    Counter Machines and Distributed Automata: A Story about Exchanging Space and Time

    We prove the equivalence of two classes of counter machines and one class of distributed automata. Our counter machines operate on finite words, which they read from left to right while incrementing or decrementing a fixed number of counters. The two classes differ in the extra features they offer: one allows to copy counter values, whereas the other allows to compute copyless sums of counters. Our distributed automata, on the other hand, operate on directed path graphs that represent words. All nodes of a path synchronously execute the same finite-state machine, whose state diagram must be acyclic except for self-loops, and each node receives as input the state of its direct predecessor.     Enzymfreisetzung und Aktivierung der Kallikrein-Kinin-Systeme bei experimenteller Pankreatitis

    Das klinische Bild der akuten Pankreatitis wird entscheidend durch die sekundĂ€re SchĂ€digung von Herz-Kreislauf-System, Lunge und Niere bestimmt. Ziel der vorliegenden Untersuchung war es, durch Messungen in venösem Pankreasblut, Pankreaslymphe und Peritonealexsudat die Kompartimente zu bestimmen, ĂŒber die die systemischen SchĂ€digungen vermittelt werden. An anĂ€sthesierten Schweinen wurden die systemischen, hĂ€modynamischen Parameter durch gesteuerte Volumentherapie konstant gehalten. Die Schweine wurden randomisiert der Kontrollgruppe (n = 9) oder einer der Pankreatitisgruppen zugeteilt (jeweils n = 10). Die Pankreatitis wurde durch Infusion von freier FettsĂ€ure in die Pankreasarterien (FFS) oder durch Infusion einer 5%igen Natrium-Taurocholat-Lösung retrograd in den Pankreasgang (NaT) ausgelöst. Nach Isolation des Pankreas wurde venöses Pankreasblut, Pankreaslymphe und Peritonealexsudat gewonnen und die AktivitĂ€t von Lipase, Phospholipase A und Plasmaprokallikrein sowie die Konzentration von Organkallikrein und Kininogen bestimmt. In beiden Pankreatitismodellen fand sich ein Anstieg der EnzymaktivitĂ€ten. Die höchsten AktivitĂ€ten fanden sich im Peritonealexsudat (Phospholipase A nach 40 min: Kontrolle 10,0 U/1, NaT 72,2 U/1). In beiden Pankreatitismodellen fanden sich außerdem Hinweise fĂŒr eine Aktivierung des Organkallikrein-Kinin-Systems durch den Anstieg der Organkallikreinkonzentration und den Abfall der Gesamtkininogenkonzentration. Die stĂ€rksten VerĂ€nderungen fanden sich wieder im Peritonealexsudat (Organkallikrein nach 40 min: Kontrolle 14,7 ng/ml, NaT 452 ng/ml).The clinical course of acute pancreatitis is strongly influenced by secondary cardiac, pulmonary and renal damage. The aim of the present study was to gather information about the compartment promoting the systemic damage. Therefore the activity of lipase, phospholipase A and plasmaprokallikrein and the concentration of tissue kallikrein and kininogen were measured in portal venous blood, pancreatic lymph and peritoneal exudate. Anaesthetized pigs were subjected to fluid resuscitation to keep systemic haemodynamic parameters constant. The pancreas was isolated in situ. The pigs were randomly assigned to a control group (n = 9) or one of the two pancreatitis groups (n = 10 each). Pancreatitis was induced by i.a. infusion of free fatty acid (FFS) or retrograde infusion of 5 % sodium taurocholate intraductally (NaT). In both pancreatitis groups the activity of lipase and phospholipase A increased. The most pronounced changes were seen in the peritoneal exsudate (phospholipase A activity 40 min after induction: control 10.0 U/1, NaT 72.2 U/1). In both pancreatitis groups there was evidence for activation of the tissue kallikreinkinin system in the form of an increase in the kallikrein concentration and a decrease in the kininogen concentration. Again the changes were most pronounced in the peritoneal exsudate (tissue kallikrein 40 min after induction: control 14.7 ng/ml, NaT 452 ng/ml)

