    Experimental studies of vortex disconnection and connection at a free surface

    An experimental study is presented that examines the interaction of a vortex ring with a free surface. The main objective of this study is to identify the physical mechanisms that are responsible for the self-disconnection of vortex filaments in the near-surface region and the subsequent connection of disconnected vortex elements to the free surface. The understanding of those mechanisms is essential for the identification and estimation of the appropriate spatial and temporal scales of the disconnection and connection process. In this regard, the velocity and vorticity fields of an obliquely approaching laminar vortex ring with a Reynolds number of 1150 were mapped by using Digital Particle Image Velocimetry (DPIV). The evolution of the near-surface vorticity field indicates that the connection process starts in the side regions of the approaching vortex ring where surface-normal vorticity already exists in the bulk. A local strain rate analysis was conducted to support this conclusion. Disconnection in the near-surface tip region of the vortex ring occurs because of the removal of surfaceparallel vorticity by the viscous flux of vorticity through the surface. Temporal and spatial mapping of the vorticity field at the surface and in the perpendicular plane of symmetry shows that the viscous flux is balanced by a local deceleration of the flow at the surface. It is found that the observed timescales of the disconnection and connection process scale with the near-surface vorticity gradient rather than with the core diameter of the vortex ring

    Evolution of microgastropods (Ellobioidea, Carychiidae): integrating taxonomic, phylogenetic and evolutionary hypotheses

    BACKGROUND: Current biodiversity patterns are considered largely the result of past climatic and tectonic changes. In an integrative approach, we combine taxonomic and phylogenetic hypotheses to analyze temporal and geographic diversification of epigean (Carychium) and subterranean (Zospeum) evolutionary lineages in Carychiidae (Eupulmonata, Ellobioidea). We explicitly test three hypotheses: 1) morphospecies encompass unrecognized evolutionary lineages, 2) limited dispersal results in a close genetic relationship of geographical proximally distributed taxa and 3) major climatic and tectonic events had an impact on lineage diversification within Carychiidae. RESULTS: Initial morphospecies assignments were investigated by different molecular delimitation approaches (threshold, ABGD, GMYC and SP). Despite a conservative delimitation strategy, carychiid morphospecies comprise a great number of unrecognized evolutionary lineages. We attribute this phenomenon to historic underestimation of morphological stasis and phenotypic variability amongst lineages. The first molecular phylogenetic hypothesis for the Carychiidae (based on COI, 16S and H3) reveals Carychium and Zospeum to be reciprocally monophyletic. Geographical proximally distributed lineages are often closely related. The temporal diversification of Carychiidae is best described by a constant rate model of diversification. The evolution of Carychiidae is characterized by relatively few (long distance) colonization events. We find support for an Asian origin of Carychium. Zospeum may have arrived in Europe before extant members of Carychium. Distantly related Carychium clades inhabit a wide spectrum of the available bioclimatic niche and demonstrate considerable niche overlap. CONCLUSIONS: Carychiid taxonomy is in dire need of revision. An inferred wide distribution and variable phenotype suggest underestimated diversity in Zospeum. Several Carychium morphospecies are results of past taxonomic lumping. By collecting populations at their type locality, molecular investigations are able to link historic morphospecies assignments to their respective evolutionary lineage. We propose that rare founder populations initially colonized a continent or cave system. Subsequent passive dispersal into adjacent areas led to in situ pan-continental or mountain range diversifications. Major environmental changes did not influence carychiid diversification. However, certain molecular delimitation methods indicated a recent decrease in diversification rate. We attribute this decrease to protracted speciation

    Next Stop: Underground. Variable degrees and variety of reasons for cave penetration in terrestrial gastropods

    Cave-dwelling animals can be classified based on their occurrence in and relationship to the subterranean environment. However, subsurface distribution data and studies addressing the initial causes for animals to enter underground habitats are sparse. By retrieving occurrence data from two voluntary biospeleological land registers in Central Germany, the degree of cave penetration in terrestrial gastropods was investigated, thus to infer potential evolutionary drivers. In total, 66 identified gastropod species entered the subterranean environment with 23 of the species also recorded from the dark zone. Gastropods possessed variable degrees of cave penetration and no obligate cave forms were observed. A decline of occurrence records from the entrance to the dark zone was the most prominent pattern. Nevertheless, several species were collected from all three light zones (i.e. entrance, transition and dark). A variety of potential reasons can be inferred to explain their subsurface appearance: exploitation of alternative food sources, aphotic above-ground movement, mating sites, temporal, seasonal and long-term refugia, and chance. Moreover, the results imply a frequent migration of species between the interconnected light zones and the surface. Consequently, terrestrial gastropods should be considered when investigating the origin and importation of allochthonous resources in caves

