LCA of the South African sugar industry
South Africa is one of the world’s leading producers of high quality sugar, producing approximately 2.5 million tonnes per annum. The South African sugar industry makes a significant contribution to the South African national economy, generating direct income of approximately 6 billion South African Rand (R) per year (US$700 million or €500 million) (SASA 2008). The industry employs approximately 85,000 people in cane production and processing, and also indirectly provides jobs in numerous support industries such as fertiliser, chemical, transport and food industries (SASA 2008)
Protein expression and genetic variability of canine Can f 1 in golden and Labrador retriever service dogs.
BackgroundValued for trainability in diverse tasks, dogs are the primary service animal used to assist individuals with disabilities. Despite their utility, many people in need of service dogs are sensitive to the primary dog allergen, Can f 1, encoded by the Lipocalin 1 gene (LCN1). Several organizations specifically breed service dogs to meet special needs and would like to reduce allergenic potential if possible. In this study, we evaluated the expression of Can f 1 protein and the inherent variability of LCN1 in two breeds used extensively as service dogs. Saliva samples from equal numbers of male and female Labrador retrievers (n = 12), golden retrievers (n = 12), and Labrador-golden crosses (n = 12) were collected 1 h after the morning meal. Can f 1 protein concentrations in the saliva were measured by ELISA, and the LCN1 5' and 3' UTRs and exons sequenced.ResultsThere was no sex effect (p > 0.2) nor time-of-day effect; however, Can f 1 protein levels varied by breed with Labrador retrievers being lower than golden retrievers (3.18 ± 0.51 and 5.35 ± 0.52 μg/ml, respectively, p < 0.0075), and the Labrador-golden crosses having intermediate levels (3.77 ± 0.48 μg/ml). Although several novel SNPs were identified in LCN1, there were no significant breed-specific sequence differences in the gene and no association of LCN1 genotypes with Can f 1 expression.ConclusionsAs service dogs, Labrador retrievers likely have lower allergenic potential and, though there were no DNA sequence differences identified, classical genetic selection on the estimated breeding values associated with salivary Can f 1 expression may further reduce that potential

Cauchy, infinitesimals and ghosts of departed quantifiers
Procedures relying on infinitesimals in Leibniz, Euler and Cauchy have been
interpreted in both a Weierstrassian and Robinson's frameworks. The latter
provides closer proxies for the procedures of the classical masters. Thus,
Leibniz's distinction between assignable and inassignable numbers finds a proxy
in the distinction between standard and nonstandard numbers in Robinson's
framework, while Leibniz's law of homogeneity with the implied notion of
equality up to negligible terms finds a mathematical formalisation in terms of
standard part. It is hard to provide parallel formalisations in a
Weierstrassian framework but scholars since Ishiguro have engaged in a quest
for ghosts of departed quantifiers to provide a Weierstrassian account for
Leibniz's infinitesimals. Euler similarly had notions of equality up to
negligible terms, of which he distinguished two types: geometric and
arithmetic. Euler routinely used product decompositions into a specific
infinite number of factors, and used the binomial formula with an infinite
exponent. Such procedures have immediate hyperfinite analogues in Robinson's
framework, while in a Weierstrassian framework they can only be reinterpreted
by means of paraphrases departing significantly from Euler's own presentation.
Cauchy gives lucid definitions of continuity in terms of infinitesimals that
find ready formalisations in Robinson's framework but scholars working in a
Weierstrassian framework bend over backwards either to claim that Cauchy was
vague or to engage in a quest for ghosts of departed quantifiers in his work.
Cauchy's procedures in the context of his 1853 sum theorem (for series of
continuous functions) are more readily understood from the viewpoint of
Robinson's framework, where one can exploit tools such as the pointwise
definition of the concept of uniform convergence.

Vertebrate Hosts as Islands: Dynamics of Selection, Immigration, Loss, Persistence, and Potential Function of Bacteria on Salamander Skin.
