    Metasomatic Reactions between Archean Dunite and Trondhjemite at the Seqi Olivine Mine in Greenland

    A metasomatic zone formed between the contact of a 2940 ± 5 Ma intrusive trondhjemite sheet in the Archean dunite of the Seqi Ultramafic Complex, SW Greenland, consists of three distinct mineral zones dominated by (1) talc, (2) anthophyllite, and (3) phlogopite. These zones supposedly resulted from a process of dissolution of olivine by silica rich fluid residual from the trondhjemite magma, with crystallization of secondary minerals along a compositional gradient in the fluid phase. A zircon crystal inclusion in a large (4 cm) olivine porphyroblast was dated in situ via LA-ICP-MS U–Pb isotope analysis, yielding a weighted mean 207Pb/206Pb age of 2963 ± 1 Ma, which coincides with granulite facies metamorphism and potential dehydration. Considering phase relations appropriate for the dunite composition, we deduced the talc forming conditions to be at temperatures of 600–650 °C and at a pressure below 1 GPa. This is supported by oxygen isotope data for talc, anthophyllite and phlogopite in the metasomatic zone, which suggests formation in the temperature range of 600–700 °C from fluids that had a δ18O of ~8‰ and a Δ’17O0.528 of about −40 ppm, i.e., from fluids that could have been derived from the late stage trondhjemite sheet

    The ecological fallacy of the role of age in chronic disease and hospital demand

    Objective: To examine the relationship between age and all-cause hospital utilization in the years preceding and following a diagnosis in hospital of heart failure, type 2 diabetes, or chronic obstructive pulmonary disease (COPD). Research Design: A cohort study of all patients in Western Australia who have had a principal diagnosis of heart failure, type 2 diabetes, or COPD, upon admission to hospital. All-cause hospital utilization 6 years preceding and 4 years following cardinal events, that is, a disease-specific diagnosis upon hospital admission, where such an event has not occurred in the previous 2 years, are examined in specific age groups. Results: Six years preceding a cardinal event, all-cause emergency department (ED) presentations are similar in all age groups, from under 55 to over 85 years of age, except in COPD where ED presentation rates are higher in younger groups. All-cause hospital inpatient days are transiently higher in the years preceding and following a cardinal event in older age groups, yet return to similar levels across all age cohorts after 4 years. ED presentations are significantly higher in the 4 years following cardinal events in younger compared with older groups. Conclusions: Longitudinal analysis of utilization around cardinal events overcomes the confounding effect of differences in chronic disease rates between age groups, avoiding a source of ecologic bias that erroneously attributes increasing utilization in individuals with chronic disease to age. Programs designed to reduce hospital demand in patients with chronic disease should possibly focus on younger, rather than older, individuals

    Repercussion of megakaryocyte-specific Gata1 Loss on megakaryopoiesis and the hematopoietic precursor compartment

