    Development of a Novel Tissue Blot Hybridization Chain Reaction for the Identification of Plant Viruses

    Assays for the high throughput screening of crops for virus monitoring need to be quick, easy, and low cost. One method involves using tissue blot immunoassays (TBIA), where plant stems are blotted onto nitrocellulose membrane and screened with available antibodies against a range of viruses. TBIAs are inexpensive but limited by antibody availability and specificity. To circumvent the antibody limitations, we developed the tissue blot hybridization chain reaction (TB-HCR). As with TBIA, plant stems are blotted onto a nitrocellulose membrane, however, TB-HCR involves using nucleic acid probes instead of antibodies. We demonstrated for the first time that TB-HCR can be used for plant viruses by designing and testing probes against species from several virus genera including begomovirus, polerovirus, luteovirus, cucumovirus, and alfamovirus. We also explored different hairpin reporter methods such as biotin/streptavidin-AP and the Alexa Fluor-488 Fluorophore. TB-HCR has applications for low-cost diagnostics for large sample numbers, rapid diagnostic deployment for new viruses, and can be performed as a preliminary triage assay prior to downstream applications

    Modified group projector technique for induced representations is a powerful tool for calculation and symmetry quantum numbers assignation of a tight binding Hamiltonian energy bands of crystals. Namely, the induced type structure of such a Hamiltonian enables efficient application of the procedure: only the interior representations of the orbit stabilizers are to be considered. Then the generalized Bloch eigen functions are obtained naturally by the expansion to the whole state space. The method is applied to the electronic pi-bands of the single wall carbon nanotubes: together with dispersion relations, their complete symmetry assignation by the full symmetry (line) groups and the corresponding symmetry-adapted eigen function are found.Comment: 10 pages 1 figur

    Influence of summer temperatures on basic economic and tourism indicators of the middle mediterranean

    The Middle Mediterranean is characterized by long, hot, and dry summers, significant historical and cultural values, and the warm Mediterranean sea, making it attractive for coastal tourism. Given these characteristics, the goal of our paper is to analyze the influence of summer temperatures in the region of the Middle Mediterranean on the values of underlying economic and tourism indicators. The method of simple linear correlation and regression was used. Based on the results of testing, we came to the conclusion that the temperatures in the summer months have no significant influence on selected economic and tourism indicators. Also, we conclude that social factors have the greatest influence on these indicators. The coefficients of variation are calculated in the observed period to analyze the variability of the tested values. It could not be identified a statistically significant relationship of indicators with summer temperatures

    Adapting the cognitive walkthrough method to assess the usability of a knowledge domain visualization

    The usability of knowledge domain visualization (KDViz) tools can be assessed at several levels. Cognitive Walkthrough (CW) is a well-known usability inspection method that focuses on how easily users can learn software through exploration. Typical applications of CW follow structured tasks where user goals and action sequences that lead to achievement of the goals are well-defined. KDViz and other information visualization tools, however, are typically designed for users to explore data and user goals and actions are less well understood. In this paper, we describe how the traditional CW method may be adapted for assessing the usability of these systems. We apply the adapted version of CW to CiteSpace, a KDViz tool that uses bibliometric analyses to create visualizations of scientific literatures. We describe usability issues identified by the adapted CW and discuss how CiteSpace supported the completion of tasks, such as identifying research fronts, and the achievement of goals. Finally, we discuss improvements to the adapted CW and issues to be addressed before applying it to a wider range of KDViz tools

    Effectiveness of some crown compounds on inhibition of polyphenoloxidase in model systems and in apple

    Enzymatic browning is (in most cases) an undesirable reaction which usually impairs the sensory properties and chemical changes in raw fruits and vegetables after mechanical operations (such as peeling, coring or slicing). A great emphasis is put on research to develop new methods to prevent enzymatic browning especially in fresh-cut (minimally processed) fruits and vegetables. The inhibition effect of crown compounds, macrocyclic ethers, benzo-18-crown-6 with sorbic acid and benzo-18-crown-6 with potassium sorbate, on polyphenoloxidase (PPO) activity was studied. The effectiveness of these compounds was evaluated by using 3,4-dihydroxy phenylalanine (L-DOPA), and chlorogenic acid (3-o-caffeoyl-D-quinic acid), the most widespread natural PPO substrates in fruits and vegetables, as well as browning inhibition substances on the cut surface of apples. Results showed that crown compounds used in this study were effective, both as inhibitors of the oxidation of phenolic compounds (PPO substrates) in model solutions and as inhibitors of enzyme discolorations of real systems (fresh-cut apples).