Skin bacterial communities can protect amphibians from a fungal pathogen; however, little is known about how these communities are maintained. We used a neutral model of community ecology to identify bacteria that are maintained on salamanders by selection or by dispersal from a bacterial reservoir (soil) and ecological drift. We found that 75% (9/12) of bacteria that were consistent with positive selection, <1% of bacteria that were consistent with random dispersal and none of the bacteria that were consistent under negative selection had a 97% or greater match to antifungal isolates. Additionally we performed an experiment where salamanders were either provided or denied a bacterial reservoir and estimated immigration and loss (emigration and local extinction) rates of bacteria on salamanders in both treatments. Loss was strongly related to bacterial richness, suggesting competition is important for structuring the community. Bacteria closely related to antifungal isolates were more likely to persist on salamanders with or without a bacterial reservoir, suggesting they had a competitive advantage. Furthermore, over-represented and under-represented operational taxonomic units (OTUs) had similar persistence on salamanders when a bacterial reservoir was present. However, under-represented OTUs were less likely to persist in the absence of a bacterial reservoir, suggesting that the over-represented and under-represented bacteria were selected against or for on salamanders through time. Our findings from the neutral model, migration and persistence analyses show that bacteria that exhibit a high similarity to antifungal isolates persist on salamanders, which likely protect hosts against pathogens and improve fitness. This research is one of the first to apply ecological theory to investigate assembly of host associated-bacterial communities, which can provide insights for probiotic bioaugmentation as a conservation strategy against disease
Microbial and metabolic succession on common building materials under high humidity conditions.
Despite considerable efforts to characterize the microbial ecology of the built environment, the metabolic mechanisms underpinning microbial colonization and successional dynamics remain unclear, particularly at high moisture conditions. Here, we applied bacterial/viral particle counting, qPCR, amplicon sequencing of the genes encoding 16S and ITS rRNA, and metabolomics to longitudinally characterize the ecological dynamics of four common building materials maintained at high humidity. We varied the natural inoculum provided to each material and wet half of the samples to simulate a potable water leak. Wetted materials had higher growth rates and lower alpha diversity compared to non-wetted materials, and wetting described the majority of the variance in bacterial, fungal, and metabolite structure. Inoculation location was weakly associated with bacterial and fungal beta diversity. Material type influenced bacterial and viral particle abundance and bacterial and metabolic (but not fungal) diversity. Molecular Targeting of Carbonic Anhydrase IX in Mice with Hypoxic HT29 Colorectal Tumor Xenografts
Background: Carbonic anhydrase IX (CAIX) is a membrane spanning protein involved in the enzymatic regulation of tumor acid-base balance. CAIX has been shown to be elevated in a number of hypoxic tumor types. The purpose of this study was to determine the efficiency of intact and IgG fragments of cG250 to target CAIX in vivo in a hypoxic tumor model. Methodology/Principal Findings: Conventional biodistribution studies were performed with 111 In-DO3A-cG250, 111 In-DO3A-F(ab’)2-cG250 and 111 In-DO3A-Fab-cG250. Additional ex vivo analysis of the tumor was performed with markers for tumor hypoxia, blood perfusion and endogenous CAIX expression. All four data sets were digitally correlated to determine the optimal agent for determining hypoxia in a HT29 colon cancer xenograft. The HT29 human colorectal tumor xenografts show strong CAIX expression in hypoxic areas of poor blood perfusion. The intact IgG had an initial high focal uptake at the periphery of these hypoxic regions and penetration into the areas of highest CAIX expression over the 7-day study period. The lower molecular weight antibody fragments had a faster uptake into areas of high CAIX expression, but had a much lower absolute uptake at the optimal imaging times. Identifying Irrigated Areas in the Snake River Plain, Idaho: Evaluating Performance Across Composting Algorithms, Spectral Indices, and Sensors
There are pressing concerns about the interplay between agricultural productivity, water demand, and water availability in semi-arid to arid regions of the world. Currently, irrigated agriculture is the dominant water user in these regions and is estimated to consume approximately 80% of the world’s diverted freshwater resources. We develop an improved irrigated land-use mapping algorithm that uses the seasonal maximum value of a spectral index to distinguish between irrigated and non-irrigated parcels in Idaho’s Snake River Plain. We compare this approach to two alternative algorithms that differentiate between irrigated and non-irrigated parcels using spectral index values at a single date or the area beneath spectral index trajectories for the duration of the agricultural growing season. Using six different pixel and county-scale error metrics, we evaluate the performance of these three algorithms across all possible combinations of two growing seasons (2002 and 2007), two datasets (MODIS and Landsat 5), and three spectral indices, the Normalized Difference Vegetation Index, Enhanced Vegetation Index and Normalized Difference Moisture Index (NDVI, EVI, and NDMI). We demonstrate that, on average, the seasonal-maximum algorithm yields an improvement in classification accuracy over the accepted single-date approach, and that the average improvement under this approach is a 60% reduction in county scale root mean square error (RMSE), and modest improvements of overall accuracy in the pixel scale validation. The greater accuracy of the seasonal-maximum algorithm is primarily due to its ability to correctly classify non-irrigated lands in riparian and developed areas of the study region
