    Environmental Air Pollutants Inhaled during Pregnancy Are Associated with Altered Cord Blood Immune Cell Profiles

    Air pollution exposure during pregnancy may be a risk factor for altered immune maturation in the offspring. We investigated the association between ambient air pollutants during pregnancy and cell populations in cord blood from babies born to mothers with asthma enrolled in the Breathing for Life Trial. For each patient (n = 91), daily mean ambient air pollutant levels were extracted during their entire pregnancy for sulfur dioxide (SO2), nitric oxide, nitrogen dioxide, carbon monoxide, ozone, particulate matter <10 ÎŒm (PM10) or <2.5 ÎŒm (PM2.5), humidity, and temperature. Ninety-one cord blood samples were collected, stained, and assessed using fluorescence-activated cell sorting (FACS). Principal Component (PC) analyses of both air pollutants and cell types with linear regression were employed to define associations. Considering risk factors and correlations between PCs, only one PC from air pollutants and two from cell types were statistically significant. PCs from air pollutants were characterized by higher PM2.5 and lower SO2 levels. PCs from cell types were characterized by high numbers of CD8 T cells, low numbers of CD4 T cells, and by high numbers of plasmacytoid dendritic cells (pDC) and low numbers of myeloid DCs (mDCs). PM2.5 levels during pregnancy were significantly associated with high numbers of pDCs (p = 0.006), and SO2 with high numbers of CD8 T cells (p = 0.002) and low numbers of CD4 T cells (p = 0.011) and mDCs (p = 4.43 × 10−6) in cord blood. These data suggest that ambient SO2 and PM2.5 exposure are associated with shifts in cord blood cell types that are known to play significant roles in inflammatory respiratory disease in childhoo

    IGR J19552+0044: A new asynchronous short period polar: "Filling the gap between intermediate and ordinary polars"

    Based on XMM--Newton X-ray observations IGR J19552+0044 appears to be either a pre-polar or an asynchronous polar. We conducted follow-up optical observations to identify the sources and periods of variability precisely and to classify this X-ray source correctly. Extensive multicolor photometric and medium- to high-resolution spectroscopy observations were performed and period search codes were applied to sort out the complex variability of the object. We found firm evidence of discording spectroscopic (81.29+/-0.01m) and photometric (83.599+/-0.002m) periods that we ascribe to the white dwarf (WD)\ spin period and binary orbital period, respectively. This confirms that IGR J19552+0044 is an asynchronous polar. Wavelength-dependent variability and its continuously changing shape point at a cyclotron emission from a magnetic WD with a relatively low magnetic field below 20 MG. The difference between the WD spin period and the binary orbital period proves that IGR J19552+0044 is a polar with the largest known degree of asynchronism (0.97 or 3%).Comment: 9 pages, 10 figures, A&A accepte

    Evaluation of Visible Diffuse Reflectance Spectroscopy in Liver Tissue: Validation of Tissue Saturations Using Extracorporeal Circulation

    Significance: Real-time information about oxygen delivery to the hepatic graft is important to direct care and diagnose vascular compromise in the immediate post-transplant period. Aim: The current study was designed to determine the utility of visible diffuse reflectance spectroscopy (vis-DRS) for measuring liver tissue saturation in vivo. Approach: A custom-built vis-DRS probe was calibrated using phantoms with hemoglobin (Hb) and polystyrene microspheres. Ex vivo (extracorporeal circulation) and in vivo protocols were used in a swine model (n=15) with validation via blood gas analysis. Results: In vivo absorption and scattering measured by vis-DRS with and without biliverdin correction correlated closely between analyses. Lin’s concordance correlation coefficients are 0.991 for ÎŒa and 0.959 for ÎŒs\u27. Hb measured by blood test and vis-DRS with (R2=0.81) and without (R2=0.85) biliverdin correction were compared. Vis-DRS data obtained from the ex vivo protocol plotted against the PO2 derived from blood gas analysis showed a good fit for a Hill coefficient of 1.67 and P50=34  mmHg (R2=0.81). A conversion formula was developed to account for the systematic deviation, which resulted in a goodness-of-fit (R2=0.76) with the expected oxygen dissociation curve. Conclusions: We show that vis-DRS allows for real-time measurement of liver tissue saturation, an indicator for liver perfusion and oxygen delivery

