    Rocket nozzle thermal shock tests in an arc heater facility

    A rocket motor nozzle thermal structural test technique that utilizes arc heated nitrogen to simulate a motor burn was developed. The technique was used to test four heavily instrumented full-scale Star 48 rocket motor 2D carbon/carbon segments at conditions simulating the predicted thermal-structural environment. All four nozzles survived the tests without catastrophic or other structural failures. The test technique demonstrated promise as a low cost, controllable alternative to rocket motor firing. The technique includes the capability of rapid termination in the event of failure, allowing post-test analysis

    Real-time assembly of ribonucleoprotein complexes on nascent RNA transcripts.

    Cellular protein-RNA complexes assemble on nascent transcripts, but methods to observe transcription and protein binding in real time and at physiological concentrations are not available. Here, we report a single-molecule approach based on zero-mode waveguides that simultaneously tracks transcription progress and the binding of ribosomal protein S15 to nascent RNA transcripts during early ribosome biogenesis. We observe stable binding of S15 to single RNAs immediately after transcription for the majority of the transcripts at 35 °C but for less than half at 20 °C. The remaining transcripts exhibit either rapid and transient binding or are unable to bind S15, likely due to RNA misfolding. Our work establishes the foundation for studying transcription and its coupled co-transcriptional processes, including RNA folding, ligand binding, and enzymatic activity such as in coupling of transcription to splicing, ribosome assembly or translation

    An Ultra-High Time Resolution Cosmic-Ray Detection Mode for the Murchison Widefield Array

    The radio-wavelength detection of extensive air showers (EASs) initiated by cosmic-ray interactions in the Earth's atmosphere is a promising technique for investigating the origin of these particles and the physics of their interactions. The Low-frequency Array (LOFAR) and the Owens Valley Long Wavelength Array (OVRO-LWA) have both demonstrated that the dense cores of low-frequency radio telescope arrays yield detailed information on the radiation ground pattern, which can be used to reconstruct key EAS properties and infer the primary cosmic-ray composition. Here, we demonstrate a new observation mode of the Murchison Widefield Array (MWA), tailored to the observation of the sub-microsecond coherent bursts of radiation produced by EAS. We first show how an aggregate 30.72MHz bandwidth (3072×10kHz frequency channels) recorded at 0.1ms resolution with the MWA's voltage capture system (VCS) can be synthesized back to the full bandwidth Nyquist resolution of 16.3ns. This process, which involves "inverting"two sets of polyphase filterbanks, retains 90.5% of the signal-to-noise of a cosmic-ray signal. We then demonstrate the timing and positional accuracy of this mode by resolving the location of a calibrator pulse to within 5m. Finally, preliminary observations show that the rate of nanosecond radio-frequency interference (RFI) events is 0.1Hz, much lower than that found at the sites of other radio telescopes that study cosmic rays. We conclude that the identification of cosmic rays at the MWA, and hence with the low-frequency component of the Square Kilometre Array, is feasible with minimal loss of efficiency due to RFI

    Andrew Melville, sacred chronology and world history: the Carmina Danielis 9 and the Antichristus

    The accepted view of the ecclesiastical reformer Andrew Melville (1545–1622) as the dynamic leader of the Presbyterian movement in Jacobean Scotland has been severely eroded in recent years, with particular criticism of the actual importance of his contribution to the Kirk and to Scottish higher education. While this reductionism has been necessary, it has resulted in an inversion of the overwhelmingly positive traditional image of Melville, and does not give us a rounded assessment of his life and works. This article attempts to partially redress this balance by looking at a neglected aspect of Melville's Latin writings, which showcase his talents as a humanist intellectual and biblical commentator. It focuses on two long poems that are both commentaries and paraphrases of Daniel and Revelation: the Carmina Danielis and the Antichristus. Through these poems, we see how Melville engaged with two problems exercising reformed theologians across Europe: the dating of key biblical events and the historicised meaning of prophecies within these texts. We also find evidence that Melville read widely among both contemporary and ancient commentators on both these issues

    High-affinity consensus binding of target RNAs by the STAR/GSG proteins GLD-1, STAR-2 and Quaking

