The Influence of Carbonate Ions on the Structure of the Electrical Double Layer at the Interface of Hydroxyapatite/Electrolyte Solution
The aim of the work was to investigate the changes in the double electrical layer at the hydroxyapatite containing different amount of carbonate ions/electrolyte solution. Besides, the main properties of the edl (electrical double layer), i.e. surface charge density and zeta potential were determined by electrophoresis measurements and potentiometer titration, respectively. The synthesized adsorbents were characterized by the following methods: X-ray diffraction, FTIR (Fourier Transform Infrared Spectroscopy), adsorption and desorption of nitrogen and scanning electron microscopy. The analyzed samples had different structures and particle sizes. It was proved that increase in the carbonate groups content is connected with the decrease of apatite grain sizes and crystallinity reduction. The characteristic parameters of the electric double layer were also different: pHpzc (point zero of charge) of hydroxyapatite was 6.5 whereas for carbonate apatite was higher – pHpzc = 8. Mitogenic potency of insulin glargine
The goal of diabetes mellitus treatment is to maintain long-term near-normoglycaemia to prevent the onset or progression of long-term
complications. In order to achieve tight glycaemic control and improve the quality of life for diabetic patients, a number of novel insulin
preparations, insulin analogues, have been constructed thanks to recombinant DNA technologies and advanced protein chemistry. Because
structurally modified insulins may differ from human insulin not only in metabolic but also in mitogenic potencies there were
concerns raised about the possibility of increased insulin analogue proliferative action or tumourigenesis. In vitro and in vivo studies on
insulin analogues in comparison to endogenous insulin have been performed to closely monitor the insulin analogue action profiles.
Insulin glargine was the only one presenting a significant increase in affinity to insulin-like growth factor type 1 (IGF-1) receptor. However,
there was controversy regarding the safety of insulin glargine use because of its potential risk of mitogenicity but it proved to be true
only for human osteosarcoma cells Saos/B10. Outcomes of the studies performed on lines other than cancer cells and on animals did not
present any increased mitogenic activity nor mitogenic potency of insulin glargine in comparison to human insulin.Celem leczenia cukrzycy jest uzyskanie i stałe utrzymywanie stanu zbliżonego do normoglikemii w celu zapobiegania powstawaniu
i progresji późnych powikłań naczyniowych. Aby uzyskać pełną kontrolę gikemii i poprawić jakość życia pacjentów chorujących na
cukrzycę, opracowano, dzięki wykorzystaniu technik rekombinacji DNA i zaawansowanych metod modyfikacji białek, wiele nowych
preparatów insulinowych - analogów insuliny. Ponieważ strukturalnie zmodyfikowana cząsteczka insuliny może różnić się od cząsteczki
ludzkiej insuliny nie tylko wpływem na metabolizm, ale również właściwościami mitogennymi, dlatego zwrócono uwagę na istnienie
ryzyka podwyższonej aktywności proliferacyjnej analogów insuliny. W celu dokładnej oceny profilu aktywności insulin analogowych
porównano je, zarówno w badaniach in vitro, jak i in vivo, z insuliną ludzką. Glargina okazała się być jedyną insuliną, która charakteryzuje się
znamiennie zwiększonym powinowactwem do receptora insulinopodobnego czynnika wzrostu 1 (IGF-1, insulin-like growth factor type 1).
