    South Wales

    Carboniferous rocks in this region occur in a broadly east-west trending syncline, the core of which includes the South Wales and Pembrokeshire coalfields (Fig. 5.1). Tournaisian and Visean strata (Avon and Pembroke Limestone groups) represent deposition on a southward prograding carbonate ramp evolving into a carbonate shelf (Wright 1987), in a succession which shows similarities to that of the Bristol and Mendips areas (Chapter 6). The main outcrops, in south Pembrokeshire, Gower and the Vale of Glamorgan, occur along the southern periphery of the coalfields and are commonly affected by Variscan thrusting and folding. Thinner successions occur along what is termed the East Crop and North Crop of the South Wales Coalfield, where much of the Visean succession is absent due to sub-Namurian and intra-Visean unconformities. Namurian fluvio-deltaic deposits (Marros Group) flank the South Wales and Pembrokeshire coalfields. Much of the lower and middle Namurian succession is absent across the region, except in the west of the South Wales Coalfield where only small parts are absent beneath an intra-Namurian unconformity. Westphalian fluvio-lacustrine deposits (South Wales Coal Measures Group) form the South Wales and Pembrokeshire coalfields, located to the east and west of Carmarthen Bay, respectively. Westphalian to Stephanian Pennant alluvial facies (Warwickshire Group) occur in the core of the South Wales Coalfield syncline. Deposition of the South Wales Coal Measures and Warwickshire groups was probably laterally contiguous with those in the Bristol and Somerset coalfields (Chapter 6), but the Usk-Cowbridge High controlled and restricted sedimentation for much of the Carboniferous, with pre-Namurian uplift and erosion removing the Tournaisian and Visean succession. Later uplift is also believed to have caused attenuation of the Warwickshire Group in the east of the South Wales Coalfield. The lithostratigraphical nomenclature for the region is that of Waters et al. (2007; 2009)

    Craven Basin and southern Pennines

    Carboniferous rocks within this area occupy the region contiguous with the northern Pennines to the north (Chapter 12) and the Peak District to the south (Chapter 10). All of the stages of the Carboniferous are present at outcrop, with the exception of Stephanian strata, which are absent. The oldest Tournaisian strata crop out within the Craven Basin, and are represented by ramp carbonate rocks (Bowland High Group) deposited on the Bowland High and adjacent Lancaster Fells and Bowland sub-basins. These carbonate rocks are overlain by mainly Visean hemipelagic mudstone and carbonate turbidites (lower part of Craven Group). To the south of the Pendle Fault System (Fig. 11.1), further platform carbonate rocks are proved in the subsurface above the Central Lancashire High (Trawden Limestone Group) and the Holme High and Heywood High (Holme High Limestone Group). These carbonate rocks, which developed during the Tournaisian to late Visean, are known only from well records and geophysical information and are not divided into formations. During the Visean, the platform carbonate rocks pass laterally into more basinal successions in the Harrogate, Rossendale and Huddersfield sub-basins (Craven Group). The lithostratigraphical nomenclature for the Tournaisian and Visean strata is that of Waters et al. (2009), adapted from Riley (1990)

    A conserved U-rich RNA region implicated in regulation of translation in Plasmodium female gametocytes.

    Translational repression (TR) plays an important role in post-transcriptional regulation of gene expression and embryonic development in metazoans. TR also regulates the expression of a subset of the cytoplasmic mRNA population during development of fertilized female gametes of the unicellular malaria parasite, Plasmodium spp. which results in the formation of a polar and motile form, the ookinete. We report the conserved and sex-specific regulatory role of either the 3’- or 5’-UTR of a subset of translationally repressed mRNA species as shown by almost complete inhibition of expression of a GFP reporter protein in the female gametocyte. A U-rich, TR-associated element, identified previously in the 3’-UTR of TR-associated transcripts, played an essential role in mediating TR and a similar region could be found in the 5’-UTR shown in this study to be active in TR. The silencing effect of this 5’-UTR was shown to be independent of its position relative to its ORF, as transposition to a location 3’ of the ORF did not affect TR. These results demonstrate for the first time in a unicellular organism that the 5’ or the 3’-UTR of TR-associated transcripts play an important and conserved role in mediating TR in female gametocytes

