    The Neglected Remains of the Royal Citadel of Messina, Sicily: A Proposal for a Suitable Conservation and Re-Use Project

    Neglected and abandoned heritage sites are a complex research topic, which always requires a thorough understanding of the site’s past and current condition. This article examines how research on history, architecture, and participation can contribute to creating a suitable re-use project for such heritage sites, focusing on a single case study: the Royal Citadel of Messina, Italy. Within the field of history, the city and fortification’s past underscores the importance of the site’s architectural and historical value. An urban analysis and documentation campaign were carried out during the field architecture. The urban analysis focused on the site as a remarkable natural landscape with industrial surroundings. The digital documentation, concentrating on the so-called cistern, emphasized the site’s values. Within a revalorization process, community participation is vital and is, therefore, one of the suggestions to consider in further developments. The last applied method is a comparative case study, namely, the submarine base at Saint-Nazaire, France. Considering the complexity and fragilities of the site, a framework is designed that provides suggestions for the conservation, revalorization, and re-use of the site of the Royal Citadel

    Shape characterization of polymersome morphologies via light scattering techniques

    Polymersomes, vesicles self-assembled from amphiphilic block copolymers, are well known for their robustness and for their broad applicability. Generating polymersomes of different shape is a topic of recent attention, specifically in the field of biomedical applications. To obtain information about their exact shape, tomography based on cryo-electron microscopy is usually the most preferred technique. Unfortunately, this technique is rather time consuming and expensive. Here we demonstrate an alternative analytical approach for the characterization of differently shaped polymersomes such as spheres, prolates and discs via the combination of multi-angle light scattering (MALS) and quasi-elastic light scattering (QELS). The use of these coupled techniques allowed for accurate determination of both the radius of gyration (Rg) and the hydrodynamic radius (Rh). This afforded us to determine the shape ratio ρ (Rg/Rh) with which we were able to distinguish between polymersome spheres, discs and rods.</p

    Soft tissue and visceral sarcomas: ESMO-EURACAN-GENTURIS Clinical Practice Guidelines for diagnosis, treatment and follow-up

    Soft tissue sarcomas (STSs) comprise ∼80 entities defined by the World Health Organization (WHO) classification based on a combination of distinctive morphological, immunohistochemical and molecular features.1 These ESMO–EURACAN–GENTURIS (European Society for Medical Oncology; European Reference Network for Rare Adult Solid Cancers; European Reference Network for Genetic Tumour Risk Syndromes) Clinical Practice Guidelines (CPGs) will cover STSs, with the exception of gastrointestinal stromal tumours (GISTs) that are covered in the ESMO–EURACAN–GENTURIS GIST CPGs.2 EURACAN and GENTURIS are the European Reference Networks connecting European institutions, appointed by their governments, to cover rare adult solid cancers and genetic cancer risk syndromes, respectively. Extraskeletal Ewing sarcoma, round cell sarcoma with EWSR1-non-ETS fusion and sarcomas with CIC rearrangements and BCOR genetic alterations are covered by the ESMO–EURACAN–GENTURIS–ERN PaedCan (European Reference Network for Paediatric Oncology) bone sarcomas CPG.3 Kaposi's sarcoma, embryonal and alveolar rhabdomyosarcoma are not discussed in this manuscript, while pleomorphic rhabdomyosarcoma is viewed as a high-grade, adult-type STS. Finally, extraskeletal osteosarcoma is also a considered a high-grade STS, whose clinical resemblance with osteosarcoma of bone is doubtful.     Immunolocalization of catecholamine enzymes, serotonin, dopamine and L-dopa in the brain of Dicentrarchus labrax (Teleostei)

