    Fabrication of self-healing injectable hyaluronic acid hydrogel for promoting angiogenesis

    Objective·To construct a self-healing injectable hyaluronic acid (HA)-based hydrogel (HAPD-Cu) and investigate the effects of different copper ions on the properties of the hydrogel and its vasogenic efficacy to evaluate its feasibility for clinical wound healing.Methods·Bisphosphonated hyaluronic acid (HAPD) was prepared via a blue-light mediated thiol-ene click reaction between thiolated hyaluronic acid (HASH) and acrylated bisphosphonate (Ac-PD) in the presence of photoinitiator 2959. Then, HAPD was further interacted with Cu2+ through metal coordination to prepare HAPD-Cu hydrogels with different Cu2+ concentrations, i.e. HAPD-Cu1, HAPD-Cu2, HAPD-Cu3 and HAPD-Cu4. The molecular structures of HASH, Ac-PD, HAPD and HAPD-Cu were verified with 1HNMR and FTIR. Microscopic morphology of HAPD-Cu was observed under SEM. The shear-thinning and self-healing properties of HAPD-Cu were verified by rheometer. The Cu2+ release from HAPD-Cu was determined with ICP. Live-dead staining and CCK-8 assay were applied to evaluate the biocompatibility of HAPD-Cu. The in vitro vasculogenic activity of HAPD-Cu was determined by a tubule-forming assay with human umbilical vein vascular endothelial cells and the in vivo vasculogenic activity of HAPD-Cu was assessed by CD31 tissue staining. A rat wound defect model was established in vitro to evaluate its actual repair effect.Results·The preparation of the materials was demonstrated through chemical qualitative and quantitative analytical means. In vitro studies showed that all HAPD-Cu with a loose porous internal structure exhibited outstanding self-healing, injectability and degradability, with a one-week degradation cycle and abrupt release behavior, which can meet the needs of wound healing cycle. All HAPD-Cu showed good biocompatibility except HAPD-Cu4, due to its high Cu2+ concentrations. Moreover, its angiogenic effect in vitro or in vivo was enhanced with increasing Cu2+ concentrations within the permissible Cu2+ concentration range. In vitro wound model experiments also showed that the HAPD-Cu hydrogel significantly promoted wound healing compared with the control group.Conclusion·HAPD-Cu hydrogel constructed via the metal coordination shows excellent shape plasticity, allowing the filling of defective sites in a minimally invasive form, and the release of Cu2+ greatly facilitates the establishment of early vascular networks, with giant potential for use in the repair of clinically irregular wounds

    Robust information hiding in low-resolution videos with quantization index modulation in DCT-CS domain

    Video information hiding and transmission over noisy channels leads to errors on video and degradation of the visual quality notably. In this paper, a video signal fusion scheme is proposed to combine sensed host signal and the hidden signal with quantization index modulation (QIM) technology in the compressive sensing (CS) and discrete cosine transform (DCT) domain. With quantization based signal fusion, a realistic solution is provided to the receiver, which can improve the reconstruction video quality without requiring significant extra channel resource. The extensive experiments have shown that the proposed scheme can effectively achieve the better trade-off between robustness and statistical invisibility for video information hiding communication. This will be extremely important for low-resolution video analytics and protection in big data era

    Endovascular Metal Devices for the Treatment of Cerebrovascular Diseases

    Cerebrovascular disease involves various medical disorders that obstruct brain blood vessels or deteriorate cerebral circulation, resulting in ischemic or hemorrhagic stroke. Nowadays, platinum coils with or without biological modification have become routine embolization devices to reduce the risk of cerebral aneurysm bleeding. Additionally, many intracranial stents, flow diverters, and stent retrievers have been invented with uniquely designed structures. To accelerate the translation of these devices into clinical usage, an in‐depth understanding of the mechanical and material performance of these metal‐based devices is critical. However, considering the more distal location and tortuous anatomic characteristics of cerebral arteries, present devices still risk failing to arrive at target lesions. Consequently, more flexible endovascular devices and novel designs are under urgent demand to overcome the deficiencies of existing devices. Herein, the pros and cons of the current structural designs are discussed when these devices are applied to the treatment of diseases ranging broadly from hemorrhages to ischemic strokes, in order to encourage further development of such kind of devices and investigation of their use in the clinic. Moreover, novel biodegradable materials and drug elution techniques, and the design, safety, and efficacy of personalized devices for further clinical applications in cerebral vasculature are discussed.Peer reviewe

