Практические аспекты применения валидационных параметров на примере методик определения количественного содержания микроорганизмов в лекарственных препаратах
Validation/verification of microbiological methods is a prerequisite for quality control of non-sterile drugs. However, the use of existing procedures for validation/verification of analytical methods is challenging, since a number of factors, such as microorganism distribution in the sample, cell morphology, and metabolic activity of microorganisms contribute to the error in microbiological testing.The aim of the study was to assess the feasibility of using the microbiological method validation parameters for validation/verification of the agar plate method.Materials and methods: 18 non-sterile medicinal products were used in the study. Experiments included determination of antimicrobial activity. The quantification of viable bacteria, yeasts and moulds was performed using the modified pour plate method. The statistical processing of the obtained results was performed using Microsoft Excel 7.0 and Statistica 8.0.Results: the paper provides the results of quantitative determination of test microorganisms inoculated into non-sterile drugs. The results were obtained as part of validation/verification of the agar plate method of the State Pharmacopoeia of the Russian Federation, XIV ed.Conclusions: the validation/verification of the test method for isolation and quantification of microorganisms revealed no deviations of the study results from the established acceptance criteria. This proves the feasibility of using the following validation parameters: accuracy, precision, robustness, and limit of quantitation when validating new methods for quantitative determination of microorganisms or verification of previously validated methods.Валидация/верификация микробиологических методик необходима при контроле качества нестерильных лекарственных средств, однако использование известных процедур валидации/верификации аналитических методик затруднительно, поскольку микробиологическая погрешность определяется такими факторами, как распределение микроорганизмов в образце, клеточная морфология и метаболическая активность микроорганизмов.Цель работы: оценка возможности применения параметров, используемых для валидации микробиологических методик, при валидации/ верификации чашечного агарового метода.Материалы и методы: нестерильные лекарственные средства 18 наименований. Предварительно исследовали антимикробное действие нестерильных лекарственных средств, для количественного определения жизнеспособных бактерий, дрожжевых и плесневых грибов применяли глубинный модифицированный метод. Статистическую обработку результатов проводили при помощи компьютерных программ Microsoft Excel 7.0 и Statistica 8.0.Результаты: представлены результаты количественного определения тест-штаммов микроорганизмов, инокулированных в нестерильные лекарственные средства, полученные в рамках валидации/верификации чашечного агарового метода Государственной фармакопеи Российской Федерации XIV изд.Выводы: при валидации/верификации методики количественного выделения микроорганизмов не было выявлено отклонений результатов исследований от установленных критериев приемлемости. Это доказывает возможность применения валидационных параметров «правильность», «прецизионность», «устойчивость», «предел количественного обнаружения» при валидации новых методик количественного определения микроорганизмов или верификации ранее валидированных
ELIMINATION OF FALSE RESULTS OF MEDICINES MICROBIOLOGICAL TESTING
The validity of medicines microbial quality testing relies on the adequacy of the test procedure employed. The aim of the study was to analyse factors triggering false results during microbial quality testing of non-sterile medicinal products, as well as to find ways of their elimination. Materials and methods: the study was focused on non-sterile medicinal products tested for microbial quality: N-methylglucamine, L-Malic acid, Xeroform, Fingolimod hydrochloride, Succinic acid, Streptocide, Aripiprazole, Doxazosin, Clopidogrel, Moxonidine, Tilorone, Mycophenolic acid, Folic acid, Gabapentin, Dutasteride, Imatinib, Temozolomide. The study involved the use of the following test strains: Bacillus subtilis, Bacillus cereus, Candida albicans, Escherichia coli, Aspergillus brasiliensis, as well as of reagents and growth media. The methods used were determination of antimicrobial activity under conditions of microbial quality testing, and modified in-depth testing of microbial quality of medicinal products according to the requirements of the State Pharmacopoeia of the Russian Federation, 13th edition. Results: the analysis of literature sources helped reveal the main factors causing false results of microbiological testing and determine ways of their elimination. The article sets forth the results of experimental comparison of two ways of sample preparation for solid formulations: the standard one described in the State Pharmacopoeia of the Russian Federation, 