Mimo że istniały kontrowersje dotyczące bezpieczeństwa stosowania glarginy z uwagi na ryzyko działania mitogennego, to okazało się
ono istotne jedynie w badaniach in vitro w odniesieniu do ludzkiej linii komórek kostniakomięsaka Saos/B10. Wyniki pozostałych badań
nie wykazały zwiększonej aktywności mitogennej lub potencjału mitogennego w porównaniu z insuliną ludzką
Influence of the Modifier Type and its Concentration on Electroosmotic Flow of the Mobile Phase in Pressurized Planar Electrochromatography
The aim of this work was to find a relationship between electroosmotic flow (EOF) velocity of the mobile phase in pressurized planar electrochromatography (PPEC) and physicochemical properties like zeta potential, dielectric constant, and viscosity of the mobile phase as well as its composition. The study included different types of organic modifiers (acetonitrile, methanol, ethanol, acetone, formamide, N-methylformamide and N,N-dimethylformamide) in the full concentration range. In all experiments, chromatographic glass plates HPTLC RP-18 W from Merck (Darmstadt) were used as a stationary phase. During the study we found that there is no linear correlation between EOF velocity of the mobile phase and single variables such as zeta potential or dielectric constant or viscosity. However, there is quite strong linear correlation between EOF velocity of the mobile phase and variable obtained by multiplying zeta potential of the stationary phase–mobile phase interface, by dielectric constant of the mobile phase solution and dividing by viscosity of the mobile phase. Therefore, it could be concluded that the PPEC system fulfilled the Helmholtz–Smoluchowski equation
Dunnigan-type familial partial lipodystrophy associated with the heterozygous R482W mutation in LMNA gene — case study of three women from one family
Lipodystrofie są heterogenną grupą chorób dotyczących rozmieszczenia tkanki tłuszczowej w organizmie. Rodzinna częściowa lipodystrofiatypu Dunnigana (FPLD, familial partial lipodystrophy of the Dunnigan type) jest rzadkim autosomalnie dominującym schorzeniem, któregoprzyczyną jest mutacja braku sensu w genie laminy A/C (gen LMNA). W chorobie tej dochodzi do selektywnej utraty podskórnej tkankitłuszczowej kończyn i tułowia oraz jej kumulacji w okolicy karku i twarzy, co zwykle jest związane z występowaniem różnych schorzeńmetabolicznych, w tym insulinooporności, cukrzycy, dyslipidemii, stłuszczenia wątroby i nadciśnienia tętniczego. W przedstawionymraporcie opisujemy kliniczne i molekularne cechy trzech polskich kobiet pochodzących z jednej rodziny charakteryzujących się fenotypemFLPD (matka i jej dwie córki). Obecność FLPD rozpoznano w trakcie leczenia cukrzycy na podstawie sekwencjonowania genu LMNAi ujawnienia heterozygotycznej mutacji R482W. W celu rozpoznania cukrzycy monogenowej związanej z lipodystrofią ważne jest przeprowadzenieszczegółowego badania fizykalnego w toku diagnostyki cukrzycy oraz świadomość konieczności diagnostyki genetycznejwątpliwych przypadków. Prawidłowa diagnostyka różnicowa cukrzycy jest niezbędna w celu ustalenia strategii leczenia, przewidywaniaprogresji choroby, określenia rokowania, a także jest potrzebna w celu wzmożenia czujności diagnostycznej w odniesieniu do rodzinyosoby będącej nosicielem mutacji.Lipodystrophies are a heterogeneous group of diseases affecting adipose tissue distribution. Familial partial lipodystrophy of the Dunnigantype (FPLD) is a rare autosomal, dominant disorder caused by missense mutations in lamin A/C (LMNA) gene where selectiveloss of subcutaneous adipose tissue from the limbs and trunk, and accumulation of fat in the neck and face, is usually associated witha variety of metabolic disorders including insulin resistance, diabetes mellitus, dyslipidemia, hepatic steatosis and high blood pressure.In this report we present clinical and molecular features of three Polish women with FLPD phenotype coming from one family (a motherand her two daughters). FPLD was recognised under the circumstances of diabetes treatment, where sequencing of LMNA gene revealedheterozygous R482W mutation. In order to be able to recognise monogenic diabetes associated with lipodystrophy, it is important to bevery precise in physical examination while diagnosing diabetes and to be aware of the necessity of performing genetic testing. Diabetesappropriatedifferential diagnosis is essential for the treatment strategy, anticipation of the disease progression, and determination of theprognosis. The polymorphism in insulin receptor substrate-1 gene and birth weight in neonates at term
Wstęp: Mutacja genu substratu 1 receptora insuliny (IRS-1, insulin receptor substrate-1) jest jednym z genetycznych czynników ryzyka,
przypuszczalnie związanych z występowaniem zjawiska oporności na insulinę lub predyspozycją do wystąpienia cukrzycy typu 2. Celem
niniejszego badania była ocena potencjalnych zależności między polimorfizmem Gly972Arg w genie IRS-1 a masą ciała u noworodków
urodzonych o czasie.
Materiał i metody: W badaniu wzięło udział 100 noworodków urodzonych o czasie (38-42 tydzień ciąży), których matki nie chorowały
podczas ciąży. Po wyekstrahowaniu genomowego DNA z leukocytów krwi pępowinowej przeprowadzono z użyciem metody PCR ocenę
polimorfizmu Gly972Arg genu IR-1.
