    Induction of Ran GTP drives ciliogenesis

    The small GTPase Ran and the importin proteins regulate nucleocytoplasmic transport. New evidence suggests that Ran GTP and the importins are also involved in conveying proteins into cilia. In this study, we find that Ran GTP accumulation at the basal bodies is coordinated with the initiation of ciliogenesis. The Ran-binding protein 1 (RanBP1), which indirectly accelerates Ran GTP → Ran GDP hydrolysis and promotes the dissociation of the Ran/importin complex, also localizes to basal bodies and cilia. To confirm the crucial link between Ran GTP and ciliogenesis, we manipulated the levels of RanBP1 and determined the effects on Ran GTP and primary cilia formation. We discovered that RanBP1 knockdown results in an increased concentration of Ran GTP at basal bodies, leading to ciliogenesis. In contrast, overexpression of RanBP1 antagonizes primary cilia formation. Furthermore, we demonstrate that RanBP1 knockdown disrupts the proper localization of KIF17, a kinesin-2 motor, at the distal tips of primary cilia in Madin–Darby canine kidney cells. Our studies illuminate a new function for Ran GTP in stimulating cilia formation and reinforce the notion that Ran GTP and the importins play key roles in ciliogenesis and ciliary protein transport

    Bacterial quality of a smoked meat product (“Suya”)

    The bacteriological quality of “suya” sold within Makurdi metropolis was determined. Suya samples from four selected locations were examined bacteriologically for both Total Plate and Coliform Counts. Mean Total Plate and Coliform Counts varied from 3.7x105cfu/g to 2.4x106cfu/g and 1.9x102cfu/g to 1.0x103cfu/g of samples respectively. Faecal coliform bacteria were isolated from all suya samples with the same pattern of distribution. There was significant difference (p>0.05) of bacterial contamination among samples from the different locations using Tukey\'s LSD multiple comparison test. The Total Plate Count and Coliform Counts of most samples were within recommended safe limit for meat except the coliform count of the Wurukum (WK) sample that was above the limit of 4x102cfu/g for Escherichia coli. Isolation of faecal coliforms from all samples in this investigation should be a serious concern to consumers. This calls for urgent improvement on the hygienic handling of the product by suya processors.     Bacterial diversity analysis of Huanglongbing pathogen-infected citrus using PhyloChips and 16S rDNA gene clone library sequencing

    The bacterial diversity associated with citrus leaf midribs was characterized for citrus groves that contained the Huanglongbing (HLB) pathogen, which has yet to be cultivated in vitro. We employed a combination of high-density phylogenetic 16S rRNA gene microarrays and 16S rRNA gene clone library sequencing to determine the microbial community composition for symptomatic and asymptomatic citrus midribs. Our results revealed that citrus leaf midribs can support a diversity of microbes. PhyloChip analysis indicated that 47 orders of bacteria in 15 phyla were present in the citrus leaf midribs, while 20 orders in 8 phyla were observed with the cloning and sequencing method. PhyloChip arrays indicated that nine taxa were significantly more abundant in symptomatic midribs than in asymptomatic midribs. "Candidatus Liberibacter asiaticus" was detected at a very low level in asymptomatic plants but was over 200 times more abundant in symptomatic plants. The PhyloChip analysis results were further verified by sequencing 16S rRNA gene clone libraries, which indicated the dominance of "Candidatus Liberibacter asiaticus" in symptomatic leaves. These data implicate "Candidatus Liberibacter asiaticus" as the pathogen responsible for HLB disease

    Some Syntactic Aspects of Lexical Anaphors in Select Munda Languages

    This paper investigates several syntactic aspects of anaphors (reflexives and reciprocals) in the Munda languages Santali, Mundari, and Ho. We intend to show that verbal reflexivization is an indigenous device, and that nominal reflexivization is a form calqued from neighbouring Indo-Aryan languages. The verbal reflexive device performs other functions, including detransitivizing marker, passive marker, and self-benefactive marker. The nominal reflexive is optional when the verbal reflexive is present, and obligatory when the verbal reflexive is absent. Long-Distance Binding is not permitted when the anaphor occurs in a subcategorized position. Reciprocity is achieved through the infixation of a morpheme -pV- in the main verb, where the vowel V in -pV- harmonizes with the nucleus of the main verb's first syllable. Some verbs have a special form when the verbal anaphor occurs

    The effects of tocotrienol-rich vitamin E (Tocovid) on diabetic neuropathy:A phase II randomized controlled trial

    Chronic hyperglycemia increases oxidative stress, activates inflammatory pathways and reduces nerve growth factor (NGF) among diabetic patients, which contribute to development of diabetic peripheral neuropathy (DPN). Tocotrienol-Rich Vitamin E (Tocovid) possesses potent antioxidant and anti-inflammatory properties which are postulated to target these pathogeneses in order to ameliorate DPN. This study aims to evaluate the effects of Tocovid on nerve conduction parameters and serum biomarkers among diabetic patients. This multicenter, prospective, randomized, double-blind, placebo-controlled clinical trial was conducted on 80 eligible participants. The intervention group (n = 39) was randomly allocated to receive 200 mg of Tocovid twice a day, and the control group (n = 41) received placebo twice a day. At the end of eight weeks, the nerve conduction parameters, as assessed by nerve conduction study, as well as serum biomarkers (NGF, malondialdehyde, vascular cell adhesion molecule 1, tumor necrosis factor receptor 1 and thromboxane B2) were compared between the two groups. Compared to placebo, Tocovid significantly improves the nerve conduction velocities of all nerves (+1.25 m/s, interquartile range [IQR] 3.35, p < 0.001, median nerve; +1.60 m/s, IQR 1.80, p < 0.001, sural nerve; +0.75 m/s, IQR 2.25, p < 0.001, tibial nerve). Meanwhile, the levels of serum NGF were significantly higher in the Tocovid group as compared to placebo at eight weeks post-intervention. Participants receiving Tocovid illustrated highly significant improvement in terms of nerve conduction velocities for all nerves tested after eight weeks of supplementation. In addition, Tocovid supplementation elevated the levels of serum NGF, in which its increase is postulated to reflect enhanced neuronal functions. This novel finding suggests that Tocovid could be a disease-modifying agent targeting serum NGF to improve nerve conduction velocities
