    A Biodegradable Polyethylenimine-Based Vector Modified by Trifunctional Peptide R18 for Enhancing Gene Transfection Efficiency In Vivo

    Lack of capacity to cross the nucleus membrane seems to be one of the main reasons for the lower transfection efficiency of gene vectors observed in vivo study than in vitro. To solve this problem, a new non-viral gene vector was designed. First, a degradable polyethylenimine (PEI) derivate was synthesized by crosslinking low-molecular-weight (LMW) PEI with N-octyl-N-quaternary chitosan (OTMCS), and then adopting a designed trifunctional peptide (RGDC- TAT-NLS) with good tumor targeting, cell uptake and nucleus transport capabilities to modify OTMCS-PEI. The new gene vector was termed as OTMCS- PEI-R18 and characterized in terms of its chemical structure and biophysical parameters. Gene transfection efficiency and nucleus transport mechanism of this vector were also evaluated. The polymer showed controlled degradation and remarkable buffer capabilities with the particle size around 100–300 nm and the zeta potential ranged from 5 mV to 40 mV. Agraose gel electrophoresis showed that OTMCS-PEI-R18 could effectively condensed plasmid DNA at a ratio of 1.0. Besides, the polymer was stable in the presence of sodium heparin and could resist digestion by DNase I at a concentration of 63U DNase I/DNA. OTMCS-PEI-R18 also showed much lower cytotoxicity and better transfection rates compared to polymers OTMCS-PEI-R13, OTMCS-PEI and PEI 25 KDa in vitro and in vivo. Furthermore, OTMCS-PEI-R18/DNA complexes could accumulate in the nucleus well soon and not rely on mitosis absolutely due to the newly incorporated ligand peptide NLS with the specific nuclear delivery pathway indicating that the gene delivery system OTMCS-PEI-R18 could reinforce gene transfection efficiency in vivo

    Manometric Measurement of the Sphincter of Oddi in Patients with Common Bile Duct Stones: A Consecutive Study of the Han Population of China

    Objective. Role of dysfunction of the sphincter of Oddi (SO) in choledocholithiasis is controversial. This study was to evaluate SO motor activity in patients with common bile duct (CBD) stones in the Han population of China. Patients and Methods. In this study, 76 patients with CBD stones were enrolled in a single tertiary endoscopy center. Data of SO motor activities was prospectively evaluated by endoscopic manometry. Mean basal SO pressure, amplitude, and frequency were collected and analyzed. Results. The mean basal SO pressure, amplitude, and frequency were 52.7±40.0 (1.60–171.1) mmHg, 39.9±19.7 (14.9–115.5) mmHg, and 5.7±3.2 (1.3–13.8)/min, respectively. The basal SO pressure was higher in patients with CBD stones < 10 mm in diameter than that in those with CBD stones larger than 10 mm in diameter (60.7±41.0 mmHg versus 36.8±29.4 mmHg, P=0.043). There was no significant difference in the basal SO pressure, amplitude, and frequency when compared with the CBD diameter, CBD stone number, prior cholecystectomy, periampullary diverticula, and symptoms. Levels of alanine aminotransferase, aspartate transaminase, Îł-glutamyl transpeptidase, and alkaline phosphatase showed no significant difference in patients with normal or elevated basal SO pressure. Conclusion. These results identify that, in Chinese Han population, abnormalities of SO motor activity are associated with CBD stones

    Segmental Membranous Glomerulopathy in Adults

    Introduction: The clinicopathological features of segmental membranous glomerulopathy (SMGN) have not been well characterized. The aim of this study was to investigate the prevalence and clinicopathological features of SMGN in adults. Methods: Adult patients with biopsy-confirmed SMGN in the native kidney at our center between January 2017 to September 2020 were identified. The clinicopathological features of SMGN were collected. The glomerular deposition of IgG subclasses, M-type phospholipase A2 receptor 1 (PLA2R), thrombospondin type 1 domain-containing 7A (THSD7A) and neural epidermal growth factor-like 1 protein (NELL1) were tested. Clinical and pathologic features were comparable between NELL1-positive and NELL1-negative SMGN. Results: A total of 167 patients with biopsy-proven SMGN were enrolled. During the same period, 32,640 (33.0%) out of 98,939 renal biopsies were diagnosed with membranous nephropathy (MN) in adults. SMGN accounted for 0.17% of total kidney biopsies and 0.51% of MN in adults. One hundred and fifty (89.8%) cases were isolated SMGN and 17 (10.2%) cases were complicated with other kidney disease. Clinically, the median age of isolated SMGN patients was 41.5 years, with female (74%) predominance, and 33.1% had full nephrotic syndrome. Pathologically, IgG1 was the dominant subclass (92.5%), followed by IgG4 (45.0%). PLA2R and THSD7A staining were done in 142 and 136 isolated SMGN cases, respectively. In which, all the cases showed negative. NELL1 staining was done in 135 isolated SMGN cases, 58 cases (43.0%) showed positive. Fifty-eight patients (41.1%) had diffuse (≄90%) foot process effacement, 119 patients (83.8%) had either stage I (38.0%) or stage II (45.8%) membranous alterations in patients with SMGN. Most patients with NELL1-positive SMGN were female. Patients with NELL1-positive SMGN were more likely with lower prevalence of full nephrotic syndrome than NELL1-negative SMGN. Conclusions: SMGN is a relatively rare pathological type. Majority of patients with isolated SMGN were female, with a median age of 41.5 years, 33.1% had full nephrotic syndrome, absence of PLA2R and THSD7A, 43.0% with NELL1-positive, and mainly stage I or II MN (83.8%).     A longitudinal resource for population neuroscience of school-age children and adolescents in China