Wyniki: Urodzeniowa masa ciała była istotnie niższa u noworodków z polimorfizmem Gly972Arg genu IRS-1 w porównaniu z grupą
kontrolną (3161,75 ± 380,86 g vs. 3427,92 ± 468,86 g). U noworodków z tym polimorfizmem zaobserwowano również mniejszą długość
ciała oraz mniejszy obwód głowy (odpowiednio: 54,38 ± 3,13 cm vs. 52,69 ± 2,91 cm oraz 34,08 ± 1,47 cm vs. 33,63 ± 0,81 cm).
Wnioski: Wyniki sugerują, że genotyp Gly972Arg u noworodków urodzonych o czasie wiąże się z niższą urodzeniową masą ciała, mniejszą
długością ciała oraz mniejszym obwodem głowy.Background: The mutation of the IRS-1 gene is one of the genetic risk factors which, it is speculated, is associated with insulin resistance
or predisposition to type 2 diabetes. The aim of our study was to evaluate the association between the Gly972Arg polymorphism in the
IRS-1 gene and birth weight in newborn children with adequate gestational age.
Material and methods: 100 newborn children with adequate gestational age (38–42 weeks), whose mother had no disorders during
pregnancy, were studied. Genomic DNA was extracted from umbilical cord blood leukocytes, and Gly972Arg polymorphism in the IRS-1
gene was genotyped using the PCR-based method.
Results: Birth weight was significantly lower in the newborn with the IRS-1 Gly972Arg polymorphism compared with a control group
(3161.75 ± 380.86 g vs. 3427.92 ± 468.86 g). Body length and head circumference at birth were also lower in the neonates with that polymorphism
(54.38 ± 3.13 cm vs. 52.69 ± 2.91 cm, and 34.08 ± 1.47 vs. 33.63 ± 0.81, respectively).
Conclusions: The results suggest that the Gly972Arg genotype is associated with lower birth weight, body length and head circumference
in neonates with adequate gestational age
Association of single nucleotide polymorphism (rs741301) of the ELMO1 gene with diabetic kidney disease in Polish patients with type 2 diabetes: a pilot study
Introduction: Multifactorial pathogenesis of diabetic kidney disease (DKD) consists of a combination of metabolic, environmental, and genetic factors. A genome-wide association study has shown that ELMO1 is a candidate gene for DKD occurrence and progression. The aim of this study was to assess the association of a single nucleotide polymorphism (rs741301) of the ELMO1 gene with DKD in Polish patients with type 2 diabetes (T2DM).
Material and methods: This was a case/control study of 272 T2DM patients with or without DKD. Patients were divided into groups depending on DKD definition according to the American Diabetes Association (ADA) and the National Kidney Foundation (NKF). The association of the rs741301 polymorphism with DKD was assessed in the whole study group as well as in the subgroups stratified according to the presence of DKD.
Results: There was no association between rs741301 polymorphisms and the presence of DKD in relation to the ADA definition (p = 0.6) or the NKF definition (p = 0.5) of DKD and with estimated glomelural filtration rate (eGFR) value reflecting the stage of the chronic kidney disease (p = 0.8).
Conclusions: Even though the results of this study are negative, there is still a great need for larger studies assessing the genetic susceptibility to DKD to identify patients who are particularly prone to this complication
Improvement of the type 2 diabetes management - role of the innovative therapies taking DPP-4 inhibitors as an example
Chorobowość cukrzycy typu 2 w Polsce oceniono na
1,6–3,7%. Około 90–95% wszystkich chorych na świecie
stanowią pacjenci z cukrzycą typu 2, a prognozy
na 2025 rok wskazują, że liczba chorych może sięgnąć
300 mln osób. Chorzy na cukrzycę są 3-krotnie
częściej hospitalizowani niż osoby bez cukrzycy. Około
70–80% chorych na cukrzycę umiera wskutek
powikłań sercowo-naczyniowych, co prowadzi do
skrócenia życia o 4–10 lat. Całkowite bezpośrednie
koszty medyczne cukrzycy typu 2 w Polsce w 2002 roku
oszacowano na 2,6 mld zł, co stanowiło 8% wydatków
poniesionych na opiekę zdrowotną. Epidemia
cukrzycy typu 2 uzasadnia umieszczenie profilaktyki
i leczenia cukrzycy wśród priorytetów polityki zdrowotnej.