    Bounded Languages Meet Cellular Automata with Sparse Communication

    Cellular automata are one-dimensional arrays of interconnected interacting finite automata. We investigate one of the weakest classes, the real-time one-way cellular automata, and impose an additional restriction on their inter-cell communication by bounding the number of allowed uses of the links between cells. Moreover, we consider the devices as acceptors for bounded languages in order to explore the borderline at which non-trivial decidability problems of cellular automata classes become decidable. It is shown that even devices with drastically reduced communication, that is, each two neighboring cells may communicate only constantly often, accept bounded languages that are not semilinear. If the number of communications is at least logarithmic in the length of the input, several problems are undecidable. The same result is obtained for classes where the total number of communications during a computation is linearly bounded

    Experimental autoimmune encephalomyelitis (EAE) is - in certain aspects - regarded as an animal model of the human CNS autoimmune disease multiple sclerosis (MS). While in EAE CNS-autoantigen-specific immunity is induced in a defined way, the initial processes leading to CNS autoimmunity in humans are so far unknown. Despite essential restrictions, which exist regarding the interpretation of EAE data towards MS, EAE might be a useful model to study certain basic aspects of CNS autoimmunity. Studies in MS have demonstrated that established autoimmune pathology can be critically influenced by environmental factors, in particular viral and bacterial infections. To investigate this interaction, EAE as an instrument to study CNS autoimmunity under defined conditions appears to be a suitable experimental tool. For this reason, we here investigated the influence of the Toll-like-receptor (TLR) ligand CpG oligonucleotide (CpG) on already established CNS autoimmunity in murine proteolipid protein (PLP)-induced EAE in SJL mice. CpG were found to co-stimulate PLPp-specific IFN-Îł production in the peripheral immune system and in the CNS. However, CpG induced Interleukin (IL)-17 production in the inflamed CNS both alone and in combination with additional PLPp stimulation. These findings might indicate a mechanism by which systemic infections and the microbial stimuli associated with them may influence already existing CNS autoimmune pathology

    The Actin Targeting Compound Chondramide Inhibits Breast Cancer Metastasis via Reduction of Cellular Contractility

    Background: A major player in the process of metastasis is the actin cytoskeleton as it forms key structures in both invasion mechanisms, mesenchymal and amoeboid migration. We tested the actin binding compound Chondramide as potential anti-metastatic agent. Methods: In vivo, the effect of Chondramide on metastasis was tested employing a 4T1-Luc BALB/c mouse model. In vitro, Chondramide was tested using the highly invasive cancer cell line MDA-MB-231 in Boyden-chamber assays, fluorescent stainings, Western blot and Pull down assays. Finally, the contractility of MDA-MB-231 cells was monitored in 3D environment and analyzed via PIV analysis. Results: In vivo, Chondramide treatment inhibits metastasis to the lung and the migration and invasion of MDA-MB-231 cells is reduced by Chondramide in vitro. On the signaling level, RhoA activity is decreased by Chondramide accompanied by reduced MLC-2 and the stretch induced guanine nucleotide exchange factor Vav2 activation. At same conditions, EGF-receptor autophosphorylation, Akt and Erk as well as Rac1 are not affected. Finally, Chondramide treatment disrupted the actin cytoskeleton and decreased the ability of cells for contraction. Conclusions: Chondramide inhibits cellular contractility and thus represents a potential inhibitor of tumor cell invasion

    Exact Solution of an Evolutionary Model without Ageing

    We introduce an age-structured asexual population model containing all the relevant features of evolutionary ageing theories. Beneficial as well as deleterious mutations, heredity and arbitrary fecundity are present and managed by natural selection. An exact solution without ageing is found. We show that fertility is associated with generalized forms of the Fibonacci sequence, while mutations and natural selection are merged into an integral equation which is solved by Fourier series. Average survival probabilities and Malthusian growth exponents are calculated indicating that the system may exhibit mutational meltdown. The relevance of the model in the context of fissile reproduction groups as many protozoa and coelenterates is discussed.Comment: LaTeX file, 15 pages, 2 ps figures, to appear in Phys. Rev.