    During the past decade, cognitive neuroscience has been calling for population diversity to address the challenge of validity and generalizability, ushering in a new era of population neuroscience. The developing Chinese Color Nest Project (devCCNP, 2013–2022), the first ten-year stage of the lifespan CCNP (2013–2032), is a two-stages project focusing on brain-mind development. The project aims to create and share a large-scale, longitudinal and multimodal dataset of typically developing children and adolescents (ages 6.0–17.9 at enrolment) in the Chinese population. The devCCNP houses not only phenotypes measured by demographic, biophysical, psychological and behavioural, cognitive, affective, and ocular-tracking assessments but also neurotypes measured with magnetic resonance imaging (MRI) of brain morphometry, resting-state function, naturalistic viewing function and diffusion structure. This Data Descriptor introduces the first data release of devCCNP including a total of 864 visits from 479 participants. Herein, we provided details of the experimental design, sampling strategies, and technical validation of the devCCNP resource. We demonstrate and discuss the potential of a multicohort longitudinal design to depict normative brain growth curves from the perspective of developmental population neuroscience.     Identification of Pink-Coloured CVD Synthetic Diamonds from Huzhou Sino-C Semiconductor Co. in China

    In recent years, increasing numbers of pink-coloured CVD synthetic diamonds have appeared on the market. One of the major sources is Huzhou SinoC Semiconductor Science and Technology Co., Ltd., Zhejiang province of China. In this article, seven pink-coloured CVD-grown diamonds produced in the last two years by Huzhou have been investigated and identified, including their gemological and spectroscopic characteristics. In DiamondView, they fluoresced orange–red, with an obscure striated growth structure, which is common for CVD synthetics. The mid-IR absorption spectra of these samples showed some single nitrogen and hydrogen-related features (1130, 1344, 3123, 3323 cm−1), which indicated that the diamonds were type Ib and were CVD-grown diamonds. The H1a defect annealed out at approximately 1400 °C, whereas the 3107 cm−1 defect was produced by annealing above 1700 or 1800 °C. This implied that the samples had undergone two separate heat treatments: first, a high-temperature anneal (possibly an HPHT treatment to reduce any brown colour), which would have produced the 3107 cm−1 defects and a small number of A centres, followed by irradiation, followed by annealing above 800 °C to make the vacancies mobile. The UV–Vis–NIR absorption spectra showed distinct NV-related features (575 and 637 nm), the main reason for the pink colour. Photoluminescence spectra obtained at liquid nitrogen temperature recorded radiation-related emissions (388.9, 503.5 nm), a strong N-V centre, H3 and H2 defects, and many unassigned emissions. These pink CVD products can be separated from natural and treated pink-coloured diamonds by a combination of optical spectroscopic properties, such as fluorescence colour, and absorption features in the infrared and UV–Vis regions

    The projectile extrusion process is an extremely important part of the initial trajectory of the artillery launch. The differentiation of the extrusion process brings about the stability problem of the extrusion process, which directly affects the stability and safety of the artillery, ballistic and projectile launching process. sex. Since the simplified chip intrusion resistance model does not pay attention to the projectile movement process and the formation process of the elastic belt groove, and only studies the change law of the intrusion resistance, the model is relatively simple and the parameters are easy to determine. In this paper, a quasi-static test device for the extrusion process of the elastic belt is established. The model is calibrated with the simplified chip resistance model with the intrusion process. Finally, the model is used to calculate the resistance along the process of a large-caliber howitzer projectile extrusion process, and it is compared with the embedded resistance test results of a large-caliber howitzer., the results show that in the whole process of extrusion, the relative error between the calculated value of the total extrusion resistance and the experimental value is basically below 10%, and the maximum relative error is 10.98%, which verifies the accuracy of the model
