Size-associated Variation and Factors Affecting the Morphology of Brown Bodies in Glycera tridactyla (Polychaeta:Glyceridae)

Size-associated variations in brown body morphology (size and shape) were studied from the individuals of Glycera tridactyla. Variables related to size, length (BL) and width (BW) of the brown bodies were measured. On the basis of these measurements, other variables such as the surface area (SA), volume (V), surface area/volume ratio (SA/V) and elongation degree (ED) were calculated. Brown body shape was quantified by the elongation degree. The immature brown bodies were significantly smaller than the mature bodies, and therefore the smaller bodies had a higher SA/V. Results obtained from the regression analysis showed that there was a significant relationship between all the variables with the exception of SA/V:BW for the immature bodies and ED: BL for both groups of the bodies. The body size (proboscis length) of the worm and the sampling time (months) were the affecting factors on size and shape of the brown body. Brown bodies tend to be elongated as the proboscis length increased. The variation in size and/or shape of the brown bodies could refer to an adaptation to the movement along the coelomic fluid

Background: The aim of this study was to evaluate the effects of β-glucan on the expression of inflammatory mediators and metabolomic profile of oral cells [keratinocytes (OBA-9) and fibroblasts (HGF-1) in a dual-chamber model] infected by Aggregatibacter actinomycetemcomitans. The periodontopathogen was applied and allowed to cross the top layer of cells (OBA-9) to reach the bottom layer of cells (HGF-1) and induce the synthesis of immune factors and cytokines in the host cells. β-glucan (10 μg/mL or 20 μg/mL) were added, and the transcriptional factors and metabolites produced were quantified in the remaining cell layers and supernatant. Results: The relative expression of interleukin (IL)-1-α and IL-18 genes in HGF-1 decreased with 10 μg/mL or 20 μg/mL of β-glucan, where as the expression of PTGS-2 decreased only with 10 μg/mL. The expression of IL-1-α increased with 20 μg/mL and that of IL-18 increased with 10 μg/mL in OBA-9; the expression of BCL 2, EP 300, and PTGS-2 decreased with the higher dose of β-glucan. The production of the metabolite 4-aminobutyric acid presented lower concentrations under 20 μg/mL, whereas the concentrations of 2-deoxytetronic acid NIST and oxalic acid decreased at both concentrations used. Acetophenone, benzoic acid, and pinitol presented reduced concentrations only when treated with 10 μg/mL of β-glucan. Conclusions: Treatment with β-glucans positively modulated the immune response and production of metabolites

The effects of β-glucan on human immune and cancer cells

Non-prescriptional use of medicinal herbs among cancer patients is common around the world. The alleged anti-cancer effects of most herbal extracts are mainly based on studies derived from in vitro or in vivo animal experiments. The current information suggests that these herbal extracts exert their biological effect either through cytotoxic or immunomodulatory mechanisms. One of the active compounds responsible for the immune effects of herbal products is in the form of complex polysaccharides known as β-glucans. β-glucans are ubiquitously found in both bacterial or fungal cell walls and have been implicated in the initiation of anti-microbial immune response. Based on in vitro studies, β-glucans act on several immune receptors including Dectin-1, complement receptor (CR3) and TLR-2/6 and trigger a group of immune cells including macrophages, neutrophils, monocytes, natural killer cells and dendritic cells. As a consequence, both innate and adaptive response can be modulated by β-glucans and they can also enhance opsonic and non-opsonic phagocytosis. In animal studies, after oral administration, the specific backbone 1→3 linear β-glycosidic chain of β-glucans cannot be digested. Most β-glucans enter the proximal small intestine and some are captured by the macrophages. They are internalized and fragmented within the cells, then transported by the macrophages to the marrow and endothelial reticular system. The small β-glucans fragments are eventually released by the macrophages and taken up by other immune cells leading to various immune responses. However, β-glucans of different sizes and branching patterns may have significantly variable immune potency. Careful selection of appropriate β-glucans is essential if we wish to investigate the effects of β-glucans clinically. So far, no good quality clinical trial data is available on assessing the effectiveness of purified β-glucans among cancer patients. Future effort should direct at performing well-designed clinical trials to verify the actual clinical efficacy of β-glucans or β-glucans containing compounds

Abstract: -Glucans belong to a group of biologically active natural compounds called biological response modifiers. These substances represent highly conserved structural components of cell walls in yeast, fungi, grain and seaweed. The effects of -glucan on immune reactions are well established however direct comparisons of the biological activities of several individual glucans are extremely rare. As this paper will show, we tested sixteen different glucans and evaluated the possibility whether individual glucans will be similarly active against each of the tested biological properties or if each glucan will affect different reactions. No direct connection between source and immunological activities was found. Based on our results, we can conclude that highly purified and highly active glucans have pleotropic effects, whereas poorly isolated glucans have only average (if any) biological effect

Immune cells and structures of Annelida represent a complex subject that was studied for over 120 years. An overwhelming mountain of data have been accumulated during these studies, most of them being the subject of many excellent reviews and books. In the this paper we focused on a brief survay of old studies and some reflection on the findings during recent years
