    Similarity and variability of blocked weather-regime dynamics in the Atlantic-European region

    Weather regimes govern an important part of the sub-seasonal variability of the mid-latitude circulation. Due to their role in weather extremes and atmospheric predictability, regimes that feature a blocking anticyclone are of particular interest. This study investigates the dynamics of these ''blocked'' regimes in the North Atlantic-European region from a year-round perspective. For a comprehensive diagnostic, we combine wave activity concepts and a piecewise potential-vorticity (PV) tendency framework. The latter essentially quantifies the well-established PV perspective of mid-latitude dynamics. All blocked regimes during the 1979&ndash;2021 period of ERA5 reanalysis are considered. Wave activity characteristics exhibit distinct differences between blocked regimes. After regime onset, one regime (Greenland Blocking) is associated with a suppression of wave activity flux, whereas two other regimes (Atlantic Ridge and European Blocking) are associated with a northward deflection of the flux without a clear net change. During onset, the envelope of Rossby wave activity retracts upstream for Greenland Blocking, whereas the envelope extends downstream for Atlantic Ridge and European Blocking. The fourth regime (Scandinavian Blocking) exhibits intermediate wave activity characteristics. From the perspective of piecewise PV tendencies projected onto the respective regime pattern, the dynamics that govern regime onset exhibit a large degree of similarity: Linear Rossby wave dynamics and nonlinear eddy PV fluxes dominate and are of approximately equal relative importance, whereas baroclinic coupling and divergent amplification make minor contributions. Most strikingly, all blocked regimes exhibit very similar (intra-regime) variability: a retrograde and an upstream pathway to regime onset. The retrograde pathway is dominated by nonlinear PV eddy fluxes, whereas the upstream pathway is dominated by linear Rossby wave dynamics. Importantly, there is a large degree of cancellation between the two pathways for some of the mechanisms before regime onset. The physical meaning of a regime-mean perspective before onset can thus be severely limited. Implications of our results for understanding predictability of blocked regimes are discussed. We further discuss the limitations of projected tendencies in capturing the importance of moist processes, which tend to occur at the fringes or outside of the regime pattern. Finally, we stress that this study investigate the variability of the governing dynamics without prior empirical stratification of data by season or by type of regime transition. We demonstrate, however, that our dynamics-centered approach does not map predominantly on variability that is associated with these factors. The main modes of dynamical variability revealed herein, and the large similarity of the blocked regimes in exhibiting this variability are thus significant results.</p

    Endothelial-Smooth Muscle Cell Interactions in a Shear-Exposed Intimal Hyperplasia on-a-Dish Model to Evaluate Therapeutic Strategies

    Intimal hyperplasia (IH) represents a major challenge following cardiovascular interventions. While mechanisms are poorly understood, the inefficient preventive methods incentivize the search for novel therapies. A vessel-on-a-dish platform is presented, consisting of direct-contact cocultures with human primary endothelial cells (ECs) and smooth muscle cells (SMCs) exposed to both laminar pulsatile and disturbed flow on an orbital shaker. With contractile SMCs sitting below a confluent EC layer, a model that successfully replicates the architecture of a quiescent vessel wall is created. In the novel IH model, ECs are seeded on synthetic SMCs at low density, mimicking reendothelization after vascular injury. Over 3 days of coculture, ECs transition from a network conformation to confluent 2D islands, as promoted by pulsatile flow, resulting in a "defected" EC monolayer. In defected regions, SMCs incorporated plasma fibronectin into fibers, increased proliferation, and formed multilayers, similarly to IH in vivo. These phenomena are inhibited under confluent EC layers, supporting therapeutic approaches that focus on endothelial regeneration rather than inhibiting proliferation, as illustrated in a proof-of-concept experiment with Paclitaxel. Thus, this in vitro system offers a new tool to study EC-SMC communication in IH pathophysiology, while providing an easy-to-use translational disease model platform for low-cost and high-content therapeutic development.     Loop Deletions Indicate Regions Important for FhuA Transport and Receptor Functions in Escherichia coli

