    Evaluation and bias correction of satellite rainfall data for drought monitoring in Indonesia

    The accuracy of three satellite rainfall products (TMPA 3B42RT, CMORPH and PERSIANN) was investigated through comparison with grid cell average ground station rainfall data in Indonesia, with a focus on their ability to detect patterns of low rainfall that may lead to drought conditions. Each of the three products underestimated rainfall in dry season months. The CMORPH and PERSIANN data differed most from ground station data and were also very different from the TMPA 3B42RT data. It proved possible to improve TMPA 3B42RT estimates by applying a single empirical bias correction equation that was uniform in space and time. For the six regions investigated, this reduced the root mean square error for estimates of dry season rainfall totals by a mean 9% (from 44 to 40 mm) and for annual totals by 14% (from 77 to 66 mm). The resulting errors represent 10% and 3% of mean dry season and annual rainfall, respectively. The accuracy of these bias corrected TMPA 3B42RT data is considered adequate for use in real-time drought monitoring in Indonesia. Compared to drought monitoring with only ground stations, this use of satellite-based rainfall estimates offers important advantages in terms of accuracy, spatial coverage, timeliness and cost efficiency

    Accelerated/reduced growth of tungsten fuzz by deposition of metals

    From the helium (He) plasma irradiations to tungsten performed in the Magnum-PSI device, the effects of deposition of metals on the helium-plasma induced fiberform nanostructures (fuzz) are discussed. It was found that fuzz was not formed at the center of the plasma cylinder if there were significant metallic impurities from the source. Deposition of metallic impurities (mainly molybdenum and copper) counteracted the growth of fuzz. In addition to the effects of metals from the source, we installed a sputtering source near the sample to replicate the deposition environment in fusion devices. The thickness of fuzzy layer was ∼7&nbsp;µm, which was about five times greater than that without deposition, at the He flux of 1.3×1026&nbsp;m−2, suggesting that the growth rate of fuzz layer was significantly accelerated due to the deposition of tungsten.</p

    B2.5-Eunomia simulations of Magnum-PSI detachment experiments: I. Quantitative comparisons with experimental measurements

    Detachment experiments have been carried out in the linear plasma device Magnum-PSI by increasing the gas pressure near the target. In order to have a proper detailed analysis of the mechanism behind momentum and power loss in detachment, a quantitative match is pursued between B2.5-Eunomia solutions and experimental data. B2.5 is a multi fluid plasma code and Eunomia is a Monte Carlo solver for neutral particles, and they are coupled together to provide steady-state solution of the plasma and neutral distribution in space. B2.5-Eunomia input parameters are adjusted to produce a close replication of the plasma beam measured in the experiments without any gas puffing in the target chamber. Using this replication as an initial condition, the neutral pressure near the plasma beam target is exclusively increased during simulation, matching the pressures measured in the experiments. Reasonable agreement is found between the electron temperature of the simulation results with experimental measurements using laser Thomson scattering near the target. The simulations also reveal the effect of increased gas pressure on the plasma current, effectively reducing the current penetration from the plasma source. B2.5-Eunomia is capable of reproducing detachment characteristics, namely the loss of plasma pressure along the magnetic field and the reduction of particle and heat flux to the target. The simulation results for plasma and neutrals will allow future studies of the exact contribution of individual plasma-neutral collisions to momentum and energy loss in detachment in Magnum-PSI.</p

    Chromosome looping at the human alpha-globin locus is mediated via the major upstream regulatory element (HS-40)

    Previous studies in the mouse have shown that high levels of alpha-globin gene expression in late erythropoiesis depend on long-range, physical interactions between remote upstream regulatory elements and the globin promoters. Using quantitative chromosome conformation capture (q3C), we have now analyzed all interactions between 4 such elements lying 10 to 50 kb upstream of the human alpha cluster and their interactions with the alpha-globin promoter. All of these elements interact with the alpha-globin gene in an erythroid-specific manner. These results were confirmed in a mouse model of human alpha globin expression in which the human cluster replaces the mouse cluster in situ (humanized mouse). We have also shown that expression and all of the long-range interactions depend largely on just one of these elements; removal of the previously characterized major regulatory element (called HS -40) results in loss of all the interactions and alpha-globin expression. Reinsertion of this element at an ectopic location restores both expression and the intralocus interactions. In contrast to other more complex systems involving multiple upstream elements and promoters, analysis of the human alpha-globin cluster during erythropoiesis provides a simple and tractable model to understand the mechanisms underlying long-range gene regulation

    High-resolution analysis of cis-acting regulatory networks at the &#945;-globin locus.

    We have combined the circular chromosome conformation capture protocol with high-throughput, genome-wide sequence analysis to characterize the cis-acting regulatory network at a single locus. In contrast to methods which identify large interacting regions (10–1000 kb), the 4C approach provides a comprehensive, high-resolution analysis of a specific locus with the aim of defining, in detail, the cis-regulatory elements controlling a single gene or gene cluster. Using the human α-globin locus as a model, we detected all known local and long-range interactions with this gene cluster. In addition, we identified two interactions with genes located 300 kb (NME4) and 625 kb (FAM173a) from the α-globin cluster

    Generation of bivalent chromatin domains during cell fate decisions

    <p>Abstract</p> <p>Background</p> <p>In self-renewing, pluripotent cells, bivalent chromatin modification is thought to silence (H3K27me3) lineage control genes while 'poising' (H3K4me3) them for subsequent activation during differentiation, implying an important role for epigenetic modification in directing cell fate decisions. However, rather than representing an equivalently balanced epigenetic mark, the patterns and levels of histone modifications at bivalent genes can vary widely and the criteria for identifying this chromatin signature are poorly defined.</p> <p>Results</p> <p>Here, we initially show how chromatin status alters during lineage commitment and differentiation at a single well characterised bivalent locus. In addition we have determined how chromatin modifications at this locus change with gene expression in both ensemble and single cell analyses. We also show, on a global scale, how mRNA expression may be reflected in the ratio of H3K4me3/H3K27me3.</p> <p>Conclusions</p> <p>While truly 'poised' bivalently modified genes may exist, the original hypothesis that all bivalent genes are epigenetically premarked for subsequent expression might be oversimplistic. In fact, from the data presented in the present work, it is equally possible that many genes that appear to be bivalent in pluripotent and multipotent cells may simply be stochastically expressed at low levels in the process of multilineage priming. Although both situations could be considered to be forms of 'poising', the underlying mechanisms and the associated implications are clearly different.</p
