    In this paper I want to focus on working with actors. How we tried to reach such extreme emotions and behaviour as described above. I will describe the process of rehearsals with the two main actors and the problems we met on the road. Its important to mention that the idea for this kind of film was also strongly inspired by my wish to work with actors on an extremely emotional level, where we would all more or less lose control

    The thesis will focus on analysing what “ The Life Of Jesus” is about and how this is portrayed through the main character, Freddy. It will discuss the concept of the filmie character as a “common human being”, who does not have access to intellectual reflection and verbal articulation of emotional conflict and crisis, but who, at the same time, is capable of feeling a great deal. He feels the emotional conflict and crisis that make up the story. I will also attempt to step inside the same filmic character itself and analyse how to visualize the morals, ethics and emotions that the character cant verbalize or communicate in any direct or intellectual way. Bruno Dumont speaks about filming the inside of a person as his highest dream. I will discuss film directing using The Life Of Jesus as an example, but I will also draw on my own experiences as a filmmaker

    Performance of p16INK4a ELISA as a primary cervical cancer screening test among a large cohort of HIV-infected women in western Kenya: a 2-year cross-sectional study.

    ObjectiveA biomarker with increased specificity for cervical dysplasia compared with human papillomavirus (HPV) testing would be an attractive option for cervical cancer screening among HIV-infected women in resource-limited settings. p16(INK4a) has been explored as a biomarker for screening in general populations.DesignA 2-year cross-sectional study.Setting2 large HIV primary care clinics in western Kenya.Participants1054 HIV-infected women in western Kenya undergoing cervical cancer screening as part of routine HIV care from October 2010 to November 2012.InterventionsParticipants underwent p16(INK4a) specimen collection and colposcopy. Lesions with unsatisfactory colposcopy or suspicious for cervical intraepithelial neoplasia 2+ (CIN2+; including CIN2/3 or invasive cervical cancer) were biopsied. Following biopsy, disease status was determined by histopathological diagnosis.Primary and secondary outcome measuresWe measured the sensitivity, specificity and predictive values of p16(INK4a) ELISA for CIN2+ detection among HIV-infected women and compared them to the test characteristics of current screening methods used in general as well as HIV-infected populations.ResultsAverage p16(INK4a) concentration in cervical samples was 37.4 U/mL. After colposcopically directed biopsy, 127 (12%) women were determined to have CIN2+. Receiver operating characteristic analysis showed an area under the curve of 0.664 for p16(INK4a) to detect biopsy-proven CIN2+. At a p16(INK4a) cut-off level of 9 U/mL, sensitivity, specificity, positive and negative predictive values were 89.0%, 22.9%, 13.6% and 93.8%, respectively. The overall p16(INK4a) positivity at a cut-off level of 9 U/mL was 828 (78.6%) women. There were 325 (30.8%) cases of correct p16(INK4a) prediction to detect or rule out CIN2+, and 729 (69.2%) cases of incorrect p16(INK4a) prediction.Conclusionsp16(INK4a) ELISA did not perform well as a screening test for CIN2+ detection among HIV-infected women due to low specificity. Our study contributes to the ongoing search for a more specific alternative to HPV testing for CIN2+ detection

    A turn propensity scale for transmembrane helices.

    Using a model protein with a 40 residue hydrophobic transmembrane segment, we have measured the ability of all the 20 naturally occurring amino acids to form a tight turn when placed in the middle of the hydrophobic segment. Turn propensities in a transmembrane helix are found to be markedly different from those of globular proteins, and in most cases correlate closely with the hydrophobicity of the residue. The turn propensity scale may be used to improve current methods for membrane protein topology prediction

    Turns in transmembrane helices – determination of the minimal length of a “helical hairpin” and derivation of a fine-grained turn propensity scale

    We have recently reported a first experimental turn propensity scale for transmembrane helices. This scale was derived from measurements of how efficiently a given residue placed in the middle of a 40 residue poly(Leu) stretch induces the formation of a "helical hairpin" with two rather than one transmembrane segment. We have now extended these studies, and have determined the minimum length of a poly(Leu) stretch compatible with the formation of a helical hairpin. We have also derived a more fine-grained turn propensity scale by (i) introducing each of the 20 amino acid residues into the middle of the shortest poly(Leu) stretch compatible with helical hairpin formation, and (ii) introducing pairs of residues in the middle of the 40 residue poly(Leu) stretch. The new turn propensities are consistent with the amino acid frequencies found in short hairpin loops in membrane proteins of known 3D structure

    Masteroppgave ledelse ORG917 - Universitetet i Agder 2017Masteroppgaven har utgangspunkt i at politiet er samfunnets maktorgan og skal løse alt fra mindre hendelser til større kriser blant befolkningen. Tillit blant befolkningen er viktig for at denne makten kan forvaltes på en god måte. Pålitelighet blant omgivelsene og internt er et nøkkelord. Målet med oppgaven var å kartlegge grad av HRO i Agder Politidistrikt. Problemstillingen i oppgaven er: ”Kan Agder Politidistrikt sies å være en High Reliability Organization (HRO)?” Jeg presenterer ulike teorier omkring kriser og katastrofer. Jeg har også plukket ut andre relevante teoretiske vinklinger, som det er naturlig å se i sammenheng med HRO-teorien. En av disse teoriene dreier seg om hvordan mennesker responderer. Mennesker kan handle enten intuitivt eller analytisk. Dette er av betydning for hvordan oppdrag blir løst. For å svare på problemstillingen har jeg gjennomført en spørreundersøkelse utviklet av Weick og Sutcliffe. (Weick og Sutcliffe, 2007). Undersøkelsen kartlegger blant annet grad av pålitelighet og evnen til å håndtere uforutsette hendelser. Summen av alle kategoriene er med på å avgjøre om Agder Politidistrikt kan sies å være en HRO. En lignende undersøkelse har ikke blitt gjennomført i politiet tidligere. I tillegg til å måle graden av HRO, er denne oppgaven en test på om HRO-teorien kan benyttes i politiet. Funn i spørreundersøkelsen viser at Agder Politidistrikt scorer høyt enkelte steder, men at det også er et stort utviklings-potensiale i organisasjonen. Jeg diskuterer årsakene til dette og tar for meg de punktene jeg mener er de viktigste. Avslutningsvis kommer jeg med noen anbefalinger for hva organisasjonen bør gjøre for å øke graden av HRO, og hvordan dette kan gjennomføres. Jeg kommer også med forslag til hvordan HRO-undersøkelsen kan utvikles videre