    Antisera to serotonin (5-HT), dopamine, and L-dopa, and to the catecholamine synthesizing enzymes, tyrosine hydroxylase (TH), dopamine beta-hydroxylase (DBH), and phenylethanolamine N-methyl transferase (PNMT), were used to localize monoamine containing neurones in the brain of Dicentrarchus labrax (sea bass). In the brain stem, 5-HT-immunoreactive (ir) neurones were recognized in the ventrolateral medulla, vagal motor area, medullary, and mesencephalic raphe nuclei and in the dorsolateral isthmal tegmentum. In the hypothalamus, liquor-contacting 5-HT neurones were seen in various regions of the paraventricular organ. Virtually all regions of the brain contained a dense innervation by 5-HT fibres and terminals. DBH-ir neurones were restricted to three brain stem areas: the locus coeruleus, the area postrema, and the reticular formation of the lower medulla. Neurones in these three groups also displayed TH-ir, and in the latter area, PNMT-ir in addition. In the locus coeruleus and area postrema, TH-ir neurones outnumbered DBH-ir neurones, an observation substantiated by the presence of dopamine-ir neurones. In the forebrain, dopamine- and TH-ir neurones were found in the olfactory bulb, ventral/central telencephalon, periventricular preoptic, and suprachiasmatic areas, dorsolateral and ventromedial thalamus, and posterior tuberal nucleus. In the paraventricular organ, the distribution and morphology of dopamine-ir neurones was similar to that observed with anti-5-HT, but the vast majority of cells were not TH-ir, suggesting accumulation of dopamine by uptake from the ventricle, rather than by synthesis. L-dopa-ir neurones were found only in the central telencephalon, preoptic recess, and dorsolateral thalamus. Fibres and terminals immunoreactive for dopamine, TH, and DBH showed a broadly similar distribution.     Effect of sensory deafferentation on the gabaergic circuitry of the adult cat visual system

    The effect of bilateral central retinal lesions on the GAD67 and GAD65 messenger RNA levels in the dorsal lateral geniculate nucleus, the perigeniculate nucleus and the visual cortex of the adult cat was investigated by in situ hybridization. Three days post-lesion, a decrease in the number of GAD67-expressing cells was apparent in the deafferented dorsal lateral geniculate nucleus. This decrease persisted until 7.5 months post-lesion and was more pronounced with longer survival times. The decrease in GAD67 mRNA was mirrored by a decrease in glutamate decarboxylase- immunoreactive cells. GAD65 messenger RNA expression levels were low in the dorsal lateral geniculate nucleus of both control and retinally-lesioned cats. In the perigeniculate nucleus the messenger RNA levels of both glutamate decarboxylase isoforms were clearly decreased over a restricted region. In the lesion-affected visual cortex, no changes at the messenger RNA level were observed for either GAD67 or GAD65 although changes in glutamate decarboxylase immunoreactivity have been previously described. Hence, in the dorsal lateral geniculate nucleus, the perigeniculate nucleus and the visual cortex, different intracellular mechanisms seem to lead to decreased GABAergic inhibition in response to sensory deafferentation.SCOPUS: ar.jinfo:eu-repo/semantics/publishe

    Early entry events in Echovirus 30 infection

    Echovirus 30 (E30), a member of the enterovirus B species, is a major cause of viral meningitis, targeting children and adults alike. While it is a frequently isolated enterovirus and the cause of several outbreaks all over the world, suprisingly little is known regarding its entry and replication strategy within cells. In this study, we used E30 Bastianni (E30B) generated from an infectious cDNA clone in order to study early entry events during infection in human RD cells. E30B required the newly discovered Fc echovirus receptor (FcRn) for succesful infection, but not the Coxsackievirus and Adenovirus Receptor (CAR) or Decay-Accelerating Factor (DAF), although an interaction with DAF was observed. Double-stranded RNA replication intermediate was generated between 2 and 3 h post-infection (p.i.). and viral capsid production was initiated between 4 and 5 h p.i. The drugs affecting Rac1 (NSC 23766) and cholesterol (Filipin III) compromised infection, whereas bafilomycin A1, dyngo, U-73122, wortmannin and nocodazole did not, suggesting the virus follows an enterovirus-triggered macropinocytic pathway rather than the clathrin pathway. Colocalization with early endosomes and increased infection due to constitutively active Rab5 expression suggests some overlap and entry to classical early endosomes. Taken together, these results suggest that E30B induces an enterovirus entry pathway, leading to uncoating in early endosomes.peerReviewe
