    Avian Influenza H9N2 Seroprevalence among Poultry Workers in Pune, India, 2010

    Avian influenza (AI) H9N2 has been reported from poultry in India. A seroepidemiological study was undertaken among poultry workers to understand the prevalence of antibodies against AI H9N2 in Pune, Maharashtra, India. A total of 338 poultry workers were sampled. Serum samples were tested for presence of antibodies against AI H9N2 virus by hemagglutination inhibition (HI) and microneutralization (MN) assays. A total of 249 baseline sera from general population from Pune were tested for antibodies against AI H9N2 and were negative by HI assay using ≥40 cut-off antibody titre. Overall 21 subjects (21/338 = 6.2%) were positive for antibodies against AI H9N2 by either HI or MN assays using ≥40 cut-off antibody titre. A total of 4.7% and 3.8% poultry workers were positive for antibodies against AI H9N2 by HI and MN assay respectively using 40 as cut-off antibody titre. This is the first report of seroprevalence of antibodies against AI H9N2 among poultry workers in India

    Transgenic tobacco plants constitutively expressing Arabidopsis NPR1 show enhanced resistance to root-knot nematode, Meloidogyne incognita

    In Arabidopsis, non-expressor of pathogenesis related genes-1, NPR1 has been shown to be a positive regulator of the salicylic acid controlled systemic acquired resistance pathway and modulates the cross talk between SA and JA signaling. Transgenic plants expressing AtNPR1 constitutively exhibited resistance against pathogens as well as herbivory. In the present study, tobacco transgenic plants expressing AtNPR1 were studied further for their response to infection by the sedentary endoparasitic root knot nematode, Meloidogyne incognita. Transgenic plants showed enhanced resistance against the root-knot nematode infection. Prominent differences in the shoot and root weights of wild type and transgenic plants were observed post-inoculation with M. incognita. This was associated with a decrease in the number of root galls and egg masses in transgenic plants compared to WT. The transgenic plants also showed constitutive and induced expression of some PR protein genes, when challenged with M. incognita

    Mature seed-derived callus of the model indica rice variety Kasalath is highly competent in Agrobacterium-mediated transformation

    We previously established an efficient Agrobacterium-mediated transformation system using primary calli derived from mature seeds of the model japonica rice variety Nipponbare. We expected that the shortened tissue culture period would reduce callus browning—a common problem with the indica transformation system during prolonged tissue culture in the undifferentiated state. In this study, we successfully applied our efficient transformation system to Kasalath—a model variety of indica rice. The Luc reporter system is sensitive enough to allow quantitative analysis of the competency of rice callus for Agrobacterium-mediated transformation. We unexpectedly discovered that primary callus of Kasalath exhibits a remarkably high competency for Agrobacterium-mediated transformation compared to Nipponbare. Southern blot analysis and Luc luminescence showed that independent transformation events in primary callus of Kasalath occurred successfully at ca. tenfold higher frequency than in Nipponbare, and single copy T-DNA integration was observed in ~40% of these events. We also compared the competency of secondary callus of Nipponbare and Kasalath and again found superior competency in Kasalath, although the identification and subsequent observation of independent transformation events in secondary callus is difficult due to the vigorous growth of both transformed and non-transformed cells. An efficient transformation system in Kasalath could facilitate the identification of QTL genes, since many QTL genes are analyzed in a Nipponbare × Kasalath genetic background. The higher transformation competency of Kasalath could be a useful trait in the establishment of highly efficient systems involving new transformation technologies such as gene targeting

    Cisgenesis and intragenesis as new strategies for crop improvement

    Cisgenesis and intragenesis are emerging plant breeding technologies which offer great promise for future acceptance of genetically engineered crops. The techniques employ traditional genetic engineering methods but are confined to transferring of genes and genetic elements between sexually compatible species that can breed naturally. One of the main requirements is the absence of selectable marker genes (such as antibiotic resistance genes) in the genome. Hence the sensitive issues with regard to transfer of foreign genes and antibiotic resistance are overcome. It is a targeted technique involving specific locus; therefore, linkage drag that prolongs the time for crop improvement in traditional breeding does not occur. It has great potential for crop improvement using superior alleles that exist in the untapped germplasm or wild species. Cisgenic and intragenic plants may not face the same stringent regulatory assessment for field release as transgenic plants which is a clear added advantage that would save time. In this chapter, the concepts of cis/intragenesis and the prerequisites for the development of cis/intragenesis plants are elaborated. Strategies for marker gene removal after selection of transformants are discussed based on the few recent reports from various plant species

