    Behavior-oriented numerical modeling of nearshore oceanic current and application on sea harbor

    590-602The West Guangdong longshore current (WG current) is a unique oceanic current system. Plenty of field survey datasets indicated that it flows uni-directionally from north-east to south-west in the entire year even during the south-west monsoon season. At present, the natural formation mechanism of the WG current remains controversial, and the traditional process-oriented modeling method could not deal with the dilemma of the scaling mismatch between the regional ocean circulation (several thousand kilometers) and harbor structure (several hundred meters). To solve this problem, in this paper, a behavior-oriented modeling concept was developed, wherein the contribution of the WG current was considered by incorporating additional net flow flux in the hydrodynamic model to separate it from the tidal currents. Through rigorous validations according to the site observed datasets, the proposed modeling concept was found to have good precision. Using the Jida Harbor as a real-life case, the modeling results showed that after the combination of the tidal current and WG current, the westward cross-flow speed in the approach channel could exceed 0.5 m/s, and at the harbor entrance the WG current induces an intense local circulation cell while ebbing, which may bring in additional maneuver risk to the ships

    Ku80 cooperates with CBP to promote COX-2 expression and tumor growth.

    Cyclooxygenase-2 (COX-2) plays an important role in lung cancer development and progression. Using streptavidin-agarose pulldown and proteomics assay, we identified and validated Ku80, a dimer of Ku participating in the repair of broken DNA double strands, as a new binding protein of the COX-2 gene promoter. Overexpression of Ku80 up-regulated COX-2 promoter activation and COX-2 expression in lung cancer cells. Silencing of Ku80 by siRNA down-regulated COX-2 expression and inhibited tumor cell growth in vitro and in a xenograft mouse model. Ku80 knockdown suppressed phosphorylation of ERK, resulting in an inactivation of the MAPK pathway. Moreover, CBP, a transcription co-activator, interacted with and acetylated Ku80 to co-regulate the activation of COX-2 promoter. Overexpression of CBP increased Ku80 acetylation, thereby promoting COX-2 expression and cell growth. Suppression of CBP by a CBP-specific inhibitor or siRNA inhibited COX-2 expression as well as tumor cell growth. Tissue microarray immunohistochemical analysis of lung adenocarcinomas revealed a strong positive correlation between levels of Ku80 and COX-2 and clinicopathologic variables. Overexpression of Ku80 was associated with poor prognosis in patients with lung cancers. We conclude that Ku80 promotes COX-2 expression and tumor growth and is a potential therapeutic target in lung cancer

    An Integrated Method to Acquire Technological Evolution Potential to Stimulate Innovative Product Design

    Fast and effective forecasting of the new generation of products is key to enhancing the competitiveness of a company in the market. Although the technological evolution laws in the theory of the solution of inventive problems (TRIZ) have been used to predict the potential states of products for innovation, there is a lack of effective methods to select the best technological evolution law consistently with product replacement and update, and acquiring potentially new technologies and solutions, which relies heavily on designers’ experience and makes it impossible for designers to efficiently use the technological evolution laws to stimulate product innovation. Aimed to bridge this gap, this paper proposes an integrated method consisting of three main steps, combining the technological evolution laws with back propagation neural network (BPNN), international patent classification (IPC) knowledge and company’s technological distance. The best technical evolution law is first searched by a BPNN. The functional verbs and effects in the IPC are then extracted and searched for potential technologies in the Spyder-integrated development environment. Finally, the company’s technological distance is used to select analogous sources of potential solutions in the patent database. The final innovative design is determined based on the ideality. The proposed method is applied in the development of a steel pipe-cutting machine to verify its feasibility. The proposed method reduces the dependence on designers’ experience and provides a way to access cross-domain technologies, providing a systematic approach for the technological evolution laws to motivate innovative product design

    Effect of phosphoglucosamine mutase on biofilm formation and antimicrobial susceptibilities in M. smegmatis glmM gene knockdown strain.

    UDP-N-acetylglucosamine (UDP-GlcNAc) is a direct glycosyl donor of linker unit (L-Rhamnose-D-GlcNAc) and an essential precursor of peptidoglycan in mycobacteria. Phosphoglucosamine mutase (GlmM) is involved in the formation of glucosamine-1-phosphate from glucosamine-6-phosphate, the second step in UDP-GlcNAc biosynthetic pathway. We have demonstrated that GlmM protein is essential for the growth of M. smegmatis. To facilitate the analysis of the GlmM protein function in mycobacteria, a tetracycline inducible M. smegmatis glmM gene knockdown strain was constructed by using an antisense RNA technology. After induction with 20 ng/ml tetracycline, the expression of GlmM protein in glmM gene knockdown strain was significantly decreased, resulting in a decline of cell growth. The morphological changes of glmM gene knockdown strain induced with 20 ng/ml tetracycline have been observed by scanning electron microscope and transmission electron microscope. Furthermore, insufficient GlmM protein reduced the biofilm formation and increased the sensitivity to isoniazid and ethambutol in M. smegmatis, indicating that GlmM protein had effect on the biofilm formation and the senstivity to some anti-tuberculosis drugs targeting the cell wall. These results provide a new insight on GlmM functions in mycobacteria, suggesting that GlmM could be a potential target for development of new anti-tuberculosis drug