    Objective: Offspring born to mothers with asthma in pregnancy are known to have lower lung function which tracks with age. Human group 2 innate lymphoid cells (ILC2) accumulate in foetal lungs, at 10-fold higher levels compared to adult lungs. However, there are no data on foetal ILC2 numbers and the association with respiratory health outcomes such as lung function in early life. We aimed to investigate cord blood immune cell populations from babies born to mothers with asthma in pregnancy. Methods: Cord blood from babies born to asthmatic mothers was collected, and cells were stained in whole cord blood. Analyses were done using traditional gating approaches and computational methodologies (t-distributed stochastic neighbour embedding and PhenoGraph algorithms). At 6 weeks of age, the time to peak tidal expiratory flow as a percentage of total expiratory flow time (tPTEF/tE%) was determined as well as Lung Clearance Index (LCI), during quiet natural sleep. Results: Of 110 eligible infants (March 2017 to November 2019), 91 were successfully immunophenotyped (82.7%). Lung function was attempted in 61 infants (67.0%), and 43 of those infants (70.5% of attempted) had technically acceptable tPTEF/tE% measurements. Thirty-four infants (55.7% of attempted) had acceptable LCI measurements. Foetal ILC2 numbers with increased expression of chemoattractant receptor-homologous molecule (CRTh2), characterised by two distinct analysis methodologies, were associated with poorer infant lung function at 6 weeks of age." Conclusion: Foetal immune responses may be a surrogate variable for or directly influence lung function outcomes in early life

    Sustaining entrepreneurial business: a complexity perspective on processes that produce emergent practice

    This article examines the management practices in an entrepreneurial small firm which sustain the business. Using a longitudinal qualitative case study, four general processes are identified (experimentation, reflexivity, organising and sensing), that together provide a mechanism to sustain the enterprise. The analysis draws on concepts from entrepreneurship and complexity science. We suggest that an entrepreneur’s awareness of the role of these parallel processes will facilitate their approaches to sustaining and developing enterprises. We also suggest that these processes operate in parallel at multiple levels, including the self, the business and inter-firm networks. This finding contributes to a general theory of entrepreneurship. A number of areas for further research are discussed arising from this result

    Boron and Lithium in Calcium Sulfate Veins: Tracking Precipitation of Diagenetic Materials in Vera Rubin Ridge, Gale Crater

    The NASA Curiosity rover’s ChemCam instrument suite has detected boron in calcium‐sulfate‐filled fractures throughout the sedimentary strata of Gale crater including Vera Rubin ridge (VRR). The presence of elevated B concentration provides insights into Martian subsurface aqueous processes. In this study we extend the dataset of B in Ca‐sulfate veins across Gale crater, comparing the detection frequency and relative abundances with Li. We report 33 new detections of B within veins analyzed between sols 1548 and 2311 where detections increase in Pettegrove Point and Jura members, which form VRR. The presence of B and Li in the Ca‐sulfate veins is possibly due to dissolution of pre‐existing B in clays of the bedrock by acids or neutral water and redistribution of the elements into the veins. Elevated frequency of B detection in veins of Gale crater correlate with presence of dehydration features such as desiccation cracks, altered clay minerals and detections of evaporites such as Mg‐sulfates, chloride salts in the host rocks. The increased observations of B also coincide with decreased Li concentration in the veins (average Li concentration of veins drops by ~15 ppm). Boron and Li have varying solubilities and Li does not form salts as readily upon dehydration as B, causing it to remain in the solution. So, the weak negative correlation between B and Li may reflect the crystallization sequence during dehydration on Vera Rubin ridge

    Empowerment or Engagement? Digital Health Technologies for Mental Healthcare

    We argue that while digital health technologies (e.g. artificial intelligence, smartphones, and virtual reality) present significant opportunities for improving the delivery of healthcare, key concepts that are used to evaluate and understand their impact can obscure significant ethical issues related to patient engagement and experience. Specifically, we focus on the concept of empowerment and ask whether it is adequate for addressing some significant ethical concerns that relate to digital health technologies for mental healthcare. We frame these concerns using five key ethical principles for AI ethics (i.e. autonomy, beneficence, non-maleficence, justice, and explicability), which have their roots in the bioethical literature, in order to critically evaluate the role that digital health technologies will have in the future of digital healthcare