    <p>Abstract</p> <p>Background</p> <p>STAR/GSG proteins regulate gene expression in metazoans by binding consensus sites in the 5' or 3' UTRs of target mRNA transcripts. Owing to the high degree of homology across the STAR domain, most STAR proteins recognize similar RNA consensus sequences. Previously, the consensus for a number of well-characterized STAR proteins was defined as a hexameric sequence, referred to as the SBE, for <b>S</b>TAR protein <b>b</b>inding <b>e</b>lement. <it>C. elegans </it>GLD-1 and mouse Quaking (Qk-1) are two representative STAR proteins that bind similar consensus hexamers, which differ only in the preferred nucleotide identities at certain positions. Earlier reports also identified partial consensus elements located upstream or downstream of a canonical consensus hexamer in target RNAs, although the relative contribution of these sequences to the overall binding energy remains less well understood. Additionally, a recently identified STAR protein called STAR-2 from <it>C. elegans </it>is thought to bind target RNA consensus sites similar to that of GLD-1 and Qk-1.</p> <p>Results</p> <p>Here, a combination of fluorescence-polarization and gel mobility shift assays was used to demonstrate that STAR-2 binds to a similar RNA consensus as GLD-1 and Qk-1. These assays were also used to further delineate the contributions of each hexamer consensus nucleotide to high-affinity binding by GLD-1, Qk-1 and STAR-2 in a variety of RNA contexts. In addition, the effects of inserting additional full or partial consensus elements upstream or downstream of a canonical hexamer in target RNAs were also measured to better define the sequence elements and RNA architecture recognized by different STAR proteins.</p> <p>Conclusions</p> <p>The results presented here indicate that a single hexameric consensus is sufficient for high-affinity RNA binding by STAR proteins, and that upstream or downstream partial consensus elements may alter binding affinities depending on the sequence and spacing. The general requirements determined for high-affinity RNA binding by STAR proteins will help facilitate the identification of novel regulatory targets <it>in vivo</it>.</p

    Appropriate inclusion of interactions was needed to avoid bias in multiple imputation

    OBJECTIVE: Missing data are a pervasive problem, often leading to bias in complete records analysis (CRA). Multiple imputation (MI) via chained equations is one solution, but its use in the presence of interactions is not straightforward. STUDY DESIGN AND SETTING: We simulated data with outcome Y dependent on binary explanatory variables X and Z and their interaction XZ. Six scenarios were simulated (Y continuous and binary, each with no interaction, a weak and a strong interaction), under five missing data mechanisms. We use directed acyclic graphs to identify when CRA and MI would each be unbiased. We evaluate the performance of CRA, MI without interactions, MI including all interactions, and stratified imputation. We also illustrated these methods using a simple example from the National Child Development Study (NCDS). RESULTS: MI excluding interactions is invalid and resulted in biased estimates and low coverage. When XZ was zero, MI excluding interactions gave unbiased estimates but overcoverage. MI including interactions and stratified MI gave equivalent, valid inference in all cases. In the NCDS example, MI excluding interactions incorrectly concluded there was no evidence for an important interaction. CONCLUSIONS: Epidemiologists carrying out MI should ensure that their imputation model(s) are compatible with their analysis model

    <em>SurfaceSlide</em>: A Multitouch Digital Pathology Platform

    Background: Digital pathology provides a digital environment for the management and interpretation of pathological images and associated data. It is becoming increasing popular to use modern computer based tools and applications in pathological education, tissue based research and clinical diagnosis. Uptake of this new technology is stymied by its single user orientation and its prerequisite and cumbersome combination of mouse and keyboard for navigation and annotation.Methodology: In this study we developed SurfaceSlide, a dedicated viewing platform which enables the navigation and annotation of gigapixel digitised pathological images using fingertip touch. SurfaceSlide was developed using the Microsoft Surface, a 30 inch multitouch tabletop computing platform. SurfaceSlide users can perform direct panning and zooming operations on digitised slide images. These images are downloaded onto the Microsoft Surface platform from a remote server on-demand. Users can also draw annotations and key in texts using an on-screen virtual keyboard. We also developed a smart caching protocol which caches the surrounding regions of a field of view in multi-resolutions thus providing a smooth and vivid user experience and reducing the delay for image downloading from the internet. We compared the usability of SurfaceSlide against Aperio ImageScope and PathXL online viewer.Conclusion: SurfaceSlide is intuitive, fast and easy to use. SurfaceSlide represents the most direct, effective and intimate human–digital slide interaction experience. It is expected that SurfaceSlide will significantly enhance digital pathology tools and applications in education and clinical practice