    Local bone metabolism balance regulation via double-adhesive hydrogel for fixing orthopedic implants

However, local metabolic imbalance around implants under osteoporosis condition could jeopardize the fixation effect. Inspired by the bone structure and the composition around implants under osteoporosis condition, alendronate (A) was grafted onto methacryloyl hyaluronic acid (H) by activating the carboxyl group of methacryloyl hyaluronic acid to be bonded to inorganic calcium phosphate on trabecular bone, which is then integrated with aminated bioactive glass (AB) modified by oxidized dextran (O) for further adhesion to organic collagen on the trabecular bone. The hybrid hydrogel could be solidified on cancellous bone in situ under UV irradiation and exhibits dual adhesion to organic collagen and inorganic apatite, promoting osteointegration of orthopedic implants, resulting in firm stabilization of the implants in cancellous bone areas. In vitro, the hydrogel was evidenced to promote osteogenic differentiation of embryonic mouse osteoblast precursor cells (MC3T3-E1) as well as inhibit the receptor activator of nuclear factor-κ B ligand (RANKL)-induced osteoclast differentiation of macrophages, leading to the upregulation of osteogenic-related gene and protein expression. In a rat osteoporosis model, the bone-implant contact (BIC) of the hybrid hydrogel group increased by 2.77, which is directly linked to improved mechanical stability of the orthopedic implants.     Association of FTH1-expressing circulating tumor cells with efficacy of neoadjuvant chemotherapy for patients with breast cancer: a prospective cohort study

    Background The association between different phenotypes and genotypes of circulating tumor cells (CTCs) and efficacy of neoadjuvant chemotherapy (NAC) remains uncertain. This study was conducted to evaluate the relationship of FTH1 gene-associated CTCs (F-CTC) with/without epithelial-mesenchymal transition (EMT) markers, or their dynamic changes with the efficacy of NAC in patients with non-metastatic breast cancer. Patients and Methods This study enrolled 120 patients with non-metastatic breast cancer who planned to undergo NAC. The FTH1 gene and EMT markers in CTCs were detected before NAC (T0), after 2 cycles of chemotherapy (T1), and before surgery (T2). The associations of these different types of CTCs with rates of pathological complete response (pCR) and breast-conserving surgery (BCS) were evaluated using the binary logistic regression analysis. Results F-CTC in peripheral blood ≥1 at T0 was an independent factor for pCR rate in patients with HER2-positive (odds ratio [OR]=0.08, 95% confidence interval [CI], 0.01-0.98, P = .048). The reduction in the number of F-CTC at T2 was an independent factor for BCS rate (OR = 4.54, 95% CI, 1.14-18.08, P = .03). Conclusions The number of F-CTC prior to NAC was related to poor response to NAC. Monitoring of F-CTC may help clinicians formulate personalized NAC regimens and implement BCS for patients with non-metastatic breast cancer

    Methylprednisolone as Adjunct to Endovascular Thrombectomy for Large-Vessel Occlusion Stroke

