    Field theoretic calculation of scalar turbulence

    The cascade rate of passive scalar and Bachelor's constant in scalar turbulence are calculated using the flux formula. This calculation is done to first order in perturbation series.     Lessons learnedThe safety profile in the patient groups who received FOLFIRI and simtuzumab did not differ from that in the FOLFIRI and placebo group.The addition of simtuzumab to chemotherapy with FOLFIRI does not improve clinical outcomes in patients with metastatic KRAS mutant colorectal carcinoma.BackgroundSimtuzumab, a humanized IgG4 monoclonal antibody to lysyl oxidase-like 2 (LOXL2), blocks desmoplastic reaction in colorectal carcinoma (CRC) cells in vitro.MethodsPatients with metastatic Kirsten rat sarcoma viral oncogene homolog (KRAS) mutant CRC were randomized to receive second-line 5-fluorouracil, leucovorin, and irinotecan (FOLFIRI) with either 200 or 700 mg simtuzumab or placebo every 2 weeks in cycles of 28 days. Progression-free survival (PFS), overall survival (OS), objective response rate (ORR), and safety were assessed.ResultsIn total, 249 patients were randomized and treated with FOLFIRI/simtuzumab 700 mg (n = 84), FOLFIRI/simtuzumab 200 mg (n = 85), and FOLFIRI/placebo (n = 80). After a median follow-up of 5.1, 3.8, and 5.5 months, respectively, median PFS for each of the respective treatment groups was 5.5 months (adjusted HR [95% CI], p value versus placebo; 1.32 [0.92, 1.89]; p = .10), 5.4 months (1.45 [1.01, 2.06]; p = .04), and 5.8 months. Median OS was 11.4 months (1.23 [0.80, 1.91]; p = .25), 10.5 months (1.50 [0.98, 2.30]; p = .06), and 16.3 months, respectively. ORR was 11.9%, 5.9%, and 10%, respectively. Simtuzumab was tolerable in metastatic KRAS mutant CRC patients.ConclusionThe addition of simtuzumab to FOLFIRI did not improve clinical outcomes in patients with metastatic KRAS mutant CRC.     Study of Distribution and Asymmetry of Solar Active Prominences During Solar Cycle 23

    In this paper we present the results of a study of the spatial distribution and asymmetry of solar active prominences (SAP) for the period 1996-2007 (solar cycle 23). For more meaningful statistical analysis we have analysed the distribution and asymmetry of SAP in two subdivisions viz. Group1 (ADF, APR, DSF, CRN, CAP) and Group2 (AFS, ASR, BSD, BSL, DSD, SPY, LPS). The north-south (N-S) latitudinal distribution shows that the SAP events are most prolific in the 21-30degree slice in the northern and southern hemispheres and east-west (E-W) longitudinal distribution study shows that the SAP events are most prolific (best visible) in the 81-90degree slice in the eastern and western hemispheres. It has been found that the SAP activity during this cycle is low compared to previous solar cycles. The present study indicates that during the rising phase of the cycle the number of SAP events were roughly equal on the north and south hemispheres. However, activity on the southern hemisphere has been dominant since 1999.     Ice in the Antarctic polar stratosphere

    On six occasions during the 1987 Airborne Antarctic Ozone Experiment, the Polar Stratospheric Cloud (PSC) ice crystals were replicated over the Palmer Peninsula at approximately 70 deg South. The sampling altitude was approximately 60 to 65 thousand feet, the temperature range was -83.5 to -72C and the atmosphere was subsaturated in all cases. The collected crystals were predominantly complete and hollow prismatic columns with maximum dimensions up to 217 microns. Evidence of scavenging of submicron particles was detected on several crystals. While the replicated crystal sizes were larger than anticipated, their relatively low concentration results in a total surface area less than one tenth that of the sampled aerosol particles. The presence of large crystals suggest that PSC ice crystals can play a very important role in stratospheric dehydration processes

    Simple Pendulum Revisited

    We describe a 8085 microprocessor interface developed to make reliable time period measurements. The time period of each oscillation of a simple pendulum was measured using this interface. The variation of the time period with increasing oscillation was studied for the simple harmonic motion (SHM) and for large angle initial displacements (non-SHM). The results underlines the importance of the precautions which the students are asked to take while performing the pendulum experiment.Comment: 17 pages with 10 figure

    Development of Protacs to Target Cancer-promoting Proteins for Ubiquitination and Degradation

    The proteome contains hundreds of proteins that in theory could be excellent therapeutic targets for the treatment of human diseases. However, many of these proteins are from functional classes that have never been validated as viable candidates for the development of small molecule inhibitors. Thus, to exploit fully the potential of the Human Genome Project to advance human medicine, there is a need to develop generic methods of inhibiting protein activity that do not rely on the target protein’s function. We previously demonstrated that a normally stable protein, methionine aminopeptidase-2 or MetAP-2, could be artificially targeted to an Skp1-Cullin-F-box (SCF) ubiquitin ligase complex for ubiquitination and degradation through a chimeric bridging molecule or Protac (proteolysis targeting chimeric molecule). This Protac consisted of an SCFß-TRCP-binding phosphopeptide derived from I{kappa}B{alpha} linked to ovalicin, which covalently binds MetAP-2. In this study, we employed this approach to target two different proteins, the estrogen (ER) and androgen (AR) receptors, which have been implicated in the progression of breast and prostate cancer, respectively. We show here that an estradiol-based Protac can enforce the ubiquitination and degradation of the {alpha} isoform of ER in vitro, and a dihydroxytestosterone-based Protac introduced into cells promotes the rapid disappearance of AR in a proteasome-dependent manner. Future improvements to this technology may yield a general approach to treat a number of human diseases, including cancer
