    Finite element analysis of an old building façade and reinforcement

    The Podestà Palace, a historical masonry construction, is dated from the XII century and is situated in Foligno, central Italy. This building has a high heritage and artistic value associated to it, mainly due to existing paintings in both the internal and external parts of the façade. Its current deteriorated state, coupled with the high seismic hazard of the region, stresses the pronounced seismic risk associated to this building. However, possible conservation and strengthening measures to be potentially applied should not affect the architectural and historical value of the Palace, i.e. its authenticity. In order to assess the seismic vulnerability of the building, a numerical model was developed. The reduced global dimensions of the building made possible the development of a simple model comprising only the consideration of the façade. The boundary conditions, due to effects of the walls perpendicular to the façade, pavements and vaults as well as the effect of the neighbouring building and later additions were duly considered. A previous and detailed structural survey of the building, together with existing architectural drawings, made possible the development and construction of a finite element mesh of the Palace’s façade. The numerical model was subjected to the self-weight of the structure and to lateral loads that took into account earthquake loads according to the Italian code. The results demonstrate that during a standard earthquake, partial or total collapse of the building should be expected. The careful analysis of the critical areas that lead the structure to collapse made possible to determine an appropriate methodology for structural strengthening, having always in mind the minimum intervention - maximum efficiency approach. Through a careful analysis of the weakened areas that caused the building collapse, it became possible to determine the best methodology for the structural strengthening, being chosen the reinforcement with carbon fibers, that allowed a localized reinforcement. In the last part of the study, the model was updated with the inclusion of the FRP strengthening and subjected to the same loads applied on the second part of the study. The results show that, due to the use of FRP composites, the collapse was avoided through the reduction of the displacements normal to the façade surface. As a conclusion, the carbon fibres located in strategic areas, increased the building façade resistance by reducing the perpendicular displacements. The strategic location of the carbon fibre becomes imperceptible, and this way, less intrusive to the patrimonial value of the construction

    Neste estudo foi efectuada uma análise da fachada do palácio Podestà, situado em Foligno, Itália, através de um modelo não linear de elementos finitos. Com base na geometria e no modelo mecânico, foi desenvolvida a análise da estrutura em três fases. Numa primeira fase, foi efectuada uma análise da estrutura para as cargas permanentes do edifício (análise estática com o peso próprio da estrutura e de elementos não estruturais, considerando os impulsos transmitidos pelas abóbadas). Numa segunda fase foi avaliado o comportamento da estrutura sujeita às cargas permanentes combinadas com a acção sísmica. Finalmente, foi avaliada numericamente a eficiência de uma solução de reforço baseada na aplicação de fibras de carbono. São comparados os resultados de cada análise tendo em consideração os deslocamentos e as tensões na fachada. Estas modulações permitiram compreender os mecanismos potenciais de dano e de colapso da fachada do edifício para cada análise efectuada, bem como estudar a eficiência de uma solução de reforço

    RSSsite: a reference database and prediction tool for the identification of cryptic Recombination Signal Sequences in human and murine genomes

    Recombination signal sequences (RSSs) flanking V, D and J gene segments are recognized and cut by the VDJ recombinase during development of B and T lymphocytes. All RSSs are composed of seven conserved nucleotides, followed by a spacer (containing either 12 ± 1 or 23 ± 1 poorly conserved nucleotides) and a conserved nonamer. Errors in V(D)J recombination, including cleavage of cryptic RSS outside the immunoglobulin and T cell receptor loci, are associated with oncogenic translocations observed in some lymphoid malignancies. We present in this paper the RSSsite web server, which is available from the address http://www.itb.cnr.it/rss. RSSsite consists of a web-accessible database, RSSdb, for the identification of pre-computed potential RSSs, and of the related search tool, DnaGrab, which allows the scoring of potential RSSs in user-supplied sequences. This latter algorithm makes use of probability models, which can be recasted to Bayesian network, taking into account correlations between groups of positions of a sequence, developed starting from specific reference sets of RSSs.     The RING domain of RAG1 ubiquitylates histone H3: a novel activity in chromatin-mediated regulation of V(D)J joining.

