    Spectral luminescence properties of indotricarbocyanine dye in biological tissues

    We have established that the shape and position of the maximum in the fluorescence spectrum of an indotricarbocyanine dye in tumor and normal tissues in vivo change over time after intravenous injection of the dye. Based on analysis of the spectral properties of the dye in vivo and in blood plasma, the dependence of the properties on the time since injection has shown that in the living body, the environment of the dye molecule changes as the photosensitizer goes from the skin into the tissue. We have established that in tissues in vivo, the dye molecules are localized in a region with low dielectric constant of the medium. We have shown that the change in the ratio of the concentrations of the different forms of hemoglobin in the blood has an effect on the absorption and shape of the fluorescence spectrum of the dye in tissues in vivo

    Метою статті є дослідження інфляційних процесів в Україні на підґрунті аналізу динаміки річного індексу споживчих цін. У зв’язку з кризовими явищами в економіці проблема стримування надмірного зростання цін стає найважливішою умовою реалізації соціально-економічної та грошово-кредитної політики держави. Досліджується вплив на зростання індексу споживчих цін наступних макроекономічних показників: обсяг валового внутрішнього продукту, курс гривні по відношенню до долара США, розмір середньої заробітної плати в Україні. Із використанням прикладних пакетів обґрунтовано, що динаміка індексу споживчих цін характеризується випадковою складовою і не може бути апроксимованою елементарними функціями, що залежать лише від часу. За допомогою електронних таблиць MS EXCEL побудовано математичну модель у вигляді множинного рівняння регресії. На підґрунті побудованої моделі доведено, що найбільше впливає на зростання індексу споживчих цін підвищення курсу національної валюти. Результати дослідження можуть бути використані при прогнозуванні річного рівня інфляції на наступний період. Обчислення індексу споживчих цін на 2021 рік на основі побудованої моделі показало статистично надійний результат.The aim of the article is to study inflationary processes in Ukraine based on the analysis of the dynamics of the annual consumer price index. Due to the crisis in the economy, the problem of curbing excessive price increases is becoming an important condition for the implementation of socio-economic and monetary policy of the state. The influence of the following macroeconomic indicators on the growth of the consumer price index is studied: the volume of gross domestic product, the exchange rate of the hryvnia against the US dollar, the size of the average wage in Ukraine. The main statistical characteristics of each indicator for the studied period are found. The growth rates of factors are calculated and their graphs are constructed. Using application packages, it is substantiated that the dynamics of the consumer price index is characterized by a random component and cannot be approximated by elementary functions that depend only on time. Using spreadsheets MS EXCEL built a mathematical model in the form of a multiple regression equation. The adequacy of the model was checked using Fisher’s criteria and finding the multiple correlation coefficient, the significance of the parameters according to Student’s criterion. Due to the different sizes of the studied variables, the power function was used, which allowed to consider the growth rates of the traits. Based on the built model, it is proved that the growth of the national currency rate has the greatest impact on the growth of the consumer price index. The results of the study can be used to predict the annual inflation rate for the next period. Also, the regression equation can be used to calculate the hryvnia exchange rate, which will provide the desired level of inflation. The calculation of the consumer price index for 2021 on the basis of the built model showed a statistically reliable result

    Spectral Properties of Thioflavin T and Its Complexes with Amyloid Fibrils

    Comparative analysis of the absorption and fluorescence spectra and fluorescence excitation spectra of thioflavin T (ThT) in various solvents and in the composition of amyloid fibrils has shown that ThT, when excited in the region of the long-wavelength absorption band, fluoresces in the spectral region with a maximum at 478–484 nm. The appearance in aqueous and alcohol solutions of a fluorescence band with a maximum near 440 nm has been attributed to the presence in the composition of the ThT preparations of an impurity with an absorption band in the 340–350-nm range. The literature data showing that in glycerol ThT has a wide fluorescence spectrum with two maxima are due to the artifact connected with the use of a high concentration of the dye. It has been suggested that the cause of the low quantum yield of ThT aqueous and alcohol solutions is the breakage of the system of conjugated bonds due to the reorientation of the benzothiozole and benzaminic rings of ThT in the excited state with respect to one another. The main factor determining the high quantum yield of fluorescence of ThT incorporated in fibrils is the steric restriction of the rotation of the rings about one another under these conditions. The suggestions made have been verified by the quantum-chemical calculation of the ThT molecule geometry in the ground and excited states

