    A conjugate gradient algorithm for the astrometric core solution of Gaia

    The ESA space astrometry mission Gaia, planned to be launched in 2013, has been designed to make angular measurements on a global scale with micro-arcsecond accuracy. A key component of the data processing for Gaia is the astrometric core solution, which must implement an efficient and accurate numerical algorithm to solve the resulting, extremely large least-squares problem. The Astrometric Global Iterative Solution (AGIS) is a framework that allows to implement a range of different iterative solution schemes suitable for a scanning astrometric satellite. In order to find a computationally efficient and numerically accurate iteration scheme for the astrometric solution, compatible with the AGIS framework, we study an adaptation of the classical conjugate gradient (CG) algorithm, and compare it to the so-called simple iteration (SI) scheme that was previously known to converge for this problem, although very slowly. The different schemes are implemented within a software test bed for AGIS known as AGISLab, which allows to define, simulate and study scaled astrometric core solutions. After successful testing in AGISLab, the CG scheme has been implemented also in AGIS. The two algorithms CG and SI eventually converge to identical solutions, to within the numerical noise (of the order of 0.00001 micro-arcsec). These solutions are independent of the starting values (initial star catalogue), and we conclude that they are equivalent to a rigorous least-squares estimation of the astrometric parameters. The CG scheme converges up to a factor four faster than SI in the tested cases, and in particular spatially correlated truncation errors are much more efficiently damped out with the CG scheme.Comment: 24 pages, 16 figures.     Quantification of cytosolic plasmid DNA degradation using high‐throughput sequencing: implications for gene delivery

    Background Although cytosolic DNA degradation plays an important role in decreasing transgene expression, the plasmid degradation pattern remains largely unexplored. Methods Illumina dye sequencing was employed to provide degradation site information for S1 and cytosolic nucleases. S1 nuclease provided a positive control for a comparison between the agarose gel method and sequencing approaches. Results The poly(A) region between the β‐lactamase gene and the cytomegalovirus (CMV) promoter was identified as the most likely cut site for polyplex‐treated cytosol. The second most likely site, at the 5' end of the β‐lactamase gene, was identified by gel electrophoresis and sequencing. Additional sites were detected in the OriC region, the SV40/poly(A) region, the luciferase gene and the CMV promoter. Sequence analysis of plasmid treated with cytosol from control cells showed the greatest cut activity in the OriC region, the β‐lactamase gene and the poly(A) region following the luciferase gene. Additional regions of cut activity include the SV40 promoter and the β‐lactamase poly(A) termination sequence. Both cytosolic nucleases and the S1 nuclease showed substantial activity at the bacterial origin of replication ( OriC ). Conclusions High‐throughput plasmid sequencing revealed regions of the luciferase plasmid DNA sequence that are sensitive to cytosolic nuclease degradation. This provides new targets for improving plasmid and/or polymer design to optimize the likelihood of protein expression.     A multi-instrument comparison of integrated water vapour measurements at a high latitude site

    We compare measurements of integrated water vapour (IWV) over a subarctic site (Kiruna, Northern Sweden) from five different sensors and retrieval methods: Radiosondes, Global Positioning System (GPS), ground-based Fourier-transform infrared (FTIR) spectrometer, groundbased microwave radiometer, and satellite-based microwave radiometer (AMSU-B). Additionally, we compare also to ERA-Interim model reanalysis data. GPS-based IWV data have the highest temporal coverage and resolution and are chosen as reference data set. All datasets agree reasonably well, but the ground-based microwave instrument only if the data are cloud-filtered. We also address two issues that are general for such intercomparison studies, the impact of different lower altitude limits for the IWV integration, and the impact of representativeness error. We develop methods for correcting for the former, and estimating the random error contribution of the latter. A literature survey reveals that reported systematic differences between different techniques are study-dependent and show no overall consistent pattern. Further improving the absolute accuracy of IWV measurements and providing climate-quality time series therefore remain challenging problems

    Low-mass e+e- pair production in 158 A GeV Pb-Au collisions at the CERN SPS, its dependence on multiplicity and transverse momentum

    We report a measurement of low-mass electron pairs observed in 158 GeV/nucleon Pb-Au collisions. The pair yield integrated over the range of invariant masses 0.2 < m < 2.0 GeV is enhanced by a factor of 3.5 +/- 0.4 (stat) +/- 0.9 (syst) over the expectation from neutral meson decays. As observed previously in S-Au collisions, the enhancement is most pronounced in the invariant-mass region 300-700 MeV. For Pb-Au we find evidence for a strong increase of the enhancement with centrality. In addition, we show that the enhancement covers a wide range in transverse momentum, but is largest at the lowest observed pt.Comment: 17 pages, 4 figures, submitted to Phys.Lett.

