Development of a Novel Antibacterial Medicine that Targets a Characteristic Lipid of the Cell Membranes of <em>Helicobacter pylori</em>
Helicobacter pylori is one of the most prevalent causes of gastritis. This pathogen colonizes for many years human stomach and asymptomatically leads the persons to chronic gastritis. The eradication of H. pylori from human stomach is, therefore, important in order to prevent the digestive diseases including peptic ulcers and gastric cancer that develop via chronic atrophic gastritis. Wide-spectrum antibiotics such as amoxicillin and metronidazole are used for the treatment for H. pylori infectious diseases. However, the H. pylori strains resistant to these antibiotics are increasing year by year around the world. On this basis, we need urgently to develop the antibacterial medicines that act on H. pylori with a novel mechanism. Recent studies by our group have demonstrated that H. pylori shows susceptibility to the bactericidal action of indene compounds derived from decomposition of vitamin D. The bactericidal action of indene compounds is selective not against commonplace bacteria but against H. pylori. The indene compounds turned out to target the H. pylori’s phosphatidylethanolamine that retains a myristic acid as the saturated fatty acid side chain. These findings will contribute to the development of new antibacterial medicines specialized to the treatment for H. pylori infectious diseases
Evaluation of the Subchronic Toxicity of Dietary Administered Equisetum arvense in F344 Rats
Equisetum arvense, commonly known as the field horsetail, has
potential as a new functional food ingredient. However, little information is
available on its side effects, and the general toxicity of Equisetum
arvense has yet to be examined in detail. In the present study, we
evaluated the influence of administration in diet at doses of 0, 0.3, 1 and 3%
for 13 weeks in male and female F344 rats. No toxicity was detected with
reference to clinical signs, body weight, urinalysis, hematology and serum
biochemistry data and organ weights. Microscopic examination revealed no
histopathological lesions associated with treatment. In conclusion, the
no-observed-adverse-effect level (NOAEL) for Equisetum arvense
was determined to be greater than 3% in both sexes of F344 rat (males and
females: >1.79 g/kg BW/day and >1.85 g/kg BW/day, respectively) under the
conditions of the present study
Targeted Proteomics of Isolated Glomeruli from the Kidneys of Diabetic Rats: Sorbin and SH3 Domain Containing 2 Is a Novel Protein Associated with Diabetic Nephropathy
To evaluate proteins associated with the development of diabetic nephropathy, a major cause of the end-stage renal disease, we analyzed protein expression in isolated glomeruli from spontaneous type 2 diabetic (OLETF) rats and their age-matched control littermates (LETO) in the early and proteinuric stages of diabetic nephropathy using QSTAR Elite LC-MS/MS. Among the 191 and 218 proteins that were altered significantly in the OLETF rats, twenty-four were actin cytoskeleton-associated proteins implicated in the formation of stress fibers, and the impairment of actin polymerization, intermediate filaments and microtubules. Importantly, sorbin and SH3 domain containing 2 (SORBS2), which is involved in the formation of stress fibers, was significantly upregulated in both stages of diabetic nephropathy (1.49- and 1.97-fold, resp.). Immunohistochemical and quantitative-PCR analyses revealed upregulation of SORBS2 in podocytes of glomeruli of OLETF rats. Our findings suggested that SORBS2 may be associated with the development of diabetic nephropathy possibility by reorganization of actin filaments
Mechanical Thrombectomy for Acute Ischemic Stroke due to Thrombus in the Pulmonary Vein Stump after Left Pulmonary Lobectomy: A Case Series
Cerebral embolic stroke caused by a thrombus in the pulmonary vein stump after left pulmonary lobectomy is a serious complication. We retrospectively analyzed four patients who underwent mechanical thrombectomy for large-vessel occlusion after left pulmonary lobectomy between January 2014 and March 2022. Two cases occurred after left upper lobectomy and the others occurred after left lower lobectomy. All patients presented with cerebral embolic stroke from the day after surgery to the 9th postoperative day, and successful reperfusion was achieved in all cases. Two patients had good outcomes at 90 days. Thrombus in the pulmonary vein stump is the probable cause of cerebral embolism, and mechanical thrombectomy is effective. Further studies are required to establish preventive measures and perioperative management strategies
Enhanced Urinary Bladder, Liver and Colon Carcinogenesis in Zucker Diabetic Fatty Rats in a Multiorgan Carcinogenesis Bioassay: Evidence for Mechanisms Involving Activation of PI3K Signaling and Impairment of p53 on Urinary Bladder Carcinogenesis
In the present study, modifying effects of diabetes on carcinogenesis induced in type 2
diabetes mellitus model Zucker diabetic fatty (ZDF) rats were investigated using a
multiorgan carcinogenesis bioassay. Our re sults demonstrated enhancement of urinary
bladder, colon and liver carcinogenesis in ZDF rats treated with five types of carcinogens
(DMBDD). Elevated insulin and leptin and decreased adiponectin levels in the serum may be
responsible for the high susceptibility of type 2 diabetes mellitus model rats to
carcinogenesis in these organs. Possible mechanisms of increased susceptibility of
diabetic rats to bladder carcinogenesis could be activation of the PI3K pathway and
suppression of p53 in the urothelium in consequence of the above serum protein
alterations
Characteristic Upregulation of Glucose-Regulated Protein 78 in an Early Lesion Negative for Hitherto Established Cytochemical Markers in Rat Hepatocarcinogenesis
Previously, we reported α2-macroglobulin (α2M) to be a
novel marker characteristic of rat hepatocellular preneoplastic and neoplastic
lesions negative for hitherto well-established markers. In the present study, we
further examined other candidate markers with specificity for the same type of
lesions. Glutathione S-transferase-placental form
(GST-P)-negative hepatocellular altered foci (HAF) were generated using a
two-stage (initiation and promotion) carcinogenesis protocol with
N,N-diethylnitrosamine (DEN) and either Wy-14,643 or clofibrate, two peroxisome
proliferators. Microarray analysis using total RNAs isolated from
laser-microdissected GST-P-negative HAF (amphophilic cell foci) and adjacent
normal tissues was conducted along with immunohistochemistry and real-time
RT-PCR. Staining for glucose-regulated protein 78 (GRP78) was detected in
GST-P-negative HAF and hepatocellular adenomas, and slightly increased GRP78
mRNA expression was observed in the lesions by real-time RT-PCR analysis. Thus,
an early increase of GRP78 expression in hepatocarcinogenesis is likely a
feature of the amphophilic subset of HAF
