Metallothionein 2A genetic polymorphisms and risk of ductal breast cancer
Metallothioneins (MTs) are a family of metal binding proteins that play an important role in cellular processes such as proliferation and apoptosis. Metallothionein 2A is the most expressed MT isoform in the breast cells. A number of studies have demonstrated increased MT2A expression in various human tumors, including breast cancer. We carried out an association study to examine whether MT2A gene polymorphisms are associated with risk of breast cancer. Information on lifestyle risk factors was collected via a self-administered questionnaire. Genotyping was conducted using polymerase chain reaction–restriction fragment length polymorphism technique. Three single nucleotide polymorphisms (SNP) rs28366003, rs1610216 and rs10636 were genotyped in 534 breast cancer cases and 556 population controls. One SNP in MT2A (rs28366003) showed a positive association with breast cancer. Compared with homozygous common allele carriers, heterozygous for the G variant [odds ratio (OR) = 1.92, 95 % confidence interval (CI):1.28–2.81, p trend <0.01; the OR assuming a dominant model 1.93 (95 % CI: 1.29–2.89, p dominant <0.02) after adjustment for age, family history, smoking status, BMI, menarche, parity, menopausal status and use of contraceptive and menopausal hormones] had a significantly increased risk of breast cancer in Polish population, as well as women with haplotypes, including variant allele of rs28366003 SNP (OR = 1.58, CI: 0.41–6.33, p global = 0.03). Association between the c.*229C>T polymorphism of the topoisomerase IIb binding protein 1 (TopBP1) gene and breast cancer
Topoisomerase IIb binding protein 1 (TopBP1)
is involved in cell survival, DNA replication, DNA damage
repair and cell cycle checkpoint control. The biological
function of TopBP1 and its close relation with BRCA1
prompted us to investigate whether alterations in the
TopBP1 gene can influence the risk of breast cancer.
The aim of this study was to examine the association
between five polymorphisms (rs185903567, rs116645643,
rs115160714, rs116195487, and rs112843513) located in
the 30UTR region of the TopBP1 gene and breast cancer
risk as well as allele-specific gene expression. Five hundred
thirty-four breast cancer patients and 556 population controls
were genotyped for these SNPs. Allele-specific Top-
BP1 mRNA and protein expressions were determined by
using real time PCR and western blotting methods,
respectively. Only one SNP (rs115160714) showed an
association with breast cancer. Compared to homozygous
common allele carriers, heterozygous and homozygous for
the T variant had significantly increased risk of breast
cancer (adjusted odds ratio = 3.81, 95 % confidence
interval: 1.63–8.34, p = 0.001). Mean TopBP1 mRNA and
protein expression were higher in the individuals with the
CT or TT genotype. There was a significant association
between the rs115160714 and tumor grade and stage. Most
carriers of minor allele had a high grade (G3) tumors
classified as T2-T4N1M0. Our study raises a possibility
that a genetic variation of TopBP1 may be implicated in
the etiology of breast cancer
Evaluation of the incidence of abnormal serum concentrations of thyroid stimulating hormone in elderly patients hospitalized in a geriatric ward
Wstęp. Częstość schorzeń tarczycy rośnie wraz z wiekiem, zwłaszcza wśród ludności zamieszkałej na terenach
do niedawna ubogich w jod. Celem pracy była ocena stężeń hormonu tyreotropowego (TSH) w populacji pacjentów
w podeszłym wieku przyjmowanych do leczenia szpitalnego na oddziale geriatrycznym.
Materiał i metody. Retrospektywną analizą objęto 1424 kolejno przyjmowanych do leczenia na oddziale geriatrycznym
pacjentów w wieku 65–99 lat (śr. 76,1 ± 6,7). Ocenie poddano częstość występowania nieprawidłowych
wartości TSH w zależności od wieku i płci oraz współistnienia wybranych schorzeń somatycznych.
Wyniki. Średnie stężenie TSH u kobiet było wyższe w stosunku do obserwowanego u mężczyzn (1,99 ± 5,13
mjm./ml v. 1,64 mjm./ml ± 5,31; p < 0,015). Nie stwierdzono korelacji stężenia surowiczego TSH z wiekiem
badanych. Nieprawidłowe stężenia TSH stwierdzono u 190 badanych (13,34%), także częściej występowały
u kobiet niż u mężczyzn [138 (15,03%) v. 52 (10,27%); p < 0,02]. Supresję TSH stwierdzono u 113 osób
(7,93%), porównywalnie często u kobiet i mężczyzn [odpowiednio: 74 (8,06%) i 39 (7,71%)], natomiast podwyższone
stężenia TSH częściej obserwowano u kobiet [64 (6,97%) i 13 (2,57%); p < 0,001]. Parametry lipidowe
oraz antropometryczne nie wykazywały korelacji z wartościami TSH. Niższe, choć nadal mieszczące się
w granicach normy stężenia TSH stwierdzono u chorych na zapalenie płuc oraz osób cierpiących na cukrzycę.
Wnioski. Ze wzgledu na wysoką częstość występowania nieprawidłowych stężeń TSH ocena tyreometabolizmu
jest koniecznym elementem całościowej oceny geriatrycznej pacjentów przyjmowanych do leczenia na oddziale
geriatrycznym.

in iodine intake until recently. The aim of study was the assessment of serum thyrotropin (TSH) level in the
population of elderly patients admitted for hospitalization in the geriatric ward.
