A tumor-associated β1 integrin mutation that abrogates epithelial differentiation control
SCC4 human keratinocytes are derived from a squamous cell carcinoma of the tongue and undergo very little spontaneous differentiation. Introduction of a wild-type β1 integrin subunit into SCC4 cells stimulates differentiation, suggesting either that the cells have a defect in the integrin signaling pathways that control differentiation or that the β1 subunit itself is defective. Here we describe a heterozygous mutation in the SCC4 β1 subunit. The mutation, T188I, maps to the I-like domain. It results in constitutive activation of ligand binding, irrespective of the partner ι subunit, in solid phase assays with recombinant protein and in living cells. The mutation promotes cell spreading, but not proliferation, motility, or invasiveness. It results in sustained activation of Erk MAPK independent of cell spreading. When introduced into SCC4 keratinocytes, the wild-type β1 integrin stimulates differentiation, whereas the mutant is inactive. Activation of β1 integrins in normal keratinocytes also suppresses differentiation. These results establish, for the first time, mutation as a mechanism by which integrins can contribute to neoplasia, because the degree of differentiation in epithelial cancers is inversely correlated with prognosis. They also provide new insights into how integrins regulate keratinocyte differentiation

Variation in the SLC23A1 gene does not influence cardiometabolic outcomes to the extent expected given its association with L-ascorbic acid
Background: Observational studies showed that circulating l-ascorbic acid (vitamin C) is inversely associated with cardiometabolic traits. However, these studies were susceptible to confounding and reverse causation.
Objectives:We assessed the relation between l-ascorbic acid and 10 cardiometabolic traits by using a single nucleotide polymorphism in the solute carrier family 23 member 1 (SLC23A1) gene (rs33972313) associated with circulating l-ascorbic acid concentrations. The observed association between rs33972313 and cardiometabolic outcomes was compared with that expected given the rs33972313-l-ascorbic acid and l-ascorbic acidâoutcome associations.
Design: A meta-analysis was performed in the following 5 independent studies: the British Women's Heart and Health Study (n = 1833), the MIDSPAN study (n = 1138), the Ten Towns study (n = 1324), the British Regional Heart Study (n = 2521), and the European Prospective Investigation into Cancer (n = 3737).
Results: With the use of a meta-analysis of observational estimates, inverse associations were shown between l-ascorbic acid and systolic blood pressure, triglycerides, and the waist-hip ratio [the strongest of which was the waist-hip ratio (â0.13-SD change; 95% CI: â0.20-, â0.07-SD change; P = 0.0001) per SD increase in l-ascorbic acid], and a positive association was shown with high-density lipoprotein (HDL) cholesterol. The variation at rs33972313 was associated with a 0.18-SD (95% CI: 0.10-, 0.25-SD; P = 3.34 Ă 10â6) increase in l-ascorbic acid per effect allele. There was no evidence of a relation between the variation at rs33972313 and any cardiometabolic outcome. Although observed estimates were not statistically different from expected associations between rs33972313 and cardiometabolic outcomes, estimates for low-density lipoprotein cholesterol, HDL cholesterol, triglycerides, glucose, and body mass index were in the opposite direction to those expected.
Conclusions: The nature of the genetic association exploited in this study led to limited statistical application, but despite this, when all cardiometabolic traits were assessed, there was no evidence of any trend supporting a protective role of l-ascorbic acid. In the context of existing work, these results add to the suggestion that observational relations between l-ascorbic acid and cardiometabolic health may be attributable to confounding and reverse causation
The artemiside-artemisox-artemisone-m1 tetrad : efficacies against blood stage p. falciparum parasites, dmpk properties, and the case for artemiside
Because of the need to replace the current clinical artemisinins in artemisinin combination
therapies, we are evaluating fitness of amino-artemisinins for this purpose. These include the
thiomorpholine derivative artemiside obtained in one scalable synthetic step from dihydroartemisinin
(DHA) and the derived sulfone artemisone. We have recently shown that artemiside undergoes facile
metabolism via the sulfoxide artemisox into artemisone and thence into the unsaturated metabolite
M1; DHA is not a metabolite. Artemisox and M1 are now found to be approximately equipotent
with artemiside and artemisone in vitro against asexual P. falciparum (Pf ) blood stage parasites (IC50
1.5â2.6 nM). Against Pf NF54 blood stage gametocytes, artemisox is potently active (IC50 18.9 nM
early-stage, 2.7 nM late-stage), although against the late-stage gametocytes, activity is expressed, like
other amino-artemisinins, at a prolonged incubation time of 72 h. Comparative drug metabolism
and pharmacokinetic (DMPK) properties were assessed via po and iv administration of artemiside,
artemisox, and artemisone in a murine model. Following oral administration, the composite Cmax
value of artemiside plus its metabolites artemisox and artemisone formed in vivo is some 2.6-fold
higher than that attained following administration of artemisone alone. Given that efficacy of short
half-life rapidly-acting antimalarial drugs such as the artemisinins is associated with Cmax, it is
apparent that artemiside will be more active than artemisone in vivo, due to additive effects of
the metabolites. As is evident from earlier data, artemiside indeed possesses appreciably greater
efficacy in vivo against murine malaria. Overall, the higher exposure levels of active drug following
administration of artemiside coupled with its synthetic accessibility indicate it is much the preferred

Mammalian Y chromosomes retain widely expressed dosage-sensitive regulators
The human X and Y chromosomes evolved from an ordinary pair of autosomes, but millions of years ago genetic decay ravaged the Y chromosome, and only three per cent of its ancestral genes survived. We reconstructed the evolution of the Y chromosome across eight mammals to identify biases in gene content and the selective pressures that preserved the surviving ancestral genes. Our findings indicate that survival was nonrandom, and in two cases, convergent across placental and marsupial mammals. We conclude that the gene content of the Y chromosome became specialized through selection to maintain the ancestral dosage of homologous X-Y gene pairs that function as broadly expressed regulators of transcription, translation and protein stability. We propose that beyond its roles in testis determination and spermatogenesis, the Y chromosome is essential for male viability, and has unappreciated roles in Turner (tm) s syndrome and in phenotypic differences between the sexes in health and disease

Activities of 11âazaartemisinin and Nâsulfonyl derivatives against asexual and transmissible malaria parasites
Dihydroartemisinin (DHA), either used in its own right or as the active drug generated inâ
vivo from the other artemisinins in current clinical useâartemether and artesunateâinduces quiescence in ringâstage parasites of Plasmodium falciparum (Pf). This induction of quiescence is linked to artemisinin resistance. Thus, we have turned to structurally disparate artemisinins that are incapable of providing DHA on metabolism. Accordingly, 11âazaartemisinin 5 and selected Nâsulfonyl derivatives were screened against intraerythrocytic asexual stages of drugâsensitive Pf NF54 and drugâresistant K1 and W2 parasites. Most displayed appreciable activities against all three strains, with IC50 values 2000 toward asexual parasites.
