    Densitometric determination of catecholamine metabolites and 5-hydroxy-indoleacetic acid after two-dimensional thin-layer chromatography on cellulose

    A quantitative two-dimensional chromatographic determination for the catecholamine metabolites vanilglycolic (vanilmandelic) acid, vanilacetic acid, vanillactic acid and vanilglycol is described. The method can also be used for the determination of 5-hydroxy-indoleacetic acid. The analytical procedure consists of the following steps: 1. extraction (after enzymatic hydrolysis when vanilglycol has to be determined); 2. two-dimensional chromatography on thin-layer cellulose; 3. quantitative evaluation of the spots by means of a chromatogram scanner. Standard deviations, recoveries, 24-h excretions in 10 normal adults and results in 10 patient suffering from neurogenic tumors are given. Some of the merits of a multi-component determination of the type described here for the clinical diagnosis of secreting neurogenic and serotonin-producing tumors are discussed

    A technique for two-dimensional electrophoresis of urinary mucopolysaccharides (MPS) is described. The method allows differentiation of a number of mucopolysaccharidoses and is suitable for application in the routine laboratory. This technique should be used to evaluate urines from patients who have a positive MPS-spot test and/or an increased urinary excretion of MPS-bound uronic acid. Urinary MPS excretion patterns are given from normal individuals, patients with the Sanfilippo syndrome and the Hunter syndrome

    Total LDH and LDH isoenzyme distribution in the serum of normal children

    Total LDH activity and LDH isoenzyme distribution were determined in sera of in normal children from 4 to 13 years old and compared to a control group of adult sera. It was found that in children the level of total LDH activity and the isoenzyme distribution did not differ significantly from that in adults. When the children were divided into age groups of one year, such differences could not be detected either. No sex-related differences were found. The relativity of the normal LDH isoenzyme distribution pattern is discussed