13th edition, and the one involving the use of a laboratory shaker. The article provides experimental data on specific aspects of elimination of antimicrobial activity against B. subtilis and B. cereus and the use of specific inactivators for particular medicinal products. Conclusions: a set of measures aimed at prevention of false-positive and false-negative testing results should include: sterility control of the growth media and reagents used, monitoring of facilities; control of growth promotion properties and selectivity of the growth media; selection of adequate incubation conditions and inoculation procedure with due regard to the dosage form; justification of the amount of sample, diluents and dilution factor used; consideration of the antimicrobial activity of a medicinal product
Экспериментальная оценка методов определения антимикробной активности препаратов хлорофиллипта
The present article describes the results of a comparative analysis of methods for determining the antimicrobial activity of chlorophyllipt. It mentions the aspects of serial dilution method and shows the advantages of using agar culture medium for subsequent recording of test results performed by the mentioned methods. The possibility of an equivalent use of cup and test-tube methods for the assessment of antimicrobial activity of chlorophyllipt in the form of alcohol and oil extracts has been confirmed. The necessity of exact compliance with the standards of laboratory procedures and initial verification of culture media quality was noted. During the experimental study of the antimicrobial activity of chlorophyllipt, using 4 types of microorganisms, the feasibility of using S. aureus. strain as a test microorganism has been confirmed. The experiment has shown that the choice of the method does not affect the result of the test in terms of «antimicrobial activity» and it only depends on the characteristics of the sample.Приведены результаты сравнительного анализа способов определения антимикробной активности препаратов хлорофиллипта. Описаны особенности методов серийных разведений и выявлены преимущества использования агаризованной питательной среды для последующего учета результатов испытания указанными методами. Подтверждена возможность равнозначного применения чашечного и пробирочного методов оценки антимикробной активности препаратов хлорофиллипта в виде спиртовых и масляных извлечений. Отмечена необходимость строгого соблюдения стандартности выполнения всех лабораторных процедур и предварительного подтверждения качества питательных сред. В ходе экспериментального изучения антимикробной активности хлорофиллипта с применением 4-х видов микроорганизмов была подтверждена целесообразность использования в качестве тест-микроорганизма штамм S. aureus. Экспериментально показано, что выбор метода не влияет на результат испытания по показателю «Антимикробная активность» и обусловлен лишь особенностями образца

Цель работы: изучение факторов, влияющих на получение ложных результатов исследования микробиологической чистоты нестерильных лекарственных средств, и определение путей их устранения. Материалы и методы: объектами исследования были нестерильные лекарственные средства, предварительно исследованные на микробиологическую чистоту: N-метилглюкамин; L-яблочная кислота; ксероформ; финголимода гидрохлорид; янтарная кислота; стрептоцид; арипипразол; доксазозин; клопидогрел; моксонидин; тилорон; микофеноловая кислота; фолиевая кислота; габапентин; дутастерид; иматиниб; темозоломид. Для исследований использовали тест-штаммы Bacillus subtilis, Bacillus cereus, Candida albicans, Escherichia coli, Aspergillus brasiliensis, реактивы, питательные среды и методы — определения антимикробного действия в условиях испытания микробиологической чистоты, модифицированный глубинный метод определения микробиологической чистоты ЛС в соответствии с требованиями Государственной фармакопеи Российской Федерации XIII изд. Результаты: на основании анализа данных литературы выявлены основные причины возникновения ложных результатов микробиологических испытаний и выбраны пути их предотвращения. Представлены результаты экспериментального сравнения двух вариантов пробоподготовки твердых лекарственных форм: стандартного по Государственной фармакопее Российской Федерации XIII изд. и с использованием встряхивателя лабораторного. Экспериментально обоснованы особенности устранения антимикробного действия в отношении B. subtilis и B. cereus и использования специфических инактиваторов для отдельных лекарственных средств. Выводы: система мероприятий по предотвращению ложноположительных и ложноотрицательных результатов исследования должна включать: контроль стерильности используемых питательных сред, реактивов, мониторинг помещений; контроль ростовых свойств и селективности питательных сред; выбор подходящих условий инкубации и методики посева с учетом лекарственной формы препарата; обоснование используемого количества образца, растворителя и фактора разведения; учет антимикробного действия лекарственного средства