W Polsce pacjenci mają ograniczony dostęp
do nowoczesnych terapii i procedur stosowanych
w leczeniu cukrzycy. Cele leczenia cukrzycy określone
przez Polskie Towarzystwo Diabetologiczne (PTD)
mogą być realizowane tylko na podstawie wykorzystania wszystkich dostępnych metod terapeutycznych,
w tym innowacyjnych. Ze względu na naturalny
przebieg choroby uzyskanie oraz utrzymanie optymalnej
kontroli glikemii nadal pozostaje trudnym
wyzwaniem. Mimo możliwości stosowania wielu
doustnych leków i nowych schematów insulinoterapii,
zwraca się uwagę na nowe klasy preparatów
przeciwcukrzycowych, jak najbardziej zbliżonych
w swym działaniu do fizjologicznego ideału. Inhibitory
DPP-IV należą do najnowszych leków doustnych
stosowanych w leczeniu cukrzycy typu 2, skutecznie
obniżających glikemię, przy dobrej tolerancji, neutralnym
wpływie na masę ciała oraz bardzo małym
ryzyku wystąpienia hipoglikemii. Stanowią jedną
z najbardziej atrakcyjnych metod poprawy kontroli
cukrzycy typu 2, zwłaszcza w skojarzeniu z metforminą,
dlatego znalazły swoje miejsce w zaleceniach
klinicznych PTD na 2009 rok. Wyniki analiz farmakoekonomicznych
wskazują również, że dołączenie inhibitora
DPP-4 (sitagliptyny) do metforminy jest efektywną
kosztowo opcją wobec stosowania pochodnych
sulfonylomocznika, przy koszcie 6,7 tys. zł za
jednostkę QALY. Nakłady na poprawę kontroli cukrzycy
typu 2 przy zastosowaniu innowacyjnych leków
mogą stanowić bardziej opłacalną strategię niż
znacznie bardziej kosztowna terapia odległych powikłań.The prevalence of type 2 diabetes in Poland is estimated
at 1.6-3.7%. Type 2 diabetes is diagnosed in
about 90-95% of the cases worldwide. The prognosis
alarms, that number of type 2 diabetes might
reach even 300 million patients in the year 2025.
Hospital admissions among diabetic patients exceed
three folds admissions in non-diabetic patients.
About 70-80% incidences of death in diabetic patients
results from the cardio-vascular complications.
These decrease life expectancy by 4-10 years. The
total direct medical costs of type 2 diabetes in the
year 2002 in Poland reached 2.6 billion PLN which
constituted 8% of the total health care spending.
The epidemic characteristic of the disease justify inclusion
of the type 2 diabetes management in the
health care policy priorities. The access of innovations
in type 2 diabetes treatment in Poland is limited.
The treatment goals determined by the Polish
Society of Diabetology could only be executed with
the use of all the available therapeutic options including
also innovative drugs. The optimal glicemic
control and maintenance of effectiveness in type 2
diabetes patients still constitutes the main therapeutic
concern. Despite the currently used oral anti-diabetics
and increasing administration of novel insulin
therapies, there is plenty of scope for improvement
with the use of innovative drugs acting through
the mechanism close to the physiological reactions.
The DPP-4 inhibitors constitute new therapeutic class
of oral anti-diabetics effective in terms of gilcemic
control and well tolerated. Moreover, DPP-4 have
neutral influence on the body weight and their use
is associated with very low risk of hypogycemy. Therefore
DPP-4 give an opportunity to improve type 2
diabetes management, particularly in combination
with metformine. As a result of their therapeutic
added value, DPP-4 have been included into therapeutic
guidelines of Polish Society of Diabetology issued
in the year 2009. Results of the pharmacoeconomics
evaluations, proved, that use of the DPP-4
(sitagliptine) as ad on therapy to metformine is cost-effective as compare to sulfonylourea addition. The
calculated cost per QALY amounted to 6.7 thousand
PLN in Polish setting. The increase of funds on innovative
anti-diabetic drugs could constitute more profitable
strategy than much more costly treatment of
the future complications related to type 2 diabetes