    Features of the cytokine profile in adolescents with microvascular complications of type 1 diabetes mellitus

    Despite advances of modern medical science, the consequences associated with management of complications in type 1 diabetes mellitus (DM1) in children and adolescents represent a serious problem. Common development of microvascular diabetic complications (retinopathy, neuropathy, kidney damage) still remains a sufficient obstacle for achieving high quality of life and social adaptation in the young patients, thus promoting studies of immune mechanisms involved in genesis of microvasculature damage under the conditions of dysmetabolic abnormalities associated with DM1. Our goal was to assess the role of altered cytokine balance in blood serum in development of microangiopathies in adolescents with DM1.140 adolescent patients with type 1 diabetes aged 14-18 years were examined being divided in 2 groups: group I included the patients with glycated hemoglobin (HbA1c) level of > 9.0% (n = 65), and group II which included adolescents with HbA1C level of ≤ 9.0% (n = 75). Each group was divided into subgroups: Ia (n = 50) and IIa (n = 38) included adolescents with diabetic retinopathy, nephropathy or neuropathy, whereas groups Ib (n = 15) and IIb (n = 37) were without microvascular complications. The control group consisted of 36 adolescents with normal body weight, without carbohydrate metabolic disorders, and family history of diabetes mellitus. Determination of TNFα, IL-1β, VCAM-1, fractalkine levels in blood serum was performed by enzyme immunoassay using test systems “RayBiotech” (USA), “BIOSCIENCE” (USA).Development of microangiopathies in adolescents with different glycemic control is associated with increased serum concentration of the factors involved in neoangiogenesis and vascular wall remodeling, i.e., TNFα, IL-1β, VCAM-1, compared with control group (p < 0.05), and a statistically significant decrease in fractalkine level in adolescent patients with either complicated, or uncomplicated DM1. The study allowed us to suggest that occurrence of microvascular complications in adolescents with DM1 is associated with impaired immune response tending for altered cytokine balance towards Th1 type, enhanced intercellular interactions, imbalance of bioregulatory molecules, contributing to development of inflammatory immunoregulatory state. The revealed patterns of laboratory markers, along with assessment of metabolic indices, will enable personalized approaches to early diagnostics of microvascular complications in adolescents with DM1 and prevent their further progression

    Fluorescence Quantum Yield of Thioflavin T in Rigid Isotropic Solution and Incorporated into the Amyloid Fibrils

    In this work, the fluorescence of thioflavin T (ThT) was studied in a wide range of viscosity and temperature. It was shown that ThT fluorescence quantum yield varies from 0.0001 in water at room temperature to 0.28 in rigid isotropic solution (T/η→0). The deviation of the fluorescence quantum yield from unity in rigid isotropic solution suggests that fluorescence quantum yield depends not only on the ultra-fast oscillation of ThT fragments relative to each other in an excited state as was suggested earlier, but also depends on the molecular configuration in the ground state. This means that the fluorescence quantum yield of the dye incorporated into amyloid fibrils must depend on its conformation, which, in turn, depends on the ThT environment. Therefore, the fluorescence quantum yield of ThT incorporated into amyloid fibrils can differ from that in the rigid isotropic solution. In particular, the fluorescence quantum yield of ThT incorporated into insulin fibrils was determined to be 0.43. Consequently, the ThT fluorescence quantum yield could be used to characterize the peculiarities of the fibrillar structure, which opens some new possibilities in the ThT use for structural characterization of the amyloid fibrils

    Extrinsic Fluorescent Dyes as Tools for Protein Characterization

    Noncovalent, extrinsic fluorescent dyes are applied in various fields of protein analysis, e.g. to characterize folding intermediates, measure surface hydrophobicity, and detect aggregation or fibrillation. The main underlying mechanisms, which explain the fluorescence properties of many extrinsic dyes, are solvent relaxation processes and (twisted) intramolecular charge transfer reactions, which are affected by the environment and by interactions of the dyes with proteins. In recent time, the use of extrinsic fluorescent dyes such as ANS, Bis-ANS, Nile Red, Thioflavin T and others has increased, because of their versatility, sensitivity and suitability for high-throughput screening. The intention of this review is to give an overview of available extrinsic dyes, explain their spectral properties, and show illustrative examples of their various applications in protein characterization
