    The epigenetic control of E-box and Myc-dependent chromatin modifications regulate the licensing of lamin B2 origin during cell cycle

    Recent genome-wide mapping of the mammalian replication origins has suggested the role of transcriptional regulatory elements in origin activation. However, the nature of chromatin modifications associated with such trans-factors or epigenetic marks imprinted on cis-elements during the spatio-temporal regulation of replication initiation remains enigmatic. To unveil the molecular underpinnings, we studied the human lamin B2 origin that spatially overlaps with TIMM 13 promoter. We observed an early G1-specific occupancy of c-Myc that facilitated the loading of mini chromosome maintenance protein (MCM) complex during subsequent mid-G1 phase rather stimulating TIMM 13 gene expression. Investigations on the Myc-induced downstream events suggested a direct interaction between c-Myc and histone methyltransferase mixed-lineage leukemia 1 that imparted histone H3K4me3 mark essential for both recruitment of acetylase complex HBO1 and hyperacetylation of histone H4. Contemporaneously, the nucleosome remodeling promoted the loading of MCM proteins at the origin. These chromatin modifications were under the tight control of active demethylation of E-box as evident from methylation profiling. The active demethylation was mediated by the Ten-eleven translocation (TET)-thymine DNA glycosylase-base excision repair (BER) pathway, which facilitated spatio-temporal occupancy of Myc. Intriguingly, the genome-wide 43% occurrence of E-box among the human origins could support our hypothesis that epigenetic control of E-box could be a molecular switch for the licensing of early replicating origins

    Critical Appraisal of Baby Friendly Hospital Initiatives among the beneficiaries of Indore Urban - A Comparative Study

    Background: Majority of the women in M.P who breastfeed their babies throw away the colostrum before they begin actual feed. Only 14.9% females feed their babies with in 1/2 an hour (National Family Health Survey-3). In Indore 2 hospitals have been given designation of baby friendly hospital (BFH) namely M.Y. hospital (MYH) which is a tertiary care center, attached to Medical College and District hospital (DH). Objectives: (1) To assess the process of implementation of baby friendly hospital initiative programme. (2) Knowledge, Attitude, Practice regarding breast feeding amongst the beneficiaries. Methods: A comparative study was conducted between two BFH and two NBFH (Non Baby Friendly Hospitals) of Indore. The respondents were categorized into health care providers and beneficiaries. A pretested questionnaire was used to interview the respondents which was made taking into consideration the essential 10 Criteria of BFHI Programme. Results: 89% in BFH and 97% in NBFH were having positive attitude towards colostrum feeding. 81% postnatal mothers in BFH and 96% in NBFH were having knowledge about benefits of exclusive breast feeding. 21% postnatal mothers in BFH and 39% in NBFH initiated breast feeding within ½ an hour. 64 % of mothers in BFH and 62 % mothers in NBFH were practicing exclusive breast Feeding. Conclusion: The above findings show that although designated as BFH, the breast feeding practices remained poor as compared to NBFH. There is an utmost need to reinforce training and constant monitoring of health care providers regarding BFHI

    Multimodal Magnetic Resonance and Near-Infrared-Fluorescent Imaging of Intraperitoneal Ovarian Cancer Using a Dual-Mode-Dual-Gadolinium Liposomal Contrast Agent.

    The degree of tumor removal at surgery is a major factor in predicting outcome for ovarian cancer. A single multimodality agent that can be used with magnetic resonance (MR) for staging and pre-surgical planning, and with optical imaging to aid surgical removal of tumors, would present a new paradigm for ovarian cancer. We assessed whether a dual-mode, dual-Gadolinium (DM-Dual-Gd-ICG) contrast agent can be used to visualize ovarian tumors in the peritoneal cavity by multimodal MR and near infra-red imaging (NIR). Intraperitoneal ovarian tumors (Hey-A8 or OVCAR3) in mice enhanced on MR two days after intravenous DM-Dual Gd-ICG injection compared to controls (SNR, CNR, p < 0.05, n = 6). As seen on open abdomen and excised tumors views and confirmed by optical radiant efficiency measurement, Hey-A8 or OVCAR3 tumors from animals injected with DM-Dual Gd-ICG had increased fluorescence (p < 0.05, n = 6). This suggests clinical potential to localize ovarian tumors by MR for staging and surgical planning, and, by NIR at surgery for resection

    Leprosy and tuberculosis concomitant infection: a poorly understood, age-old relationship

    Historically, archaeological evidence, post-mortem findings and retro- spective analysis of leprosy institutions’ data demonstrates a high observed incidence of concomitant infection with leprosy and tuberculosis (TB). However, reports of concomitant infection in the modern literature remain scarce, with estimates of annual new case detection rates of concomitant infection at approximately 0·02 cases per 100,000 population. Whilst the mechanism for this apparent decline in concomitant infections remains unclear, further research on this topic has remained relatively neglected. Modelling of the interaction of the two organisms has suggested that the apparent decline in observations of concomitant infection may be due to the protective effects of cross immunity, whilst more recently others have questioned whether it is a more harmful relationship, predisposing towards increased host mortality. We review recent evidence, comparing it to previously held understanding on the epidemiological relationship and our own experience of concomitant infection. From this discussion, we highlight several under-investigated areas, which may lead to improvements in the future delivery of leprosy management and services, as well as enhance understanding in other fields of infection management. These include, a) highlighting the need for greater understanding of host immunogenetics involved in concomitant infection, b) whether prolonged courses of high dose steroids pre-dispose to TB infection? and, c) whether there is a risk of rifampicin resistance developing in leprosy patients treated in the face of undiagnosed TB and other infections? Longitudinal work is still required to characterise these temporal relationships further and add to the current paucity of literature on this subject matter