Material and method. The group of 1424 patients consecutive admitted for treatment in the geriatric ward at
the age of 65–99 years (76.1 ± 6.7) was retrospectively estimated. The incidence of abnormal TSH values
depending on age and sex, and co-existence of certain somatic illnesses were assessed.
Results. The average serum thyrotropin level in women was higher compared with men (1.99 ± 5.13 mIU/ml vs
1.64 ± 5.31 mIU/mL, p < 0.015). No correlation between serum TSH level and age was revealed. The abnormal
TSH concentrations in 190 participants (13.34%) were noticed, more commonly in the female group (138
= 15.03% vs 52 = 10.27%). The suppression of serum TSH level in 113 patients (7.93%) was found, equally
often in women (8.06%) and men (7.71%), but TSH elevation in the female group more frequently was observed
(64 [6.97%] vs 13 [2.57%]; p < 0.001). There was no correlation between plasma TSH level and lipoid parameters
and between TSH and anthropometrical indices. Lower, but still within normal limits, serum TSH concentrations
in patients suffering from pneumonia and diabetes mellitus compared to healthy were observed.
Conclusions. Given the high prevalence of abnormal serum TSH concentrations, valuation of thyrometabolism is
a necessary component of an overall assessment of geriatric patients admitted for treatment in a geriatric ward.

Polimorfizm genu Klotho a ryzyko raka pęcherza moczowego
Introduction: Bladder cancer is the most frequent tumor of the urinary tract in Poland. Klotho gene can act as an suppressor gene. Therefore, variability of this gene might be implicated in the carcinogenesis of urinary bladder. The aim of the study was analysis of the association between the g.33590184 G>A (rs1207568), g.33634983 C>T (rs564481), g.33628193 G>C (rs9527025) polymorphisms of the Klotho gene and bladder cancer risk. Materials and methods: The study included 96 patients diagnosed with transitional cell carcinoma of the bladder (TCC) and 114 healthy, cancer-free individuals. Three selected polymorphisms were typed by PCR with confronting two-pair primers (PCR-CTPP) and Real Time PCR with TaqMan probes. Results: The GA and AA genotypes of the rs1207568 polymorphism increased the risk of bladder cancer (OR = 1.86, 95% CI [1.04-3.33], p = 0.03 and OR = 6.58, 95% CI [1.27-34.02], p = 0.01, respectively). Individuals who were heterozygous and homozygous for the A variant had 2.10-fold higher risk of bladder cancer (OR = 2.10, 95% CI [1.20-3.65], p = 0.009). On the other hand, heterozygous subjects and homozygous carriers of the wild-type allele (G) had a decreased bladder cancer risk (OR = 0.19, 95% CI [0.04-0.95], p = 0.043). Also, the occurrence of bladder cancer was positively correlated with the presence of the GC genotype of the rs9527025 polymorphism (OR = 2.84, 95% CI [1.57-5.15], p = 0.0001). Conclusions: Two polymorphisms of Klotho gene (rs1207568 and rs9527025) may play a role in susceptibility to bladder cancer.Wstęp: Rak pęcherza moczowego jest najczęściej występującym nowotworem układu moczowego w Polsce. Sugeruje się, że gen Klotho może pełnić funkcje genu supresorowego. W związku z tym, polimorfizm genu Klotho może mieć wpływ na proces transformacji nowotworowej pęcherza moczowego. Celem przedstawionych badań był analiza związku pomiędzy występowaniem wybranych polimorfizmów pojedynczych nukleotydów g.33590184 G>A (rs1207568), g.33634983 C>T (rs564481), g.33628193 G>C (rs9527025) a ryzykiem zachorowania na raka pęcherza moczowego. Materiały i metody: Do badań włączono 96 pacjentów ze zdiagnozowanym przejściowokomórkowym rakiem pęcherza moczowego (TCC; transitional cell carcinoma) oraz 114 osób zdrowych, u których nie stwierdzono choroby nowotworowej. Występowanie trzech wybranych polimorfizmów analizowano przy użyciu techniki PCR z dwiema parami przeciwstawnych starterów (PCR-CTPP; PCR with confronting two-pair primers) oraz metody Real Time PCR z sondami fluorescencyjnymi TaqMan. Wyniki: Genotypy GA i AA polimorfizmu rs1207568 wpływają na wzrost ryzyka zachorowania na przejściowokomórkowego raka pęcherza moczowego (OR = 1,86, 95% PU [1,04-3,33], p = 0,03 oraz OR = 6,58, 95% CI [1,27-34,02], p = 0,01, odpowiednio). U osób będących heterozygotami lub homozygotami pod względem allela A wykazano ponad 2-krotnie wyższe ryzyko zachorowania na raka pęcherza moczowego (OR = 2,10, 95% PU [1,20-3,65], p = 0,009). Natomiast w przypadku nosicieli allela G, w układzie homozygotycznym lub heterozygotycznym, obserwowano spadek ryzyka zachorowania na badany nowotwór (OR = 0,19, 95% PU [0,04-0,95], p = 0,043). W przypadku polimorfizmu rs952705 wykazano, że genotyp GC zwiększa ryzyko zachorowania na przejściowokomórkowego raka pęcherza moczowego (OR = 2,84, 95% PU [1,57-5,15], p = 0,0001).Wnioski: Dwa spośród badanych polimorfizmów genu Klotho (rs1207568 i rs9527025) mogą mieć wpływ na predyspozycję do zachorowania na przejściowokomórkowego raka pęcherza moczowego
