    Minimisation of risk exposure at the pre-production stage through the use of contractor-led design management

    Many large-scale construction projects (LSPs) are designed in a collaboration between famous international architects and local design firms. Design management usually evolves as a tool at the design stage for designers and design solution. One of the special characteristics of the Korean construction environment places a duty and responsibility on the contractor to coordinate and check design information. Hence the contractor must manage and integrate diverse design information into the production process. This research considers how the design management diagram (DMD) can help as a part of the system at the pre-production stage of LSPs in Korea. The pre-production stage receives insufficient attention from the research community from the perspective of design management; it is a complex process involving interdependence, risk, and uncertainty. Through the application of a DMD from the pre-production stage, the contractor can predict and manage the design-related uncertainty during production stages. The design management factors (DMFs) were analysed by the analytic hierarchy process (AHP), and then used complexity system theory to understand the interrelationship between the causal factors. DMFs are presented as a causal loop diagram which can help the contractor to cope with design related uncertainties at the early production phase

    Artificial Intelligence (AI)-Based Technology Adoption in the Construction Industry: A Cross National Perspective Using the Technology Acceptance Model

    The research has chosen the workers in construction-related companies in South Korea and the United Kingdom (UK) as research subjects in order to analyse factors that influence their usage intention of Artificial Intelligence (AI) based technologies. The perceived usefulness had a positive impact (+) on technological satisfaction and usage intention in terms of the commonalities shown by the construction industry workers in both countries, South Korea and the UK, in adopting AI-based technologies. Moreover, the most remarkable differences were personal competence and social influence when choosing AI-based technologies. It was analysed that in the case of South Korea, personal competence had a positive impact (+) on perceived ease of use, whereas the UK had a positive impact (+) on perceived usefulness and perceived ease of use. This study holds particular significance in the domain of cross-cultural research within the construction industry. It conducts an analysis of the factors influencing the adoption of AI-driven technologies or products, with a specific focus on the cultural differences between two nations: South Korea and the UK, which represent Eastern and Western cultural paradigms, respectively

    Synthetic DNA immunotherapy in biochemically relapsed prostate cancer

    Background: INO-5150 (PSA and PSMA) +/- INO-9012 (IL-12), a synthetic DNA immunotherapy, was assessed for safety, immunogenicity and efficacy in biochemically recurrent prostate cancer patients (pts). Methods: Phase I, open-label, multi-center study in the US included pts with rising PSA after surgery and/or RT, PSA doubling time (PSADT) \u3e3 months (mos), testosterone \u3e150 ng/dL and no concurrent ADT. Safety, immunogenicity and efficacy (PSA kinetics, PFS) were evaluated in 4 treatment arms of 15 pts each. Arms A: 2mg INO-5150, B: 8.5 mg INO-5150, C: 2mg INO-5150 + 1mg INO-9012 and D: 8.5mg INO-5150 + 1mg INO-9012. Pts received 4 IM doses of vaccine followed by electroporation on day 0, wks 3, 12 and 24 and were followed for 72 wks. Results: 50/61 (82%) pts completed all visits and treatments were well tolerated with no safety concerns. Median PFS for overall population [N = 61, baseline (D0) PSADT range (mos) 1.5-217.1, median 9.8] and for a subset of pts with D0 PSADT ≤12mos (N = 36) has not yet been reached (FU 3-19 mos). 86% of pts with D0 PSADT ≤12 mos were progression free through 19mos FU. 27 out of 36 (75%) pts with D0 PSADT≤ 12 mos had disease stabilization at wks 27 evidenced by significant improvement in log2PSA change over time (slope) and PSADT from D0 (Slope=0.19 declined to 0.1, PSADT=5.3 improved to 10.1 mos, p = \u3c0.0001). This effect was maintained at wk 72 (Slope=0.09, PSADT=10.6, p = \u3c0.0001). Immunogenicity was observed in 77% (47/61) of pts by multiple immunologic assessments. Patient immunogenicity to INO-5150 as determined by CD38 and Perforin + CD8 T cell immune reactivity correlated with attenuated % PSA rise compared to pts without reactivity (p = 0.05, n = 50). Conclusions: INO-5150 +/- INO-9012 was safe, well tolerated and immunogenic. Clinical efficacy was observed in the patients with D0 PSADT≤ 12 mos as evidenced by a significant dampening of log2PSA change over time and increased PSADT up to 72 weeks FU. Additional genomic analyses are ongoing to further elucidate the correlation of immunologic efficacy and clinical benefit. (NCT02514213)

    Regulation of the hTERT telomerase catalytic subunit by the c-Abl tyrosine kinase

    BACKGROUND: Telomeres consist of repetitive (TTAGGG) DNA sequences that are maintained by the multisubunit telomerase ribonucleoprotein. Telomerase consists of an RNA, which serves as template for the sequence tracts, and a catalytic subunit that functions in reverse transcription of the RNA template. Cloning and characterization of the human catalytic subunit of telomerase (hTERT) has supported a role in cell transformation. How telomerase activity is regulated, however, is largely unknown. RESULTS: We show here that hTERT associates directly with the c-Abl protein tyrosine kinase. We also found that c-Abl phosphorylates hTERT and inhibits hTERT activity. Moreover, our findings demonstrate that exposure of cells to ionizing radiation induces tyrosine phosphorylation of hTERT by a c-Abl-dependent mechanism. The functional significance of the c-Abl-hTERT interaction is supported by the demonstration that cells deficient in c-Abl show telomere lengthening. CONCLUSIONS: The ubiquitously expressed c-Abl tyrosine kinase is activated by DNA double-strand breaks. Our finding of telomere lengthening in c-Abl-deficient cells and the functional interactions between c-Abl and hTERT support a role for c-Abl in the regulation of telomerase function

    The comparison between the NCTU and Caltech PQ-PMMA material shows that the difference in their behavior lies in the different concentration of residual MMA in the samples. Experimental evidence shows that during recording, PQ molecules attach to MMA but no diffusion takes place at room temperature. However, the excess of monomer during recording enables photoinduced polymerization as a mechanism for hologram formation leading to high diffraction efficiencies without the need of baking. The grating formed by the PQ-MMA groups is unstable and it can be erased within a few hours of baking

    RNA interference demonstrates a novel role for integrin-linked kinase as a determinant of pancreatic adenocarcinoma cell gemcitabine chemoresistance

    Integrin-linked kinase (ILK) facilitates signal transduction between extracellular events and important intracellular survival pathways involving protein kinase B/Akt. We examined the role of ILK in determining pancreatic adenocarcinoma cellular chemoresistance to the nucleoside analogue gemcitabine. Cellular ILK expression was quantified by Western blot analysis. We examined the effects of overexpression of active ILK and of ILK knockdown induced by RNA interference on gemcitabine chemoresistance. We also examined the effects of modulating ILK expression on gemcitabine-induced caspase 3– mediated apoptosis, phosphorylation status of Akt (Ser(473)) and glycogen synthase kinase. Overexpression of ILK increased cellular gemcitabine chemoresistance, whereas ILK knockdown induced chemosensitization via increased caspase 3– mediated apoptosis. ILK knockdown attenuated Akt Ser(473) and glycogen synthase kinase phosphorylation, whereas overexpression of constitutively active myristoylated Akt was sufficient to induce significant recovery in gemcitabine chemoresistance in the presence of ILK knockdown. Levels of ILK expression affect gemcitabine chemoresistance in pancreatic adenocarcinoma cells. This novel finding suggests that therapies directed against ILK and its downstream signaling targets may have the potential to enhance the efficacy of gemcitabine-based chemotherapy