    Precise deletions of cell surface-exposed loops of FhuA resulted in mutants of Escherichia coli with distinct phenotypes. Deletion of loop 3 or 11 inactivated ferrichrome transport activity. Deletion of loop 8 inactivated receptor activity for colicin M and the phages T1, T5, and φ80. The loop 7 deletion mutant was colicin M resistant but fully phage sensitive. The loop 4 deletion mutant was resistant to the TonB-dependent phages T1 and φ80 but fully sensitive to the TonB-independent phage T5. The phenotypes of the deletion mutants revealed important sites for the multiple FhuA transport and receptor activities. The ligand binding sites are nonidentical and are distributed among the entire exposed surface. Presumably, FhuA evolved as a ferrichrome transporter and was subsequently used as a receptor by the phages and colicin M, which selected the same as well as distinct loops as receptor sites

    Multi-purpose deep learning framework for MPI based on contrastive learning

    Deep learning can be used in many tasks for MPI, prominently to reduce the calibration time of system matrix (SM)-based reconstruction by recovering undersampled SMs or directly reconstructing measurements without an SM. The success of supervised machine learning methods depends on the used training data, which should have high quality, match the distribution of the desired test cases and be cleanly labeled. For MPI, such data rarely exists. To find robust features in complex input data, the unsupervised method contrastive learning can be used. In this work, we show its applicability to MPI voltage signals and that the learned features improve the performance of tasks like SM recovery and direct reconstruction of real MPI data in 2D. SM recovery is performed by predicting voltage signals of samples placed in the MPI FOV, which could also provide an alternative to classic simulation frameworks that cannot match real MPI measurements

    Deep learning MPI super-resolution by implicit representation of the system matrix

    Image reconstruction in MPI is often performed with the system matrix (SM) approach, where the signal of a reference particle sample is measured on a predefined grid in the scanner. This calibration measurement is not only very time-consuming but also places an upper limit to the spatial resolution of the reconstructed image, given by the spacing between two adjacent SM grid points. Recently, implicit neural representations have shown great results in computer vision. They allow for oversampling to gain a higher frequency explicit representation of an object without fixing a certain upsampling scale. We show that this can be used to mostly restore an SM with up to 16x subsampling in 2D and to generate SMs of arbitrary size as an additional tool for image quality improvement. However, we also found that classic spline interpolation is a reasonable tool for this task as well

    In Vivo Reconstitution of the FhuA Transport Protein of Escherichia coli K-12

    The FhuA protein in the outer membrane of Escherichia coli actively transports ferrichrome and the antibiotics albomycin and rifamycin CGP 4832 and serves as a receptor for the phages T1, T5, and φ80 and for colicin M and microcin J25. The crystal structure reveals a ÎČ-barrel with a globular domain, the cork, which closes the channel formed by the barrel. Genetic deletion of the cork resulted in a ÎČ-barrel that displays no FhuA activity. A functional FhuA was obtained by cosynthesis of separately encoded cork and the ÎČ-barrel domain, each endowed with a signal sequence, which showed that complementation occurs after secretion of the fragments across the cytoplasmic membrane. Inactive complete mutant FhuA and an FhuA fragment containing 357 N-proximal amino acid residues complemented the separately synthesized wild-type ÎČ-barrel to form an active FhuA. Previous claims that the ÎČ-barrel is functional as transporter and receptor resulted from complementation by inactive complete FhuA and the 357-residue fragment. No complementation was observed between the wild-type cork and complete but inactive FhuA carrying cork mutations that excluded the exchange of cork domains. The data indicate that active FhuA is reconstituted extracytoplasmically by insertion of separately synthesized cork or cork from complete FhuA into the ÎČ-barrel, and they suggest that in wild-type FhuA the ÎČ-barrel is formed prior to the insertion of the cork

    Sample preparations for reproducible measurements in MPS and MPI

    MPI is a promising new technology for highly sensitive medical imaging. To acquire reproducible measurements,stable scanner and sample preparations are needed for both system matrix (SM) acquisition and measurement.Therefore, it is important to analyze and subsequently minimize the influence of the scanner hardware, as wellas of the preparation method. In this work, we studied the evolution of the MPI signal of particle samplesusing five sealing methods (tape, hot glue, silicone, silicone + tape, UV glue) in combination with two differenttracers (perimagÂź and SHP20). In a background examination, we obtained high variations in the signal of theempty scanner over time.