    On the decay of a turbulent vortex ring

    The spatiotemporal evolution of a turbulent vortex ring with an initial Reynolds number of 7500 is experimentally investigated using the technique of digital particle image velocimetry. The flow is initially characterized by the laminar/turbulent transition via azimuthal bending instabilities. After transition, the shedding of vorticity from peripheral regions of the ring is found to be responsible for the formation of a wake region. This shedding process results in the staircase‐like decay of the circulation and propagation speed of the vortex ring

    Measurements of neutron-induced reactions in inverse kinematics and applications to nuclear astrophysics

    Neutron capture cross sections of unstable isotopes are important for neutron-induced nucleosynthesis as well as for technological applications. A combination of a radioactive beam facility, an ion storage ring and a high flux reactor would allow a direct measurement of neutron induced reactions over a wide energy range on isotopes with half lives down to minutes. The idea is to measure neutron-induced reactions on radioactive ions in inverse kinematics. This means, the radioactive ions will pass through a neutron target. In order to efficiently use the rare nuclides as well as to enhance the luminosity, the exotic nuclides can be stored in an ion storage ring. The neutron target can be the core of a research reactor, where one of the central fuel elements is replaced by the evacuated beam pipe of the storage ring.     Paternal sepsis induces alterations of the sperm methylome and dampens offspring immune responses—an animal study

    Background: Sepsis represents the utmost severe consequence of infection, involving a dysregulated and self-damaging immune response of the host. While different environmental exposures like chronic stress or malnutrition have been well described to reprogram the germline and subsequently offspring attributes, the intergenerational impact of sepsis as a tremendous immunological stressor has not been examined yet. Methods: Polymicrobial sepsis in 12-week-old male C57BL/6 mice was induced by cecal ligation and puncture (CLP), followed by a mating of the male survivors (or appropriate sham control animals) 6 weeks later with healthy females. Alveolar macrophages of offspring animals were isolated and stimulated with either LPS or Zymosan, and supernatant levels of TNF-α were quantified by ELISA. Furthermore, systemic cytokine response to intraperitoneally injected LPS was assessed after 24 h. Also, morphology, motility, and global DNA methylation of the sepsis survivors’ sperm was examined. Results: Comparative reduced reduction bisulfite sequencing (RRBS) of sperm revealed changes of DNA methylation (n = 381), most pronounced in the intergenic genome as well as within introns of developmentally relevant genes. Offspring of sepsis fathers exhibited a slight decrease in body weight, with a more pronounced weight difference in male animals (CLP vs. sham). Male descendants of sepsis fathers, but not female descendants, exhibited lower plasma concentrations of IL-6, TNF-alpha, and IL-10 24 h after injection of LPS. In line, only alveolar macrophages of male descendants of sepsis fathers produced less TNF-alpha upon Zymosan stimulation compared to sham descendants, while LPS responses kept unchanged. Conclusion: We can prove that male—but surprisingly not female—descendants of post-sepsis fathers show a dampened systemic as well as pulmonary immune response. Based on this observation of an immune hypo-responsivity, we propose that male descendants of sepsis fathers are at risk to develop fungal and bacterial infections and might benefit from therapeutic immune modulation

    Development and Projected Performance of the Red Kite Sounding Rocket Motor

    Averaging about ten launches per year, DLR Mobile Rocket Base (MORABA) has been supporting rocket borne research for more than five decades. Major fields of experimentation include atmospheric physics, microgravity-based research in material physics and biology as well as hypersonic flight research and technology development. Over the last decade, a sustained demand has evolved for sounding rocket vehicles with the capacity to deliver payloads in the order of 400 kg gross mass into trajectories with apogees beyond 250 km or extended dwell time in the hypersonic regime. To leverage cost efficient and reliable military surplus motors such as the Improved Malemute for use in this regime, DLR has contracted Bayern-Chemie GmbH for a joint development and delivery of a suitable solid propellant motor to be used as a first stage. Currently in project phase C, the definition of the motor performance, materials and design are completed and manufacturing of first components has begun. The paper gives an overview of the motor performance and safety characteristics, applications of the motor in vehicle combinations with their projected performances and a schedule of tasks until qualification flight