    During hematopoiesis, transcriptional programs are essential for the commitment and differentiation of progenitors into the different blood lineages. GATA1 is a transcription factor expressed in several hematopoietic lineages and essential for proper erythropoiesis and megakaryopoiesis. Megakaryocyte-specific genes, such as GP1BA, are known to be directly regulated by GATA1. Mutations in GATA1 can lead to dyserythropoietic anemia and pseudo gray-platelet syndrome. Selective loss of Gata1 expression in adult mice results in macrothrombocytopenia with platelet dysfunction, characterized by an excess of immature megakaryocytes. To specifically analyze the impact of Gata1 loss in mature committed megakaryocytes, we generated Gata1-Lox|Pf4-Cre mice (Gata1cKOMK). Consistent with previous findings, Gata1cKOMK mice are macrothrombocytopenic with platelet dysfunction. Supporting this notion we demonstrate that Gata1 regulates directly the transcription of Syk, a tyrosine kinase that functions downstream of Clec2 and GPVI receptors in megakaryocytes and platelets. Furthermore, we show that Gata1cKOMK mice display an additional aberrant megakaryocyte differentiation stage. Interestingly, these mice present a misbalance of the multipotent progenitor compartment and the erythroid lineage, which translates into compensatory stress erythropoiesis and splenomegaly. Despite the severe thrombocytopenia, Gata1cKOMK mice display a mild reduction of TPO plasma levels, and Gata1cK-OMK megakaryocytes show a mild increase in Pf4 mRNA levels; such a misbalance might be behind the general hematopoietic defects observed, affecting locally normal TPO and Pf4 levels at hematopoietic stem cell niches. © 2016 Meinders et al.     Whole rock and olivine geochemistry, together with high 3He/4He (>MORB=8 R/RA, where RA denotes 3He/4He of air) found in the East African Rift System (EARS) suggests the presence of an underlying deep mantle plume contributing to the magmatism throughout the rift. This thesis contains new major, minor and trace elements of olivine crystals from a suite of 17 mafic lava and tephra samples from six volcanic series within the Rungwe Volcanic Province (RVP), Tanzania. The olivine chemistry shows considerable variations in their elemental concentrations and elemental ratios. Olivine chemistry proxies were used to identify the lithology of the source supplying magmas to the RVP and estimating the degrees of crustal contamination, partial melting and crystallization temperatures affecting the source. Variations in Ni content in the olivines suggest that the source has likely undergone a series of mafic recharge events and subsequent mixing. High Ni and low Mn found in the Ngozi -younger extrusives series, however, suggests the presence of pyroxenite in the source, which is attributed to a lithologically heterogenous (hybrid) source. Furthermore, our findings were compared to the whole rock chemistry, isotopic ratios and helium isotope systematics of the samples of Castillo et al (2014) and Hilton et al (2011). These findings concur with recent hypotheses where a primordial deeply sourced plume, characterized by high 3He/4He, is present underneath the RVP. Additionally, this hybrid plume component mixed with an old lithospheric component associated with the Tanzania Craton could result in the wide variations observed in the RVP lava chemistry

    The ratchet effect: dramatic and sustained changes in health care utilization following admission to hospital with chronic disease

    Objective: To describe the previously unexamined association between admissions to hospital with chronic disease and changes in all-cause health service utilization over time. Research Design: A cohort study examining the population of Western Australia with hospitalizations for chronic disease from 2002 to 2010. A rolling clearance period is used to define cardinal events, that is, a disease-specific diagnosis upon hospital admission, where such an event has not occurred in the previous 2 years. Changes in the rate of cardinal events associated with diagnoses of heart failure, type 2 diabetes, chronic obstructive pulmonary disease, cataract with diabetes, asthma, and dialysis are examined. Health service utilization (defined as inpatient days or emergency department presentations) 6 years preceding and 4 years following such events is presented. Results: Cardinal events make up 40%-60% of all chronic disease admissions. A previously undescribed ratchet effect following cardinal events specifically associated with type 2 diabetes, heart failure, and chronic obstructive pulmonary disease is observed. This involves a 2- to 3-fold increase in inpatient days and emergency department presentations that are sustained for at least 4 years. Conclusions: Cardinal events represent an important reference point to understand the impact of chronic disease on health service utilization. Events that herald such a marked transition in health service demand have not been previously described

    Predictors of Urinary Pyrethroid and Organophosphate Compound Concentrations among Healthy Pregnant Women in New York

    Our study aimed to investigate dietary and non-dietary predictors of exposure to pyrethroids, organophosphates pesticides and 2,4-D herbicide in two cohorts of pregnant women in New York City: 153 women from the Thyroid Disruption and Infant Development (TDID) cohort and 121 from the Sibling/Hermanos Cohort(S/H). Baseline data on predictors were collected from the women at time of recruitment. We used three different modeling strategies to address missing data due to biomarker values below the limit of detection (<LOD): (1) logistic regression models with biomarkers categorized as (<median, ≥median); (2) linear regression models, imputing the <LOD values with (LOD/√2); (3) regression models, considering <LOD values as left-censored. Generally, all three models identified similar predictors of exposure. We found that ethnicity, higher income and education predicted higher concentrations of most of the biomarkers in both cohorts. Mothers who consumed processed meat in the TDID cohort, and broiled, barbequed food or burgers in the S/H cohort, tended to have lower concentrations of organophosphates and 2,4-D. The choice of modeling led to a few different predictors identified, and the selection of modeling strategy should be based on the study question