    How auditory experience differentially influences the function of left and right superior temporal cortices

    To investigate how hearing status, sign language experience and task demands influence functional responses in the human superior temporal cortices (STC) we collected fMRI data from deaf and hearing participants (male and female), who either acquired sign language early or late in life. Our stimuli in all tasks were pictures of objects. We varied the linguistic and visuospatial processing demands in three different tasks that involved decisions about (1) the sublexical (phonological) structure of the British Sign Language (BSL) signs for the objects; (2) the semantic category of the objects; and (3) the physical features of the objects. Neuroimaging data revealed that in participants who were deaf from birth, STC showed increased activation during visual processing tasks. Importantly, this differed across hemispheres. Right STC was consistently activated regardless of the task whereas left STC was sensitive to task demands. Significant activation was detected in the left STC only for the BSL phonological task. This task, we argue, placed greater demands on visuospatial processing than the other two tasks. In hearing signers, enhanced activation was absent in both left and right STC during all three tasks. Lateralisation analyses demonstrated that the effect of deafness was more task-dependent in the left than the right STC whereas it was more task-independent in the right than the left STC. These findings indicate how the absence of auditory input from birth leads to dissociable and altered functions of left and right STC in deaf participants

    Trends in Competition and Profitability in the Banking Industry: A Basic Framework

    This paper brings to the forefront the assumptions that we make when focusing on a particular type of explanation for bank profitability. We evaluate a broad field of research by introducing a general framework for a profit maximizing bank and demonstrate how different types of models can be fitted into this framework. Next, we present an overview of the current major trends in European banking and relate them to each model’s assumptions, thereby shedding light on the relevance, timeliness and shelf life of the different models. This way, we arrive at a set of recommendations for a future research agenda. We advocate a more prominent role for output prices, and suggest a modification of the intermediation approach. We also suggest ways to more clearly distinguish between market power and efficiency, and explain why we need time-dependent models. Finally, we propose the application of existing models to different size classes and sub-markets. Throughout we emphasize the benefits from applying several, complementary models to overcome the identification problems that we observe in individual models.

    Salivary gland-specific <i>P. berghei</i> reporter lines enable rapid evaluation of tissue-specific sporozoite loads in mosquitoes

    Malaria is a life-threatening human infectious disease transmitted by mosquitoes. Levels of the salivary gland sporozoites (sgs), the only mosquito stage infectious to a mammalian host, represent an important cumulative index of &lt;i&gt;Plasmodium&lt;/i&gt; development within a mosquito. However, current techniques of sgs quantification are laborious and imprecise. Here, transgenic &lt;i&gt;P. berghei&lt;/i&gt; reporter lines that produce the green fluorescent protein fused to luciferase (GFP-LUC) specifically in sgs were generated, verified and characterised. Fluorescence microscopy confirmed the sgs stage specificity of expression of the reporter gene. The luciferase activity of the reporter lines was then exploited to establish a simple and fast biochemical assay to evaluate sgs loads in whole mosquitoes. Using this assay we successfully identified differences in sgs loads in mosquitoes silenced for genes that display opposing effects on &lt;i&gt;P. berghei&lt;/i&gt; ookinete/oocyst development. It offers a new powerful tool to study infectivity of &lt;i&gt;P. berghei&lt;/i&gt; to the mosquito, including analysis of vector-parasite interactions and evaluation of transmission-blocking vaccines

    Identification of the regions involved in phonological assembly using a novel paradigm.