    Template-based object detectors such as the deformable parts model of Felzenszwalb et al. [11] achieve state-ofthe-art performance for a variety of object categories, but are still outperformed by simpler bag-of-words models for highly flexible objects such as cats and dogs. In these cases we propose to use the template-based model to detect a distinctive part for the class, followed by detecting the rest of the object via segmentation on image specific information learnt from that part. This approach is motivated by two observations: (i) many object classes contain distinctive parts that can be detected very reliably by template-based detectors, whilst the entire object cannot; (ii) many classes (e.g. animals) have fairly homogeneous coloring and texture that can be used to segment the object once a sample is provided in an image. We show quantitatively that our method substantially outperforms whole-body template-based detectors for these highly deformable object categories, and indeed achieves accuracy comparable to the state-of-the-art on the PASCAL VOC competition, which includes other models such as bagof-words. 1

    Glimpses of Patala (Stereospermum Suaveolens roxb. Dc.) in Rasashastra: A Review

    In the universe of Ayurveda, Patala (Stereospermum suaveolens Roxb. DC.) is renowned drug credited with variety of therapeutic properties as well as other specific uses. It is also the member of much celebrated Mishraka Varga like Bruhatpanchmoola and Dashmoola. Rasashastra i.e., Indian alchemy or Iatrochemistry deals primarily with metals, minerals, gems etc. but it is inseparable from different flora and fauna. This sincere attempt helps ease out the efforts to draw information on Patala, Bruhatpanchmoola and Dashmoola from Rasashastra domain. For this purpose, information pertaining to Patala and other two herbal groups was screened from classical Rasashastra texts available at central library of ITRA, Jamnagar. Gathered information according to relevance and specificity was arranged in tabular form under the headings like name of group, processes like Shodhana, Marana, formulation name, mode of administration, dosage form etc. Total 18 groups of herbs, 164 formulations (excluding references of processes like Shodhana/Marana) were found having their therapeutic efficacy in almost 28 indications with 14 dosage forms at their disposal. Dashmoola in formulations, Jvara in therapeutic indications and Rasaushadhi in dosage forms were found in majority. Lastly general information of Patala, in the context of Dinacharya, Rutucharya, Rasashala, its simile also quoted. This review depicts multimodal applications of Patala along with its group of herbs in Rasashastra. The write-up will serve as in hand guide for the knowledge of Patala and its associated groups of herbs in the branch of Rasashastra

    Molecular characterization of marker-free transgenic lines of indica rice that accumulate carotenoids in seed endosperm

    A single Agrobacterium strain harbouring two binary plasmids was successfully used for the first time to develop a marker-free transgenic rice of improved nutritional value. Sixty-eight T0 co-transformants were obtained in three indica rice cultivars—two popular high-yielding Bangladeshi varieties (BR28 and BR29), and one high-iron rice cultivar (IR68144). Marker-free lines were obtained from 14 out of 24 selected co-transformants screened in the T1 generation. The accumulation of total carotenoids in polished T2 rice seeds of the primary transgenic VPBR29-17-37 reached levels of up to 3.0 μg/g, with the level of β-carotene reaching 1.8 μg/g. In the cultivars BR28 and IR68144, total carotenoid levels in the transformants reached 2.0 μg/g of polished rice seeds. The levels of lutein and other carotenoids in the seeds were also significantly enhanced. T1 plants obtained from primary transgenics with simple gene-integration patterns tended to have a lower carotenoid content than the original parental lines. This study describes the development of marker-free transgenic rice lines containing high levels of carotenoids, and addresses the relationship between the rearrangement of transgenes and the presence of metabolic end products in transgenic rice
