    Validation and psychometric properties of the Polish version of the Skin Picking Scale-Revised

    The Skin Picking Scale-Revised (SPS-R) is an 8-item self-report measure of skin picking behaviors. It includes two subscales related to skin picking symptom severity and picking-related impairments. The study aimed to assess the psychometric properties of the Polish version of the SPS-R in a sample of adults reporting skin picking. The sample of 764 participants was recruited from the general population through an online survey. Among them, 159 meet the criteria of pathological skin picking applied in the original SPS-R validation study, and 57 endorsed all of the DSM-5 criteria for excoriation disorder. The SPS-R was back-translated into Polish. Factor structure, reliability, convergent and divergent validity, and diagnostic accuracy were assessed. Confirmatory factor analyses revealed a two-factor structure of the scale. High internal consistency and convergent and divergent validity were confirmed for the total score as well as for the subscales. High prognostic ability of the SPS-R total score was also demonstrated using ROC analysis: ≥5 was accepted as an optimal cut-off point for distinguishing skin picking sufferers from healthy controls. The Polish version of the SPS-R shows good psychometric properties and appears to be a reliable measure of skin picking symptoms and picking-related impairment

    Cytotoxic and antioxidant activity of hypericum perforatum L. extracts against human melanoma cells from different stages of cancer progression, cultured under normoxia and hypoxia

    Oxidative stress and the hypoxic microenvironment play a key role in the progression of human melanoma, one of the most aggressive skin cancers. The aim of our study was to evaluate the effect of Hypericum perforatum extracts of different origins (both commercially available (HpEx2) and laboratory-prepared from wild grown (HpEx12) and in vitro cultured (HpEx13) plants) and hyperforin salt on WM115 primary and WM266-4 lymph node metastatic human melanoma cells cultured under normoxic and hypoxic conditions. The polyphenol content, radical scavenging activity, and hyperforin concentration were determined in the extracts, while cell viability, apoptosis, ROS production, and expression of NRF2 and HO-1, important oxidative stress-related factors, were analyzed after 24 h of cell stimulation with HpExs and hyperforin salt. We found that cytotoxic, pro-apoptotic and antioxidant effects depend on the extract composition, the stage of melanoma progression, and the oxygen level. Hyperforin salt showed lower activity than H. perforatum extracts. Our study for the first time showed that the anticancer activity of H. perforatum extracts differs in normoxia and hypoxia. Importantly, the composition of extracts of various origins, including in vitro cultured, resulting in their unique properties, may be important in the selection of plants for therapeutic application

    Burnout, neurotic symptoms and coping strategies in medical students

Medical studies are considered one of the most stressful majors, leading to early burnout and other related symptoms such as neurotic symptoms. Our aim was to examine this topic by assessing burnout and neurotic symptoms as well as strategies of coping with stress experienced during each year of studies. Method: We used a web-based questionnaire, consisting of the Maslach Burnout Inventory-Student Survey (MBI-SS), Coping Inventory for Stressful Situations (CISS) and Symptom Checklist S-III, and invited medical students at various stages of a 6-year medical course to fill it in online. Questionnaire was filled by 781 students in total. Results: Statistical analysis revealed an interesting pattern of symptoms severity in students, with highest scores at the beginning and at the end of the medical course and the lowest score during the 3rd year of studies. This pattern was clearly visible for MBI-SS Exhaustion, and somewhat less pronounced for MBI-SS Cynicism and S-III scores, where only the decrease of symptoms was significant. Coping strategies seemed to be similar for all medical students with a higher score for the Distraction scale among the 3rd - year students compared with the 2nd-year students. Discussion: These results, however unexpected, seem to be consistent with available literature, emphasizing higher levels of stress experienced during great changes regarding expectations in students at the beginning of their course and in soon-to-be doctors. Conclusions: The results prompt to reflect on ways of countering emerging symptoms of burnout not only in experienced students, but also among those starting medical college

    Clinical presentation of simple and combined or syndromic arteriovenous malformations.

    OBJECTIVES Arteriovenous malformations of the lower extremities (AVMLE) can present as simple or complex combined or syndromic forms (e.g. Parkes Weber Syndrome). We aimed to characterize the differences in clinical presentation and natural history of these potentially life and limb threatening congenital vascular malformations. METHODS We conducted a retrospective analysis of a consecutive series of patients with AVMLE, who presented to a tertiary referral center in Switzerland between 2008 and 2018. Clinical baseline characteristics, D-dimer level and course were summarized and differences between simple, non-syndromic and combined or syndromic AVMLE determined. Odds ratios (OR) and 95% confidence intervals (CI) were estimated using logistic regression models. RESULTS Overall, 506 patients were prospectively enrolled in the Bernese Congenital Vascular Malformation Registry, 31 (6%) with AVMLE. There were 16 women and 15 men with a mean age of 18 years at first diagnosis (1 month - 72 years). Simple AVMLE was present in 22 (71%), combined or syndromic AVMLE with limb overgrowth in 9 patients (29%), respectively. Common symptoms and signs were pain 25 (81%), swelling 21 (68%) and soft tissue hypertrophy 13 (42%). Among combined or syndromic patients, 3 patients died from wound infection with sepsis or disseminated intravascular coagulation with bleeding complications (intracranial hemorrhage and bleeding from extensive leg ulcers). Combined or syndromic patients presented more often with bleeding (67% vs. 5%; p<0.001), malformation related infection (44% vs. 5%; p=0,017) and leg length difference (56% vs. 14%; p=0.049). D-dimer levels were elevated (mean 17256 μg/L, range 1557 μg/L to 80000 μg/L) and angiographic appearance showed complex, mixed type of AVMs, including interstitial type IV, in all patients with combined or syndromic AVMLE. CONCLUSION Patients with congenital simple AVMLE most often present with benign clinical features and rarely complications related to hemodynamic changes. Patients with combined or syndromic AVMLE often face serious outcomesdominated by complications other than direct high flow related heart failure