    The RAG1 and RAG2 proteins are the only lymphoid-specific factors required to perform the first step of V(D)J recombination, DNA cleavage. While the catalytic domain of RAG1, the core region, has been well characterized, the role of the noncore region in modulating chromosomal V(D)J recombination efficiency remains ill defined. Recent studies have highlighted the role of chromatin structure in regulation of V(D)J recombination. Here we show that RAG1 itself, through a RING domain within its N-terminal noncore region, preferentially interacts directly with and promotes monoubiquitylation of histone H3. Mutations affecting the RAG1 RING domain reduce histone H3 monoubiquitylation activity, decrease V(D)J recombination activity in vivo, reduce formation of both signal-joint and coding-joint products on episomal substrates, and decrease efficiency of V(D)J recombination at the endogenous IgH locus in lymphoid cells. The results reveal that RAG1-mediated histone monoubiquitylation activity plays a role in regulating the joining phase of chromosomal V(D)J recombination

    Clinical impact of subclonal EGFR T790M mutations in advanced-stage EGFR-mutant non-small-cell lung cancers.

    Advanced non-small-cell lung cancer (NSCLC) patients with EGFR T790M-positive tumours benefit from osimertinib, an epidermal growth factor receptor-tyrosine kinase inhibitor (EGFR-TKI). Here we show that the size of the EGFR T790M-positive clone impacts response to osimertinib. T790M subclonality, as assessed by a retrospective NGS analysis of 289 baseline plasma ctDNA samples from T790M-positive advanced NSCLC patients from the AURA3 phase III trial, is associated with shorter progression-free survival (PFS), both in the osimertinib and the chemotherapy-treated patients. Both baseline and longitudinal ctDNA profiling indicate that the T790M subclonal tumours are enriched for PIK3CA alterations, which we demonstrate to confer resistance to osimertinib in vitro that can be partially reversed by PI3K pathway inhibitors. Overall, our results elucidate the impact of tumour heterogeneity on response to osimertinib in advanced stage NSCLC patients and could help define appropriate combination therapies in these patients

    DETERMINANTS OF 6-MONTH MORTALITY IN SURVIVORS OF MYOCARDIAL-INFARCTION AFTER THROMBOLYSIS - RESULTS OF THE GISSI-2 DATA-BASE

    none647Background. Current knowledge of risk assessment in survivors of myocardial infarction is largely based on data gathered before the advent of thrombolysis. It must be determined whether and to what extent available information and proposed criteria of prognostication are applicable in the thrombolytic era. Methods and Results. We reassessed risk prediction in the 10 219 survivors of myocardial infarction with follow-up data available (ie, 98% of the total) who had been enrolled in the GISSI-2 trial, relying on a set of prespecified variables. The 3.5% 6-month all-cause mortality rate of these patients compared with the higher value of 4.6% found in the corresponding GISSI-1 cohort, originally allocated to streptokinase therapy, indicates a 24% reduction in postdischarge 6-month mortality. On multivariate analysis (Cox model), the following variables were predictors of 6-month all-cause mortality: ineligibility for exercise test for both cardiac (relative risk [RR], 3.30; 95% confidence interval [CI], 2.36-4.62) and noncardiac reasons (RR, 3.28; 95% CI, 2.23-4.72), early left ventricular failure (RR, 2.41; 95% Cl, 1.87-3.09), echocardiographic evidence of recovery phase left ventricular dysfunction (RR, 2.30; 95% CI, 1.78-2.98), advanced (more than 70 years) age (RR, 1.81; 95% Cl, 1.43 -2.30), electrical instability (ie, frequent and/or complex ventricular arrhythmias) (RR, 1.70; 95% Cl, 1.32-2.19), late left ventricular failure (RR, 1.54; 95% Cl, 1.17-2.03), previous myocardial infarction (RR, 1.47; 95% CI, 1.14-1.89), and a history of treated hypertension (RR, 1.32; 95% Cl, 1.05-1.65). Early post-myocardial infarction angina, a positive exercise test, female sex, history of angina, history of insulin-dependent diabetes, and anterior site of myocardial infarction were not risk predictors. On further multivariate analysis, performed on 8315 patients with the echocardiographic indicator of left ventricular dysfunction available, only previous myocardial infarction was not retained as an independent risk predictor. Conclusions. A decline in 6-month mortality of myocardial infarction survivors, seen within 6 hours of symptom onset, has been observed in recent years. Ineligibility for exercise test, early left ventricular failure, and recovery-phase left ventricular dysfunction are the most powerful (RR, >2) predictors of 6-month mortality among patients recovering from myocardial infarction after thrombolysis. Qualitative variables reflecting residual myocardial ischemia do not appear to be risk predictors. The lack of an independent adverse influence of early post-myocardial infarction angina on 6-month survival represents a major difference between this study and those of the prethrombolytic era.noneVOLPI A;
