    Observations on the Life Cycles of \u3ci\u3eHydropsyche bifida\u3c/i\u3e Banks and \u3ci\u3eHydropsyche morosa\u3c/i\u3e Hagen (Trichoptera: Hydropsychidae)

    Two different laboratory streams were.used to examine the life cycles of Hydropsyche bifida and H. morosa (Trichoptera:Hydropsychidae). Effectiveness of the artificial streams was determined. Hydropsyche bifida and H. morosa were shown to be useful bioassay organisms. Variations of the species-specific larval head patterns were drawn for both H. oifida and H. morosa. Field studies were used to determine nicheΒ·separations for H. bifida and H. morosa in riffle areas of the Sauk River, St. Cloud, Minnesota. Physical-spatial factors such as retreat material, substrate preference, area of riffle and location on substrate were used to evaluate thi\u27s separation. Eggs found in the river were indistin-Β· guishable from laboratory eggs obtained. The river eggs developed and photographs were made of the first instar, tentatively identified as Hydropsyche sp

Without uniforms, flags, and formations, the task of identifying and targeting these combatants represented an unprecedented operational challenge for which Cold War era doctrinal methods were largely unsuited. This monograph examines the doctrinal, technical, and bureaucratic innovations that evolved in response to these new operational challenges.     Increasing Estrus Expression in Lactating Dairy Cows

    This report summarizes the use of various hormones in an attempt to induce greater estrus expression of lactating dairy cows. Average detection of estrus (\u3c 50%) in most U.S. dairy herds has been identified as a problem limiting reproductive efficiency. Detection of estrus plays an important role in reproductive management in U.S. dairy herds despite the adoption of fixed-time artificial insemination programs. When estrus was detected by an activity monitoring system or a rump-mounted device, supplementing cows with progesterone before induction of luteolysis resulted in greater intensity of estrus when compared with controls. In addition, administering estradiol cypionate at 24 h after induced luteolysis resulted in greater frequency of estrus expression compared with cows treated with testosterone propionate or controls when assessed by an activity monitor. Activity monitoring systems achieved heat-detection rates of approximately 70% (80% with estradiol) and are likely cost effective for herds achieving less than that level of heat detection

    Vertical Structure and Color of Jovian Latitudinal Cloud Bands during the Juno Era

    The identity of the coloring agent(s) in Jupiter's atmosphere and the exact structure of Jupiter's uppermost cloud deck are yet to be conclusively understood. The Cr\`{e}me Br\^ul\'ee model of Jupiter's tropospheric clouds, originally proposed by Baines et al. (2014) and expanded upon by Sromovsky et al. (2017) and Baines et al. (2019), presumes that the chromophore measured by Carlson et al. (2016) is the singular coloring agent in Jupiter's troposphere. In this work, we test the validity of the Cr\`{e}me Br\^ul\'ee model of Jupiter's uppermost cloud deck using spectra measured during the Juno spacecraft's 5th^{\mathrm{th}} perijove pass in March 2017. These data were obtained as part of an international ground-based observing campaign in support of the Juno mission using the NMSU Acousto-optic Imaging Camera (NAIC) at the 3.5-m telescope at Apache Point Observatory in Sunspot, NM. We find that the Cr\`{e}me Br\^ul\'ee model cloud layering scheme can reproduce Jupiter's visible spectrum both with the Carlson et al. (2016) chromophore and with modifications to its imaginary index of refraction spectrum. While the Cr\`{e}me Br\^ul\'ee model provides reasonable results for regions of Jupiter's cloud bands such as the North Equatorial Belt and Equatorial Zone, we find that it is not a safe assumption for unique weather events, such as the 2016-2017 Southern Equatorial Belt outbreak that was captured by our measurements.Comment: 38 pages, 21 figures; Accepted for publication in AAS Planetary Science Journa

    The near real-time synthesis of a non-uniformly correlated partially coherent source using a low-actuator-count deformable mirror is demonstrated. The statistical optics theory underpinning the synthesis method is reviewed. The experimental results of a non-uniformly correlated source are presented and compared to theoretical predictions. A discussion on how deformable mirror characteristics such as actuator count and pitch affect source generation is also included

    Automated Analysis of Cryptococcal Macrophage Parasitism Using GFP-Tagged Cryptococci

    The human fungal pathogens Cryptococcus neoformans and C. gattii cause life-threatening infections of the central nervous system. One of the major characteristics of cryptococcal disease is the ability of the pathogen to parasitise upon phagocytic immune effector cells, a phenomenon that correlates strongly with virulence in rodent models of infection. Despite the importance of phagocyte/Cryptococcus interactions to disease progression, current methods for assaying virulence in the acrophage system are both time consuming and low throughput. Here, we introduce the first stable and fully characterised GFP–expressing derivatives of two widely used cryptococcal strains: C. neoformans serotype A type strain H99 and C. gattii serotype B type strain R265. Both strains show unaltered responses to environmental and host stress conditions and no deficiency in virulence in the macrophage model system. In addition, we report the development of a method to effectively and rapidly investigate macrophage parasitism by flow cytometry, a technique that preserves the accuracy of current approaches but offers a four-fold improvement in speed

    A Diverse Population of Cryptococcus gattii Molecular Type VGIII in Southern Californian HIV/AIDS Patients

    Cryptococcus gattii infections in southern California have been reported in patients with HIV/AIDS. In this study, we examined the molecular epidemiology, population structure, and virulence attributes of isolates collected from HIV/AIDS patients in Los Angeles County, California. We show that these isolates consist almost exclusively of VGIII molecular type, in contrast to the VGII molecular type isolates causing the North American Pacific Northwest outbreak. The global VGIII population structure can be divided into two molecular groups, VGIIIa and VGIIIb. Isolates from the Californian patients are virulent in murine and macrophage models of infection, with VGIIIa significantly more virulent than VGIIIb. Several VGIII isolates are highly fertile and produce abundant sexual spores that may serve as infectious propagules. The a and Ξ± VGIII MAT locus alleles are largely syntenic with limited rearrangements compared to the known VGI (a/Ξ±) and VGII (Ξ±) MAT loci, but each has unique characteristics including a distinct deletion flanking the 5β€² VGIII MATa alleles and the Ξ± allele is more heterogeneous than the a allele. Our studies indicate that C. gattii VGIII is endemic in southern California, with other isolates originating from the neighboring regions of Mexico, and in rarer cases from Oregon and Washington state. Given that >1,000,000 cases of cryptococcal infection and >620,000 attributable mortalities occur annually in the context of the global AIDS pandemic, our findings suggest a significant burden of C. gattii may be unrecognized, with potential prognostic and therapeutic implications. These results signify the need to classify pathogenic Cryptococcus cases and highlight possible host differences among the C. gattii molecular types influencing infection of immunocompetent (VGI/VGII) vs. immunocompromised (VGIII/VGIV) hosts