    Parkes Weber Syndrome: Contribution of the Genotype to the Diagnosis

    Objectives: Parkes Weber syndrome (PWS) is a rare disorder that combines overgrowth, capillary malformations, and arteriovenous malformations (AVM)/arteriovenous fistulas, for which underlying activating mutations in the ras/mitogen-activated protein kinase/extracellular-signal-regulated kinase signaling pathway have been described. The clinical overlap with Klippel-Trenauny syndrome, associated with mutations in PIK3CA, is significant. This case series aimed to elaborate on the phenotypic description of PWS, to underline its clinical overlap with Klippel-Trenauny syndrome and nonsyndromic AVM, and to evaluate the contribution of genotypic characterization to the diagnosis. Methods: All patients diagnosed with PWS upon enrollment in the Bernese VAScular COngenital Malformations (VASCOM) cohort were included. The diagnostic criteria of PWS were retrospectively reviewed. A next-generation sequencing (NGS) gene panel (TSO500, Illumina) was used on tissue biopsy samples. Results: Overall, 10/559 patients of the VAScular COngenital Malformations cohort were initially diagnosed with PWS. Three patients were reclassified as nonsyndromic AVM (Kristen Rat Sarcoma Viral oncogene homolog [KRAS], KRAS+tumor protein p53, and protein tyrosine phosphatase non-receptor type 11). Finally, 7 patients fulfilled all clinical diagnostic criteria of PWS. Genetic testing was available in 5 PWS patients. Only 1 patient had the classic RASA1 mutation; another patient had mutations in G protein subunit alpha q (GNAQ) and phosphatase and tensin homolog. In a third case, a PIK3CA mutation was detected. In 2 patients, no mutations were identified. Conclusion: Overgrowth syndromes with vascular malformations are rare and their clinical overlap hampers the classification of individual phenotypes under specific syndrome labels, sometimes even despite genetic testing. To provide optimal patient care, an accurate phenotypic description combined with the identification of molecular targets for precision medicine may be more meaningful than the syndrome classification itself

    Synergids and filiform apparatus in the sexual and apomictic dandelions from section Palustria (Taraxacum, Asteraceae)

    An evolutionary trend to reduce “unnecessary costs” associated with the sexual reproduction of their amphimictic ancestors, which may result in greater reproductive success, has been observed among the obligatory apomicts. However, in the case of the female gametophyte, knowledge about this trend in apomicts is not sufficient because most of the ultrastructural studies of the female gametophyte have dealt with amphimictic angiosperms. In this paper, we tested the hypothesis that, in contrast to amphimictic plants, synergids in apomictic embryo sacs do not form a filiform apparatus. We compared the synergid structure in two dandelions from sect. Palustria: the amphimictic diploid Taraxacum tenuifolium and the apomictic tetraploid, male-sterile Taraxacum brandenburgicum. Synergids in both species possessed a filiform apparatus. In T. brandenburgicum, both synergids persisted for a long time without any degeneration, in spite of the presence of an embryo and endosperm. We propose that the persistent synergids in apomicts may play a role in the transport of nutrients to the embryo

    W ostatnich latach gry wideo zyskały znaczącą popularność jako forma rozrywki. Celem tego badania było sprawdzenie, czy preferowane przez graczy style radzenia sobie ze stresem mogą zapośredniczać związek między cechami osobowości a objawami uzależnienia od gier. 159 uczestników (69% mężczyzn, 29% kobiet) w wieku 18 do 63 lat (M = 23,01; SD = 5,92) wypełniło ankietę online składającą się z kwestionariusza IPIP-BFM-50 do pomiaru cech osobowości, Mini-COPE w celu oceny preferowanych strategii radzenia sobie, oraz POGQ do oszacowania objawów związanych z objawami uzależnienia od gier komputerowych. Przeprowadzono analizę korelacji oraz 4 niezależne analizy mediacji. Ekstrawersja była dodatnio związana z radzeniem sobie skoncentrowanym na problemie i emocjach. Stabilność emocjonalna dodatnio korelowała z radzeniem sobie skoncentrowanym na problemie a ujemnie z radzeniem sobie przez unikanie. Niestety żadne z modeli mediacji nie okazały się istotne. Kolejne badania dotyczące uwarunkowań uzależnienia od gier wideo, zamiast koncentrować się na trzech szerokich kategoriach stylów radzenia sobie ze stresem powinny raczej uwzględnić bardziej specyficzne strategie.In recent years, video games have gained significant prominence and popularity as a form of entertainment. The aim of this study was to examine whether gamers’ preferred coping styles might mediate the relationship between their personality trats and problematic gaming symptoms. 159 participants (69% male, 29% female), aged 18 to 63 (M = 23,01; SD = 5,92) completed the survey consisting of IPIP-BFM-50 to measure their Big Five Personality Traits, Brief-COPE to assess their preferred coping strategies and POGQ to estimate the Internet Gaming Disorder symptoms. Correlation analysis and 4 independent mediation analyses were undertaken. Extraversion was positively and significantly correlated with problem-focused and emotion-focused coping styles. Emotional stability correlated positively with problem-focused coping and negatively with avoidant coping. Unfortunately, none of the hypothesized mediational models were found to be significant. Further studies, instead of focusing on three overarching categories of coping should rather incorporate more specific coping strategies