    Airborne measurements of submicron aerosols across the coastline at Bhubaneswar during ICARB

    Airborne measurements of the number concentration and size distribution of aerosols from 13 to 700 nm diameter have been made at four vertical levels across a coastline at Bhubaneswar (20◦25N, 85◦83E) during the Integrated Campaign for Aerosols, gases and Radiation Budget (ICARB) programme conducted in March–April 2006. The measurements made during the constant-level flights at 0.5, 1, 2 and 3 km altitude levels extend ∼100 km over land and ∼150km over ocean. Aerosol number concentrations vary from 2200 to 4500 cm−3 at 0.5 km level but are almost constant at ∼ 6000 cm−3 and ∼ 800 cm−3 at 2 and 3 km levels, respectively. At 1km level, aerosol number concentration shows a peak of 18,070 cm−3 around the coastline. Most of the aerosol size distribution curves at 0.5 km and 1 km levels are monomodal with a maxima at 110nm diameter which shifts to 70 nm diameter at 2 and 3 km levels. However, at the peak at 1 km level, number concentration has a bimodal distribution with an additional maximum appearing in nucleation mode. It is proposed that this maxima in nucleation mode at 1 km level may be due to the formation and transport of new particles from coastal region

    The ion-aerosol interactions from the ion mobility and aerosol particle size distribution measurements on January 17 and February 18, 2005 at Maitri, Antarctica - A case study

    A case study for the ion–aerosol interactions is presented from the simultaneous measurements of mobility spectra of atmospheric ions in the mobility range of 2.29 to 2.98 × 10 − 4 cm2 V − 1 s −1 (diameter range 0.41–109 nm) and of size distribution of atmospheric aerosol particles in the size ranges of 4.4–700 nm and 500–20,000 nm diameters made at Maitri (70°45′52′ ′S, 11°44′2.7′ ′E; 130 m above mean sea level), Antarctica, on two days January 17 and February 18, 2005, with contrasting meteorological conditions. In contrast to January 17, on February 18, winds were stronger from the morning to noon and lower from the noon to evening, atmospheric pressure was lower, cloudiness was more, the land surface remained snow-covered after a blizzard on February 16 and 17 and the airmass over Maitri, descended from an altitude of ~3 km after an excursion over ocean. On these days mobility spectra showed two modes, corresponding to intermediate ions and light large ions and an indication of additional one/two maxima for small/cluster ions and heavy large ions. The small ions generated by cosmic rays, and the nucleation mode particles generated probably by photochemical reactions grew in size by condensation of volatile trace gases on them and produced the cluster and intermediate ion modes and the Aitken particle mode in ion/particle spectra. Particles in the size range of 9–26 nm have been estimated to grow at the rate of 1.9 nm h − 1 on February 18, 2005. Both, ions and aerosol particles show bimodal size distributions in the 16–107 nm size range, and comparison of the two size distributions suggests the formation of multiple charged ions. Attachment of small ions to particles in this bimodal distribution of Aitken particles together with the formation of multiple charged ions are proposed to result in the light and heavy large ion modes. Growth of the nucleation mode particles on February 18, 2005 is associated with the passage of the airmass over ocean. In contrast, though the ion size distributions were not much different, the aerosol size distributions did not show a dominant peak for the formation and growth of nucleation mode particles on January 17. More measurements are needed before the conclusion of this case study is generalized

    Changes in concentration and size distribution of aerosols during fog over the south Indian Ocean

    Measurements of the concentration and size distribution of aerosol particles in the size-ranges of 0.5-20 μm and 16-700 nm diameters were made during six fog episodes over the south Indian Ocean. Observations show that concentrations of particles of all sizes start decreasing 1-2 hours before the occurrence of fog. This decrease is more prominent for coarse particles of >1 μm diameter and continues until 10-20 minutes before the onset of fog when particle concentrations in all size ranges rapidly increase by one/two orders of magnitude in ~20 minutes. Thereafter, concentrations of particles of all sizes gradually decrease until the dissipation of fog. After the fog dissipation, concentrations of coarse mode particles rapidly increase and restore to their pre-fog levels but concentrations of the Aitken mode particles decrease slowly and reach their pre-fog levels only after 1-2 hours. The net effect of fog is to change the bimodal size distributions of aerosols with a coarse mode at 1.0 μm and an accumulation mode at 40-60 nm to a power law size distribution. It is proposed that the preferential growth and sedimentation of the coarse mode hygroscopic particles in the initial phase cause a large decrease in the aerosol surface area. As a result, the low vapour pressure gases which were initially being used for the growth of coarse mode particles, now accelerate the growth rates of the accumulation and Aitken mode particles