    Importance It is uncertain whether intravenous methylprednisolone improves outcomes for patients with acute ischemic stroke due to large-vessel occlusion (LVO) undergoing endovascular thrombectomy. Objective To assess the efficacy and adverse events of adjunctive intravenous low-dose methylprednisolone to endovascular thrombectomy for acute ischemic stroke secondary to LVO. Design, Setting, and Participants This investigator-initiated, randomized, double-blind, placebo-controlled trial was implemented at 82 hospitals in China, enrolling 1680 patients with stroke and proximal intracranial LVO presenting within 24 hours of time last known to be well. Recruitment took place between February 9, 2022, and June 30, 2023, with a final follow-up on September 30, 2023.InterventionsEligible patients were randomly assigned to intravenous methylprednisolone (n = 839) at 2 mg/kg/d or placebo (n = 841) for 3 days adjunctive to endovascular thrombectomy. Main Outcomes and Measures The primary efficacy outcome was disability level at 90 days as measured by the overall distribution of the modified Rankin Scale scores (range, 0 [no symptoms] to 6 [death]). The primary safety outcomes included mortality at 90 days and the incidence of symptomatic intracranial hemorrhage within 48 hours. Results Among 1680 patients randomized (median age, 69 years; 727 female [43.3%]), 1673 (99.6%) completed the trial. The median 90-day modified Rankin Scale score was 3 (IQR, 1-5) in the methylprednisolone group vs 3 (IQR, 1-6) in the placebo group (adjusted generalized odds ratio for a lower level of disability, 1.10 [95% CI, 0.96-1.25]; P = .17). In the methylprednisolone group, there was a lower mortality rate (23.2% vs 28.5%; adjusted risk ratio, 0.84 [95% CI, 0.71-0.98]; P = .03) and a lower rate of symptomatic intracranial hemorrhage (8.6% vs 11.7%; adjusted risk ratio, 0.74 [95% CI, 0.55-0.99]; P = .04) compared with placebo. Conclusions and Relevance Among patients with acute ischemic stroke due to LVO undergoing endovascular thrombectomy, adjunctive methylprednisolone added to endovascular thrombectomy did not significantly improve the degree of overall disability.Trial RegistrationChiCTR.org.cn Identifier: ChiCTR210005172

    Abstract. Ganglia [1] is a scalable distributed monitoring system for high performance computing systems such as clusters and Grids. We propose an improved Ganglia-like clusters monitoring system, which has more reliability with federation node and associated link failures; some monitoring data is accessed by permission; adding control functions such as restart or shutdown confusion processes; send email or pager to cluster administrator when important event occurs; and optionally select some data to federation node based on user policy in order to speedup the WAN access. We have implemented a prototype system.

    Cloud robotics in smart manufacturing environments : challenges and countermeasures

    In Smart Manufacturing Environments (SME), the use of cloud robotics is based on the integration of cloud computing and industrial robots, which provides a new technological approach to task execution and resource sharing compared to traditional industrial robots. However, research on cloud robotics in SME still faces some challenges. First, highly flexible load scheduling mechanisms are immature. Second, traditional optimization mechanisms for the network service quality do not meet the requirements of smart manufacturing due to time variability and service quality dynamics. And, finally, existing learning algorithms used without cloud-assisted resources cause great resource wasting. Accordingly, this paper explores main technologies related to cloud robotics in SME. The research contents include self-adaptive adjustment mechanisms for the service quality of a cloud robot network, computing load allocation mechanisms for cloud robotics, and group learning based on a cloud platform. The results presented in this paper are helpful to understand the internal mechanisms of perception and interaction, intelligent scheduling and control of cloud robot systems oriented to smart manufacturing, and the design of a cloud architecture oriented to group learning. © 201

    Spatiotemporal Characterization and Suppression Mechanism of Supersonic Inlet Buzz with Proper Orthogonal Decomposition Method

    The buzz phenomenon of a typical supersonic inlet is analyzed using the unsteady Reynolds Average Navier-Stokes (RANS) simulation and proper orthogonal decomposition (POD) method. The dominant flow patterns and characteristics of the buzzed flow are obtained by decoupling the computed pressure field into spatial and temporal sub-parts based on the POD method. The supersonic inlet buzz phenomenon could be approximated as a product of decoupled temporal and spatial terms, and the one-dimensional (1D) mathematical model is therefore proposed. The standard deviations of the unsteady pressure fields from both the numerical simulation and the model prediction are compared. The limited discrepancy can be observed, and the good agreement validates the credibility of the proposed 1D model. The numerical simulation and the 1D model prediction are presented to explore the unsteady-jet control with a small perturbation. The results of the 1D model and the numerical simulation achieve good agreements with each other in terms of the overall trend. Finally, POD modal energy is employed to analyze the buzz suppression mechanism. When the jet frequency is identical to the dominant frequency of the buzz and the jet phase is opposite to the oscillation phase of the captured mass flow, the buzz suppression could be more efficient. The buzz suppression mechanism could be explained in two aspects. For one thing, the complex flow structure is suppressed and the first average modal energy in the inlet is increased. For another, the energy redistribution among each POD mode is achieved and the flow stability is gradually enhanced
