    Coupled aquaponics systems

    Coupled aquaponics is the archetype form of aquaponics. The technical complexity increases with the scale of production and required water treatment, e.g. filtration, UV light for microbial control, automatic controlled feeding, computerization and biosecurity. Upscaling is realized through multiunit systems that allow staggered fish production, parallel cultivation of different plants and application of several hydroponic subsystems. The main task of coupled aquaponics is the purification of aquaculture process water through integration of plants which add economic benefits when selecting suitable species like herbs, medicinal plants or ornamentals. Thus, coupled aquaponics with closed water recirculation systems has a particular role to fulfil. Under fully closed recirculation of nutrient enriched water, the symbiotic community of fish, plants and bacteria can result in higher yields compared with stand-alone fish production and/or plant cultivation. Fish and plant choices are highly diverse and only limited by water quality parameters, strongly influenced by fish feed, the plant cultivation area and component ratios that are often not ideal. Carps, tilapia and catfish are most commonly used, though more sensitive fish species and crayfish have been applied. Polyponics and additional fertilizers are methods to improve plant quality in the case of growth deficiencies, boosting plant production and increasing total yield. The main advantages of coupled aquaponics are in the most efficient use of resources such as feed for nutrient input, phosphorous, water and energy as well as in an increase of fish welfare. The multivariate system design approach allows coupled aquaponics to be installed in all geographic regions, from the high latitudes to arid and desert regions, with specific adaptation to the local environmental conditions. This chapter provides an overview of the historical development, general system design, upscaling, saline and brackish water systems, fish and plant choices as well as management issues of coupled aquaponics especially in Europe

    Evaluation of Dicentrarchus labrax Meats and the Vegetable Quality of Beta vulgaris var. cicla Farmed in freshwater and saltwater aquaponic systems

    The purpose of this study is to exploit the euryhaline nature of commercially attractive species for their cultivation in freshwater aquaponic systems. This approach may increase the profitability of aquaponic production in coastal countries where the consumption of marine fish is traditional and of commercial relevance. For this purpose, juvenile European sea bass (Dicentrarchus labrax) were reared in an aquaponic freshwater (AFW) system and an aquaponic saltwater (ASW) system (salinity 20 ppt), in combination with chard (Beta vulgaris var. cicla) seedlings, a salt tolerant plant. At the end of the trial, nitrate and phosphate concentration in water significantly increased in the ASW system, suggesting that the ability of B. vulgaris to absorb these substances was limited by salinity. Total reflection X-ray fluorescence spectrometry revealed that the concentration of some oligoelements such as Fe remained lower with respect to the concentration in the freshwater hydroponic solution, in both AFW and ASW. FTIR-Fourier transform infrared spectroscopy on plants showed that growth at high salinity affected their lipid content. In the case of fish, freshwater had no effects on mono- and poly-unsaturated fatty acid profiles, although saturated fatty acids were significantly decreased in D. labrax reared in AFW. Our results demonstrates that it is possible to increase aquaponic profitability by farming D. labrax juveniles in an aquaponic freshwater system together with Beta vulgaris, obtaining good quality products

    Xenobiotic-contaminated diets affect hepatic lipid metabolism: Implications for liver steatosis in Sparus aurata juveniles

    The metabolic effects induced by feed contaminated with a lower or a higher concentration of -nonylpnenol (NP), 4-tert-octylphenol (t-OP) or bisphenol A (BPA), three environmental endocrine disruptors, were assessed in juvenile sea bream liver. Histological analysis demonstrated that all these three xenobiotics induced hepatic lipid accumulation and steatosis. These findings prompted analysis of the expression of the major molecules involved in lipid metabolism: peroxisome proliferator activated receptors (which is encoded by ppars), fatty acid synthase (encoded by fas), lipoprotein lipase (encoded by lpl) and hormone-sensitive lipase (encoded by hsl). The enzymes encoded by ppars and fas are in fact responsible for lipid accumulation, whereas lpl- and hsl- encoded proteins play a pivotal role in fat mobilization. The three xenobiotics modulated ppar mRNA expression: ppar\u3b1 mRNA expression was induced by the higher dose of each contaminant; ppar\u3b2 mRNA expression was upregulated by the lower doses and in BPA2 fish ppary mRNA overexpression was induced by all pollutants. These data agreed with the lipid accumulation profiles documented by histology. Fas mRNA levels were modulated by the two NP doses and the higher BPA concentration. Lpl mRNA was significantly upregulated in all experimental groups except for BPA1 fish while hsl mRNA was significantly downregulated in all groups except for t-OP2 and BPA1 fish. The plasma concentrations of cortisol, the primary stress biomarker, were correlated with the levels of pepck mRNA level. This gene encodes phosphoenolpyruvate carboxykinase which is one of the key enzymes of gluconeogenesis. Pepck mRNA was significantly overexpressed in fish exposed to NP2 and both t-OP doses. Finally, the genes encoding cyclooxygenase 2 (cox2) and 5-lipoxygenase (5 lox), the products of which are involved in the inflammatory response, transcriptions were significantly upregulated in NP and BPA fish, whereas they were unchanged in t-OP specimens. The present findings suggest that dietary xenobiotic contamination can give rise to metabolic disorders also in fish and highlight the potential for their vertical transfer through the trophic levels and ultimately to humans

    In the last decades there have been several evidences that traditionally used live preys like rotifers and Artemia salina have nutritional deficiencies that result in a general decrease of fish health, causing anomalies in the development, in growth and in pigmentation. In this study a partial of total replacement of traditional live preys with preserved copepods that represent the natural food of the larvae was evaluated during Solea solea culture. In this study a positive effect of co-feeding preserved copepods in sole larviculture was observed since larvae fed this diet growth and survived better, showed a better tolerance to captive conditions and had a better response to the final thermal/density stress-test with respect to larvae fed a traditional diet. Morphometric data were fully supported by molecular and biochemical ones. Moreover, liver histological investigations, revealed that the inclusion of preserved copepods in the larval diet was able to improve lipid assimilation. In conclusion, preserved copepods may be considered a suitable food for sole when used as a supplement to the traditional diet based on rotifers and Artemia nauplii

    A developmental hepatotoxicity study of dietary bisphenol A in Sparus aurata juveniles

    Previous studies in rats have indicated that a diet enriched with Bisphenol A adversely effects metabolism and reproductive success. In rats exposed to BPA by maternal gavage, alteration in the developmental programming, higher obesity rates and reproductive anomalies were induced. Starting with this evidence, the aim of this study was to provide important insights on the effects induced by a BPA enriched diet, on the reproductive physiology and metabolismof juvenile fish, simulating the scenario occurringwhenwild fish fed on prey contaminatedwith environmental BPA. Seabreamwas chosen asmodel, as it is one of the primary commercial species valued by consumers and these results could provide important findings on adverse effects that could be passed on to humans by eating contaminated fish. A novel method for measuring BPA in the food and water by affinity chromatography was developed. Analysis of signals involved in reproduction uncovered altered levels of vtg and Zp, clearly indicating the estrogenic effect of BPA. Similarly, BPA up-regulated catd and era gene expression. A noteworthy outcome from this study was the full length cloning of two vtg encoding proteins, namely vtgA and vtgB, which are differently modulated by BPA. Cyp1a1 and EROD activity were significantly downregulated, confirming the ability of estrogenic compounds to inhibit the detoxification process. GST activitywas unaffected by BPA contamination, while CAT activitywas down regulated. These results collectively confirmthe estrogenic effect of BPA and provide additional characterization of novel vtg genes in Sparus aurata
