Nanoparticle transport in saturated porous medium using magnetic resonance imaging
Transport study of nanoparticle (NP) through matrix flow dominated aquifer sand and soils have significant influence in natural systems. To quantify the transport behaviour, magnetic resonance imaging (MRI) was used to image the iron oxide based nanoparticle, Molday ION (carboxyl terminated) through saturated sandstone rock core. T2-weighted images were acquired and the changes in image intensity were calibrated to get a quantitative concentration profiles at various time intervals. These profiles were evaluated through CXTFIT transport model to estimate the transport parameters. These parameters are estimated at various points along the length of the column while classical breakthrough curve analysis cannot provide these details. NP–surface interactions were investigated using DLVO (Derjaguin–Landau–Verwey–Overbeek) theory. The dispersion coefficients (2.55–1.21 × 10−7 m2/s) were found to be decrease with distance, deposition rate constant k (6.70–9.13 × 10−4 (1/s)) and fast deposition rate constant kfast (4.32–8.79 × 10−2 (1/s)) were found to be increase with distance. These parameter variations over length will have a scaling up impact in developing transport models for environmental remediation and risk assessment schemes
An Integrated Study of Pervious Concrete Mixture Design for Wearing Course Applications
This report presents the results of the largest and most comprehensive study to date on portland cement pervious concrete (PCPC). It is designed to be widely accessible and easily applied by designers, producers, contractors, and owners. The project was designed to begin with pervious concrete best practices and then to address the unanswered questions in a systematic fashion to allow a successful overlay project. Consequently, the first portion of the integrated project involved a combination of fundamental material property investigations, test method development, and addressing constructability issues before actual construction could take place. The second portion of the project involved actual construction and long-term testing before reporting successes, failures, and lessons learned. The results of the studies conducted show that a pervious concrete overlay can be designed, constructed, operated, and maintained. A pervious concrete overlay has several inherent advantages, including reduced splash and spray and reduced hydroplaning potential, as well as being a very quiet pavement. The good performance of this overlay in a particularly harsh freeze-thaw climate, Minnesota, shows pervious concrete is durable and can be successfully used in freeze-thaw climates with truck traffic and heavy snow plowing
Metagenomic sequencing unravels gene fragments with phylogenetic signatures of O2-tolerant NiFe membrane-bound hydrogenases in lacustrine sediment
Many promising hydrogen technologies utilising hydrogenase enzymes have been slowed by the fact that most hydrogenases are extremely sensitive to O2. Within the group 1 membrane-bound NiFe hydrogenase, naturally occurring tolerant enzymes do exist, and O2 tolerance has been largely attributed to changes in iron–sulphur clusters coordinated by different numbers of cysteine residues in the enzyme’s small subunit. Indeed, previous work has provided a robust phylogenetic signature of O2 tolerance [1], which when combined with new sequencing technologies makes bio prospecting in nature a far more viable endeavour. However, making sense of such a vast diversity is still challenging and could be simplified if known species with O2-tolerant enzymes were annotated with information on metabolism and natural environments. Here, we utilised a bioinformatics approach to compare O2-tolerant and sensitive membrane-bound NiFe hydrogenases from 177 bacterial species with fully sequenced genomes for differences in their taxonomy, O2 requirements, and natural environment. Following this, we interrogated a metagenome from lacustrine surface sediment for novel hydrogenases via high-throughput shotgun DNA sequencing using the Illumina™ MiSeq platform. We found 44 new NiFe group 1 membrane-bound hydrogenase sequence fragments, five of which segregated with the tolerant group on the phylogenetic tree of the enzyme’s small subunit, and four with the large subunit, indicating de novo O2-tolerant protein sequences that could help engineer more efficient hydrogenases

Bayes linear analysis of imprecision in computer models, with application to understanding galaxy formation.
Imprecision arises naturally in the context of computer models and their relation to reality. An imprecise treatment of general computer models is presented, illustrated with an analysis of a complex galaxy formation simulation known as Galform. The analysis involves several different types of uncertainty, one of which (the Model Discrepancy) comes directly from expert elicitation regarding the deficiencies of the model. The Model Discrepancy is therefore treated within an Imprecise framework to reflect more accurately the beliefs of the expert concerning the discrepancy between the model and reality. Due to the conceptual complexity and computationally intensive nature of such a Bayesian imprecise uncertainty analysis, Bayes Linear Methodology is employed which requires consideration of only expectations and variances of all uncertain quantities. Therefore incorporating an Imprecise treatment within a Bayes Linear analysis is shown to be relatively straightforward. The impact of an imprecise assessment on the input space of the model is determined through the use of an Implausibility measure

Gene expression profile of cervical tissue compared to exfoliated cells: Impact on biomarker discovery
BACKGROUND: Exfoliated cervical cells are used in cytology-based cancer screening and may also be a source for molecular biomarkers indicative of neoplastic changes in the underlying tissue. However, because of keratinization and terminal differentiation it is not clear that these cells have an mRNA profile representative of cervical tissue, and that the profile can distinguish the lesions targeted for early detection. RESULTS: We used whole genome microarrays (25,353 unique genes) to compare the transcription profiles from seven samples of normal exfoliated cells and one cervical tissue. We detected 10,158 genes in exfoliated cells, 14,544 in the tissue and 7320 genes in both samples. For both sample types the genes grouped into the same major gene ontology (GO) categories in the same order, with exfoliated cells, having on average 20% fewer genes in each category. We also compared microarray results of samples from women with cervical intraepithelial neoplasia grade 3 (CIN3, n = 15) to those from age and race matched women without significant abnormalities (CIN1, CIN0; n = 15). We used three microarray-adapted statistical packages to identify differential gene expression. The six genes identified in common were two to four fold upregulated in CIN3 samples. One of these genes, the ubiquitin-conjugating enzyme E2 variant 1, participates in the degradation of p53 through interaction with the oncogenic HPV E6 protein. CONCLUSION: The findings encourage further exploration of gene expression using exfoliated cells to identify and validate applicable biomarkers. We conclude that the gene expression profile of exfoliated cervical cells partially represents that of tissue and is complex enough to provide potential differentiation between disease and non-disease
