    In Vivo Consumption of Cranberry Exerts ex Vivo Antiadhesive Activity against <i>FimH</i>-Dominated Uropathogenic Escherichia coli: A Combined in Vivo, ex Vivo, and in Vitro Study of an Extract from Vaccinium macrocarpon

    For investigation of the molecular interaction of cranberry extract with adhesins of uropathogenic Escherichia coli (UPEC), urine from four volunteers consuming standardized cranberry extract (proanthocyanidin content = 1.24%) was analyzed within ex vivo experiments, indicating time-dependent significant inhibition of 40–50% of bacterial adhesion of UPEC strain NU14 to human T24 bladder cells. Under in vitro conditions a dose-dependent increase in bacterial adhesion was observed with proanthocyanidin-enriched cranberry Vaccinium macrocarpon extract (proanthocyanidin content = 21%). Confocal laser scanning microscopy and scanning electron microscopy proved that <i>V.m.</i> extract led to the formation of bacterial clusters on the outer plasma membrane of the host cells without subsequent internalization. This agglomerating activity was not observed when a PAC-depleted extract (<i>V.m.</i> extract<sup>≠PAC</sup>) was used, which showed significant inhibition of bacterial adhesion in cases where type 1 fimbriae dominated and mannose-sensitive UPEC strain NU14 was used. <i>V.m.</i> extract<sup>≠PAC</sup> had no inhibitory activity against P- and F1C-fimbriae dominated strain 2980. Quantitative gene expression analysis indicated that PAC-containing as well as PAC-depleted cranberry extracts increased the <i>fimH</i> expression in NU14 as part of a feedback mechanism after blocking <i>FimH</i>. For strain 2980 the PAC-containing extract led to up-regulation of P- and F1C-fimbriae, whereas the PAC-depleted extract had no influence on gene expression. <i>V.m.</i> and <i>V.m.</i> extract<sup>≠PAC</sup> did not influence biofilm and curli formation in UPEC strains NU14 and 2980. These data lead to the conclusion that also proanthocyanidin-free cranberry extracts exert antiadhesive activity by interaction with mannose-sensitive type 1 fimbriae of UPEC

Zadání pro bakalářskou práci definovala firma FULLCOM systems s.r.o., která se zabývá vývojem ERP systémů zejména pro potravinářský průmysl s cílem analyzovat a vytvořit rozšíření funkcionality jejich ERP systému FIS.     Import 05/08/2014Tato bakalářská práce se zabývá analýzou možností tvorby efektivní webové prezentace za použití redakčního systému. První část práce se věnuje firmě. Je zde nastíněn popis a organizace firmy a její informační toky. Dále jsou jmenovány požadavky na funkce redakčního systému. Druhá část práce se zabývá redakčními systémy. V teoretické části jsou vysvětleny základní funkce a principy redakčních systémů. Praktická část porovnává, nakolik funkce jednotlivých vybraných redakčních systémů splňují požadavky firmy. Nakonec je na základě zjištěných poznatků navrženo vhodné typové řešení pro danou firmu.This bachelor thesis is dedicated to analysis how to create an effective web presentation by means of content management system. The first part of the thesis is dedicated to a company. It contains description and organization of a company and its information flows. Furthermore, requirements for content management functions are stated. The second part of the thesis is dedicated to content management systems. In the theoretical part, basic functions and principles of content management systems are explained. In the practical part, it is compared to which extend functions of selected content management systems fulfill requirements of a company.     Import 05/08/2014Tato diplomová práce se zabývá vyhotovením geometrických plánů v České a Slovenské republice a jejich vzájemným porovnáním. Porovnává vyhotovení geometrických plánů z pohledu jejich tvorby, právních předpisů, formálních náležitostí, obsahu a dalších důležitých kritérií. Aby bylo možné geometrické plány vytvořit a porovnat, bylo nutné důkladně se seznámit s legislativou v obou zemích. V diplomové práci byly použity citace ze zákonů, vyhlášek a metodických pokynů jednotlivých zemí a příslušných katastrech nemovitostí. Práce se zároveň v jejím úvodu zabývá historickým vývojem katastru nemovitostí na územích České a Slovenské republiky obecně, jakož i konkrétní vývojem samotných geometrických plánů. Podrobně popisuje a porovnává jejich grafické a písemné části a zbylou dokumentaci. Obsahuje informace o aktuálních legislativních předpisech, které upravují tvorbu a obsah geometrických plánů na Slovensku a v České republice.This Master Thesis delves into the complex process of geometrical survey and map production in Slovakia and in the Czech Republic and compares geometrical survey processes in the two countries. This work assesses the processes involved in geometrical surveys in Slovakia and in the Czech Republic and compares them based on crucial criteria including survey concept and creation, legal requirements and other formalities. In order to realize such a comprehensive Master Thesis, it was necessary to familiarize oneself fully with the researched topic through extensive study of legislature, Land Registry guidelines and regulations and methodical directives of both countries in question. In its Intro, this Master Thesis elaborates on the history of Land Registries in Slovakia and in the Czech Republic, as well as on the history of geometrical maps in both countries. The author examines the graphic and the written part of geometrical maps in great detail and cross-examines further supporting documentation.     <p><i>(A)</i> Whole mount urogenital tracts of uninfected wild-type or CEAtg female mice were fixed and processed for scanning electron microscopy (SEM).
