    Laser-induced periodic surface structures:Fingerprints of light localization

    The finite-difference time-domain (FDTD) method is used to study the inhomogeneous absorption of linearly polarized laser radiation below a rough surface. The results are first analyzed in the frequency domain and compared to the efficacy factor theory of Sipe and coworkers. Both approaches show that the absorbed energy shows a periodic nature, not only in the direction orthogonal to the laser polarization, but also in the direction parallel to it. It is shown that the periodicity is not always close to the laser wavelength for the perpendicular direction. In the parallel direction, the periodicity is about lambda/Re((n) over tilde), with (n) over tilde being the complex refractive index of the medium. The space-domain FDTD results show a periodicity in the inhomogeneous energy absorption similar to the periodicity of the low-and high-spatial-frequency laser-induced periodic surface structures depending on the material's excitation

    Molecular characterization and identification of proteins regulated by Hfq in Neisseria meningitidis

    Hfq is a highly conserved pleiotropically acting prokaryotic RNA-binding protein involved in the post-transcriptional regulation of many stress-responsive genes by small RNAs. In this study, we show that Hfq of the strictly human pathogen Neisseria meningitidis is involved in the regulation of expression of components involved in general metabolic pathways, iron metabolism and virulence. A meningococcal hfq deletion strain (H44/76Ī”hfq) is impaired in growth in nutrient-rich media and does not grow at all in nutrient-limiting medium. The growth defect was complemented by expression of hfq in trans. Using proteomics, the expression of 28 proteins was found to be significantly affected upon deletion of hfq. Of these, 20 proteins are involved in general metabolism, among them seven iron-responsive genes. Two proteins (PilE, TspA) are involved in adherence to human cells, a step crucial for the onset of disease. One of the differentially expressed proteins, GdhA, was identified as an essential virulence factor for establishment of sepsis in an animal model, studied earlier. These results show that in N. meningitidis Hfq is involved in the regulation of a variety of components contributing to the survival and establishment of meningococcal disease

    Nonsteroidal antiinflammatory drugs and the risk of Alzheimer's disease

    BACKGROUND: Previous studies have suggested that the use of nonsteroidal antiinflammatory drugs (NSAIDs) may help to prevent Alzheimer's disease. The results, however, are inconsistent. METHODS: We studied the association between the use of NSAIDs and Alzheimer's disease and vascular dementia in a prospective, population-based cohort study of 6989 subjects 55 years of age or older who were free of dementia at base line, in 1991. To detect new cases of dementia, follow-up screening was performed in 1993 and 1994 and again in 1997 through 1999. The risk of Alzheimer's disease was estimated in relation to the use of NSAIDs as documented in pharmacy records. We defined four mutually exclusive categories of use: nonuse, short-term use (1 month or less of cumulative use), intermediate-term use (more than 1 but less than 24 months of cumulative use), and long-term use (24 months or more of cumulative use). Adjustments were made by Cox regression analysis for age, sex, education, smoking status, and the use or nonuse of salicylates, histamine Hz-receptor antagonists, antihypertensive agents, and hypoglycemic agents. RESULTS: During an average follow-up period of 6.8 years, dementia developed in 394 subjects, of whom 293 had Alzheimer's disease, 56 vascular dementia, and 45 other types of dementia. The relative risk of Alzheimer's disease was 0.95 (95 percent confidence interval, 0.70 to 1.29) in subjects with short-term use of NSAIDs, 0.83 (95 percent confidence interval, 0.62 to 1.11) in those with intermediate-term use, and 0.20 (95 percent confidence interval, 0.05 to 0.83) in those with long-term use. The risk did not vary according to age. The use of NSAIDs was not associated with a reduction in the risk of vascular dementia. CONCLUSIONS: The long-term use of NSAIDs may protect against Alzheimer's disease but not against vascular dementia

    Increased incidence of cytogenetic abnormalities in chorionic villus samples from pregnancies established by in vitro fertilization and embryo transfer (IVF-ET)

    We studied 201 pregnancies that were established by in vitro fertilization and embryo transfer (IVFā€“ET) and compared the frequency of cytogenetic abnormalities with that found in a large control population matched for indication group (advanced maternal age) and time of sampling. A total of 252 IVFā€“ET fetuses were cytogenetically analysed by either chorionic villus sampling (CVS; n = 80) or amniocentesis (n = 172). Eleven chromosome abnormalities were found in the CVS group (13Ā·8 per cent); among them, a 45, X/46, X, dic(q11)/46, X, del(Y)(q11) mosaic that was found in an IVF pregnancy established by intracytoplasmic sperm injection (ICSI), four cases of trisomy 21, and three cases of trisomy 7 confined to the placenta. The results indicate a statistically significant threeā€to fiveā€fold increase in both confined placental abnormalities (P<0Ā·008) and true fetal chromosome anomalies (P<0Ā·04). In the amniocentesis group, identical rates (1Ā·7 per cent) of chromosome abnormalities were found in the IVFā€“ET and control groups.     Mitochondrial Ca2+ Overload Underlies AĪ² Oligomers Neurotoxicity Providing an Unexpected Mechanism of Neuroprotection by NSAIDs

    Dysregulation of intracellular Ca2+ homeostasis may underlie amyloid Ī² peptide (AĪ²) toxicity in Alzheimer's Disease (AD) but the mechanism is unknown. In search for this mechanism we found that AĪ²1ā€“42 oligomers, the assembly state correlating best with cognitive decline in AD, but not AĪ² fibrils, induce a massive entry of Ca2+ in neurons and promote mitochondrial Ca2+ overload as shown by bioluminescence imaging of targeted aequorin in individual neurons. AĪ² oligomers induce also mitochondrial permeability transition, cytochrome c release, apoptosis and cell death. Mitochondrial depolarization prevents mitochondrial Ca2+ overload, cytochrome c release and cell death. In addition, we found that a series of non-steroidal anti-inflammatory drugs (NSAIDs) including salicylate, sulindac sulfide, indomethacin, ibuprofen and R-flurbiprofen depolarize mitochondria and inhibit mitochondrial Ca2+ overload, cytochrome c release and cell death induced by AĪ² oligomers. Our results indicate that i) mitochondrial Ca2+ overload underlies the neurotoxicity induced by AĪ² oligomers and ii) inhibition of mitochondrial Ca2+ overload provides a novel mechanism of neuroprotection by NSAIDs against AĪ² oligomers and AD

    On the surface topography of ultrashort laser pulse treated steel surfaces

    This paper concentrates on observations of the surface topography by scanning electron microscopy (SEM) on alloyed and stainless steels samples treated by ultrashort laser pulses with duration of 210 fs and 6.7 ps. Globular-like and jet-like objects were found depending on the various levels of the fluence applied. It is shown that these features appear due to solid-liquid and liquid-gas transitions within surface layer irradiated by intense laser light. The observations are confronted to the theory of short-pulsed laser light-matter interactions, including interference, excitation of electrons, electron-phonon coupling as well as subsequent ablation. It is shown that the orientation of small ripples does not always depend on the direction of the polarization of laser light. (C) 2011 Elsevier B.V. All rights reserved
