    The light scalar mesons as tetraquarks

    We present a numerical solution of the four-quark Bethe-Salpeter equation for ground-state scalar tetraquarks with J^PC = 0^++. We find that the four-body equation dynamically generates pseudoscalar-meson poles in the Bethe-Salpeter amplitude. The resulting tetraquarks are genuine four-quark states that are dominated by pseudoscalar meson-meson correlations. Diquark-antidiquark contributions are subleading because of their larger mass scale. In the light quark sector, the sensitivity of the tetraquark wave function to the pion poles leads to an isoscalar tetraquark mass M_sigma ~ 350 MeV which is comparable to that of the sigma/f0(500). The masses of its multiplet partners kappa and a0/f0 follow a similar pattern. This provides support for a tetraquark interpretation of the light scalar meson nonet in terms of 'meson molecules'.Comment: 7 pages, 4 figures; version accepted by PL

    Increasing the Ratio of PP2A Core Enzyme to Holoenzyme Inhibits Tat-Stimulated HIV-1 Transcription and Virus Production

    AbstractWe demonstrated previously that PP2A exists in many cell types as two abundant forms: (1) holoenzyme composed of two regulatory subunits, A and B, and a catalytic subunit C; and (2) core enzyme consisting of the A and C subunits. These two forms have different substrate specificities. Since published data suggested that HIV-1 transcription may be regulated by a cellular protein phosphatase, it was of interest to determine whether changing the ratio between PP2A core and holoenzyme affects HIV-1 gene expression. This question was addressed by expression in COS cells of an N-terminal mutant of the A subunit, AΔ5, which binds the C but not the B subunit. This resulted in an increase in the amount of core enzyme and a decrease in the amount of holoenzyme concomitant with the expected change in phosphatase activity. Tat-stimulated transcription from the HIV-1 LTR was inhibited 5-fold by mutant AΔ5, whereas mRNA synthesis directed by the actin promoter was not affected. Furthermore, virus production in COS, HeLa, and Jurkat T cells was inhibited 45-, 5-, and 3-fold, respectively, by mutant AΔ5. These results demonstrate that the balance between PP2A holoenzyme and core enzyme is important for HIV-1 gene expression and virus production

    Overweight and Obesity: Risk Factors in Calcium Oxalate Stone Disease?

    Introduction. Several studies showed an association of overweight and obesity with calcium oxalate stone disease (CaOx). However, there are no sufficient data on the influence of body weight on the course of the disease and the recurrence rate. Patients and Methods. N = 100 consecutive stone formers with pure CaOx were studied. Different parameters were investigated. According to the BMI, patients were divided into three groups: (1) BMI ≤ 25; (2) BMI 25.1–30; (3) BMI > 30. Results. N = 32 patients showed a BMI ≤ 25, n = 42 patients showed a BMI of 25.1–30 and n = 26 patients showed a BMI ≥ 30. The groups differed significantly concerning BMI (by definition), urine pH, and urine citrate. The recurrence rate was not significantly different. Discussion. Our study demonstrated that body weight negatively influences single risk factors in CaOx, but obesity is not a predictor for the risk of recurrence in CaOx

    Sensors and Techniques for On-Line Determination of Cell Viability in Bioprocess Monitoring

    In recent years, the bioprocessing industry has experienced significant growth and is increasingly emerging as an important economic sector. Here, efficient process management and constant control of cellular growth are essential. Good product quality and yield can only be guaranteed with high cell density and high viability. Whereas the on-line measurement of physical and chemical process parameters has been common practice for many years, the on-line determination of viability remains a challenge and few commercial on-line measurement methods have been developed to date for determining viability in industrial bioprocesses. Thus, numerous studies have recently been conducted to develop sensors for on-line viability estimation, especially in the field of optical spectroscopic sensors, which will be the focus of this review. Spectroscopic sensors are versatile, on-line and mostly non-invasive. Especially in combination with bioinformatic data analysis, they offer great potential for industrial application. Known as soft sensors, they usually enable simultaneous estimation of multiple biological variables besides viability to be obtained from the same set of measurement data. However, the majority of the presented sensors are still in the research stage, and only a few are already commercially available

    Tocopherol and plastoquinone synthesis in spinach chloroplasts subfractions

    Subfractions isolated from intact purified spinach chloroplasts are able to prenylate the aromatic moiety of -tocopherol and plastoquinone-9 precursors. The biosynthesis of -tocopherol and plastoquinone-9 is a compartmentalized process. The chloroplast envelope membranes are the only site of the enzymatic prenylation in -tocopherol synthesis whereas the thylakoid membrane is also involved in the prenylation and methylation sequence of plastoquinone-9 biosynthesis. A very active kinase which forms phytyl-PP is localized in the stroma. Phytol but not geranylgeraniol is the polyprenol precursor of the side chain of -tocopherol in spinach chloroplasts