    Abstract In order to achieve the high brightness promised by the single-pass Free Electron Laser (FEL) at the TESLA Test Facility (TTF) at DESY, the electron beam position must be controlled better than 10 m over a distance of 15 m. The design of a monitor system with this accuracy is described. The silicon pixel detector, which is critical to this monitor, must have a position resolution of 1 m and an excellent sensitivity for soft X-rays above 150 eV. The silicon pixel detector and its readout electronics are also described

    Dual fluorescence of 9-anthryl-substituted oligothiophenes in nonpolar environment

    9-Anthryl-oligothiophenes (9A-Tn, n = 1–4) have been studied in n-hexane solution between helium and room temperature using absorption, fluorescence emission and excitation spectra and time resolved fluorescence measurements. These compounds are reference systems for studies on intramolecular energy and charge transfer in donor/acceptor-substituted conjugated chain molecules. The absorption spectra show contribution of both substituents anthracene and oligothiophene, but no additional bands due to mixed electronic states. Dual fluorescence is observed at Tgreater-or-equal, slanted 120 K for the compounds 9A-Tn (n = 1–3), but not for 9A-T4. Time resolved spectroscopy reveals a dynamical coupling between the two emission components whose relative quantum yields are strongly temperature dependent. The occurrence of the dual fluorescence is explained by an intramolecular torsional motion between the two molecular subunits

    Recent results from Pb-Au collisions at 158 GeV/c per nucleon obtained with the CERES spectrometer

    During the 1996 lead run time, CERES has accumulated 42 million events, corresponding to a factor of 5 more statistics than in 1995 and 2.5 million events of a special photon-run. We report on the results of the low-mass e+^+e^--pair analysis. Since the most critical item is the poor signal-to-background ratio we also discuss the understanding of this background, in absolute terms, with the help of a detailed Monte Carlo simulation.     e+e--pair production in Pb-Au collisions at 158 GeV per nucleon

    We present the combined results on electron-pair production in 158 GeV/n {Pb-Au} (s\sqrt{s}= 17.2 GeV) collisions taken at the CERN SPS in 1995 and 1996, and give a detailed account of the data analysis. The enhancement over the reference of neutral meson decays amounts to a factor of 2.31±0.19(stat.)±0.55(syst.)±0.69(decays)\pm0.19 (stat.)\pm0.55 (syst.)\pm0.69 (decays) for semi-central collisions (28% σ/σgeo\sigma/\sigma_{geo}) when yields are integrated over m>m> 200 MeV/c2c^2 in invariant mass. The measured yield, its stronger-than-linear scaling with NchN_{ch}, and the dominance of low pair ptp_t strongly suggest an interpretation as {\it thermal radiation} from pion annihilation in the hadronic fireball. The shape of the excess centring at mm\approx 500 MeV/c2c^2, however, cannot be described without strong medium modifications of the ρ\rho meson. The results are put into perspective by comparison to predictions from Brown-Rho scaling governed by chiral symmetry restoration, and from the spectral-function many-body treatment in which the approach to the phase boundary is less explicit.Comment: 39 pages, 40 figures, to appear in Eur.Phys.J.C.     Responses of the putative trachoma vector, Musca sorbens, to volatile semiochemicals from human faeces

    The putative vector of trachoma, Musca sorbens, prefers to lay its eggs on human faeces on the ground. This study sought to determine whether M. sorbens females were attracted to volatile odours from human faeces in preference to odours from the faeces of other animals, and to determine whether specific volatile semiochemicals mediate selection of the faeces. Traps baited with the faeces of humans and local domestic animals were used to catch flies at two trachoma-endemic locations in The Gambia and one in Ethiopia. At all locations, traps baited with faeces caught more female M. sorbens than control traps baited with soil, and human faeces was the most successful bait compared with soil (mean rate ratios 44.40, 61.40, 10.50 [P<0.001]; 8.17 for child faeces [P = 0.004]). Odours from human faeces were sampled by air entrainment, then extracts of the volatiles were tested by coupled gas chromatography-electroantennography with laboratory-reared female M. sorbens. Twelve compounds were electrophysiologically active and tentatively identified by coupled mass spectrometry-gas chromatography, these included cresol, indole, 2-methylpropanoic acid, butanoic acid, pentanoic acid and hexanoic acid. It is possible that some of these volatiles govern the strong attraction of M. sorbens flies to human faeces. If so, a synthetic blend of these chemicals, at the correct ratios, may prove to be a highly attractive lure. This could be used in odour-baited traps for monitoring or control of this species in trachoma-endemic regions