    Here we adopt a novel strategy to investigate phonological assembly. Participants performed a visual lexical decision task in English in which the letters in words and letterstrings were delivered either sequentially (promoting phonological assembly) or simultaneously (not promoting phonological assembly). A region of interest analysis confirmed that regions previously associated with phonological assembly, in studies contrasting different word types (e.g. words versus pseudowords), were also identified using our novel task that controls for a number of confounding variables. Specifically, the left pars opercularis, the superior part of the ventral precentral gyrus and the supramarginal gyrus were all recruited more during sequential delivery than simultaneous delivery, even when various psycholinguistic characteristics of the stimuli were controlled. This suggests that sequential delivery of orthographic stimuli is a useful tool to explore how readers, with various levels of proficiency, use sublexical phonological processing during visual word recognition

    Activation of chimeric and full-length growth hormone receptors by growth hormone receptor monoclonal antibodies: A specific conformational change may be required for full-length receptor signaling

    Signal transduction by the growth hormone receptor (GHR) occurs through growth hormone (GH)-induced dimerization of two GHRs to form a trimeric complex, It is thought that dimerization alone is sufficient for signaling, since monoclonal antibodies (mAbs) against the extracellular domain of the GHR elicit proliferation of FDC-P1 cells transfected with a chimeric receptor comprising the extracellular domain of the GHR and the fibronectin and cytoplasmic domains of the murine granulocyte colony-stimulating factor receptor, We have screened 14 GHR mAbs for proliferative activity against characterized FDC-P1 and BaF-B03 cell lines stably expressing the full-length human, rabbit, or rat GHR, or the chimeric human GHR/granulocyte colony-stimulating factor receptor, and for transactivation of the c-fos promoter and STAT activation. With the chimeric receptor, eight mAbs were able to elicit proliferation, although there was no correlation between inhibition of hormone binding and agonist activity. In contrast, no mAbs were able to act as agonists with the full-length GHR FDC-P1 cell lines, although nine competed with GH for binding, A weak proliferative response was observed in the BaF-B03 cell lines with two of the mAbs (263 and 1C9), and the addition of anti-mouse F(ab)(2) resulted in increased signaling in the hGHR BaF-B03 cell line to a plateau of 28 +/- 4% of the GH maximum for mAb 263. These data could indicate considerable stringency in the ability of mAbs to correctly dimerize the full-length GI-IR. However, the ability of mAb 263 to stimulate a mutant hGHR altered in the F'-G' loop of domain 2 was nearly abolished, concurrent with an increased affinity of this mAb for the receptor. Since the F'-G' loop undergoes a conformational change on GH binding and is necessary for full proliferative signaling, we propose that in addition to promoting receptor dimerization, mAb 263 may induce specific changes in receptor conformation similar to GH, which are required for the biological response

    Climate instability and tipping points in the Late Devonian: Detection of the Hangenberg Event in an open oceanic island arc in the Central Asian Orogenic Belt

    Sedimentary petrology and trace element geochemistry indicate that the Late Devonian to Early Carboniferous Heishantou Formation near Boulongour Reservoir (NW Xinjiang, China) was deposited on a steep slope, mid-latitude accreting island arc complex in an open oceanic system. Bulk 87Sr/86Sr ratios show excursion patterns that are consistent with excursions at the Devonian-Carboniferous (D-C) boundary in epicontinental margin sediments. Sedimentation rates for the Boulongour Reservoir sediments show highly variable rates that range from 0.5 cm/ky to 10 cm/ky, consistent with other Late Devonian sections and modern arc environments. Multiple whole rock geochemical proxies for anoxia and the size and distribution of pyrite framboids suggest the presence of the Hangenberg Event in the sediments associated with the D-C boundary, despite the lack of visible black shale. The presence of anoxia in an open ocean, island arc environment cannot be explained by upwelling of anoxic bottom waters at this paleolatitude, but can be explained by the global infliction of oceanic shallow water eutrophication on to a climate system in distress

    This article reports early case-study data gathered from 20 schools involved in the ‘Impact of School Leadership on Pupil Outcomes’ project. We present and discuss the perceptions of headteachers and other school leaders regarding leadership factors that directly and indirectly affect pupil outcomes in these improving schools. Included are issues relating to the pivotal role played by the headteacher in setting and communicating a strategic vision for the school; models of distributed leadership; and the building of leadership capacity so as to build a collective responsibility for the improvement of pupil outcomes
