A Novel P53/POMC/Gas/SASH1 Autoregulatory Feedback Loop and Pathologic Hyperpigmentation
P53-regulated proteins in transcriptional level are associated with many signal transduction pathways and p53 plays a pivotal role in a number of positive and negative autoregulatory feedback loops. Although POMC/α-MSH productions induced by ultraviolet (UV) are directly mediated by p53, p53 is related to UV-independent pathological pigmentation. In the process of identifying the causative gene of dyschromatosis universalis hereditaria (DUH), three mutations encoding amino acid substitutions were found in the gene SAM and SH3 domain containing 1 (SASH1). SASH1 was identified to interact with guanine nucleotide-binding protein subunit-alpha isoforms short (Gαs). However, for about 90 years, the pathological gene and the pathological mechanism of DUH are unclear. Our study indicates that SASH1 is physiologically medicated by p53 upon UV stimulation and a reciprocal SASH-p53 inducement is existed physiologically and pathophysiologically. A novel p53/POMC/α-MSH/Gαs/SASH1 signal cascade regulates SASH1 to foster melanogenesis. SASH1 mutations control a novel p53/POMC/Gαs/SASH1 autoregulatory positive feedback loop to promote pathological hyperpigmentation phenotype in DUH-affected individuals. Our work illustrates a novel p53/POMC/Gαs/SASH1 autoregulatory positive feedback loop that is mediated by SASH1 mutations to foster pathological hyperpigmentation phenotype
Enhanced hydrogen storage properties of LiAlH4 catalyzed by CoFe2O4 nanoparticles
The catalytic effects of CoFe2O4 nanoparticles on the hydrogen storage properties of LiAlH4 prepared by ball milling were investigated. The onset desorption temperature of the LiAlH4 + 2 mol% CoFe2O4 sample is 65 °C, which is 90 °C lower that of the as-received LiAlH4, with approximately 7.2 wt% hydrogen released at 250 °C. The isothermal desorption results show that for the 2 mol% CoFe2O4 doped sample dehydrogenated at 120 °C, 6.8 wt% of hydrogen can be released within 160 min, which is 6.1 wt% higher than that of the as-received LiAlH4 under the same conditions. Through the differential scanning calorimetry (DSC) and the Kissinger desorption kinetics analyses, the apparent activation energy, Ea, of the 2 mol% CoFe2O4 doped sample is calculated as 52.4 kJ mol -1 H2 and 86.5 kJ mol-1 H2 for the first two decomposition processes. This is 42.4 kJ mol-1 H 2 and 86.1 kJ mol-1 H2 lower compared with the pristine LiAlH4, respectively, indicating considerably improved dehydrogenation kinetics by doping the CoFe2O4 catalyst in the LiAlH4 matrix. From the Fourier transform infrared spectroscopy (FTIR) and X-ray diffraction (XRD) analyses, a series of finely dispersed Fe and Co species with a range of valence states, produced from the reactions between LiAlH4 and CoFe2O4, play a synergistic role in remarkably improving LiAlH4 dehydrogenation properties. Strong structural and electronic coupling in metavalent PbS moire superlattices
Moire superlattices are twisted bilayer materials, in which the tunable
interlayer quantum confinement offers access to new physics and novel device
functionalities. Previously, moire superlattices were built exclusively using
materials with weak van der Waals interactions and synthesizing moire
superlattices with strong interlayer chemical bonding was considered to be
impractical. Here using lead sulfide (PbS) as an example, we report a strategy
for synthesizing of moire superlattices coupled by strong chemical bonding. We
use water-soluble ligands as a removable template to obtain free-standing
ultra-thin PbS nanosheets and assemble them into direct-contact bilayers with
various twist angles. Atomic-resolution imaging shows the moire periodic
structural reconstruction at superlattice interface, due to the strong
metavalent coupling. Electron energy loss spectroscopy and theoretical
calculations collectively reveal the twist angle26 dependent electronic
structure, especially the emergent separation of flat bands at small twist
angles. The localized states of flat bands are similar to well-arranged quantum
dots, promising an application in devices. This study opens a new door to the
exploration of deep energy modulations within moire superlattices alternative
to van der Waals twistronics
A synchrotron study of defect and strain inhomogeneity in laser-assisted three-dimensionally-printed Ni-based superalloy
Synchrotron X-ray microdiffraction was employed to investigate the inhomogeneous distribution of defect and residual strain in the transitional region between the dendritic and stray grains in a laser-assisted 3D printed Ni-based superalloy. The dendritic region was found to be under tensile strain transversely to the primary dendrite arm directions. The dendrite edges, where high level of strains and geometrically necessary dislocations were detected, were discerned as low angle grain boundaries. High angle grain boundaries were observed in the stray grain region, and the orientation of the strain tensor in this region varied dramatically at the micron scale, in contrast with the more or less homogeneous distribution in the dendritic region