    Sepsis Induces a Long-Lasting State of Trained Immunity in Bone Marrow Monocytes

    Innate immune memory describes the functional reprogramming of innate immune cells after pathogen contact, leading to either a boosted (trained immunity) or a diminished (immune tolerance) response to a secondary stimulus. Immune tolerance or “sepsis-induced immunosuppression” is a typical hallmark of patients after sepsis survival, characterized by hypo-responsiveness of the host's immune system. This condition renders the host vulnerable for a persisting infection or the occurrence of secondary, often opportunistic infections, along with an increased mortality rate. The mechanisms involved in the maintenance of this long-lasting condition are not examined yet. Polymicrobial abdominal sepsis was induced in 12 week old male C57BL/6 mice by cecal ligation and puncture. Mice were euthanized 3 months after insult. Immune cell composition of the spleen and whole blood, as well as stem and progenitor cells of the bone marrow, were assessed by flow cytometry. Whole blood and bone marrow monocytes were stimulated with LPS and supernatant levels of TNF and IL-6 detected by ELISA. Furthermore, naïve bone marrow monocytes were analyzed for metabolic (Seahorse technology) and transcriptomic (RNA sequencing) changes. Flow cytometric analysis revealed an increase of inflammatory monocytes and regulatory T cells in the spleen, whereby immune composition of whole blood kept unchanged. Granulocyte-monocyte progenitor cells are increased in sepsis survivors. Systemic cytokine response was unchanged after LPS challenge. In contrast, cytokine response of post-septic naïve bone marrow monocytes was increased. Metabolic analysis revealed enhanced glycolytic activity, whereas mitochondrial indices were not affected. In addition, RNA sequencing analysis of global gene expression in monocytes revealed a sustained signature of 367 differentially expressed genes. We here demonstrate that sepsis via functional reprogramming of naïve bone marrow monocytes induces a cellular state of trained immunity, which might be counteracted depending on the compartmental localization of the cell. These findings shed new light on the complex aftermath of sepsis and open up a new pathophysiological framework in need for further research

    Intravenous versus epidural analgesia to reduce the incidence of gastrointestinal complications after elective pancreatoduodenectomy (the PAKMAN trial, DRKS 00007784): study protocol for a randomized controlled trial

    Background: Despite substantial improvements in surgical and anesthesiological practices leading to decreased mortality of less than 5 % at high-volume centers, pancreatic surgery is still associated with high morbidity rates of up to 50 %. Attention is increasingly directed toward the optimization of perioperative management to reduce complications and enhance postoperative recovery. Currently, two different strategies for postoperative pain management after pancreatoduodenectomy are being routinely used: patient-controlled intravenous analgesia and thoracic epidural analgesia. Evidence is lacking to assess which strategy entails fewer postoperative complications. Methods/design: The PAKMAN trial is designed as an adaptive, pragmatic, randomized, controlled, multicenter, open-label, superiority trial with two parallel study groups. A total of 370 patients scheduled for elective pancreatoduodenectomy will be randomized after giving written informed consent, and 278 patients are needed for analysis. Patients with chronic pancreatitis, severe chronic obstructive pulmonary disease (COPD), American Society of Anesthesiologists (ASA) physical status classification ≥ IV, or chronic pain syndrome will be excluded. The group A intervention includes intraoperative general anesthesia and postoperative patient-controlled intravenous analgesia; the group B intervention comprises combined intraoperative general anesthesia and epidural analgesia with postoperative epidural analgesia. The primary endpoint of this trial is a composite of the gastrointestinal complications (delayed gastric emptying, pancreatic fistula, biliary leak, gastrointestinal bleeding, and postoperative ileus) up to postoperative day 30. The aim is to investigate whether the frequency of gastrointestinal complications following pancreatoduodenectomy can be reduced by 15 % using postoperative, patient-controlled intravenous analgesia compared with epidural analgesia. Discussion: Several previous studies investigating the two different strategies for postoperative pain management have mainly focused on their effectiveness in pain control. However, the PAKMAN trial is the first to compare them with regard to their impact on the surgical endpoint “postoperative gastrointestinal complications” after pancreatoduodenectomy.
