    In-Out Intermittency in Gap Junction-Coupled Class I^* Neurons

    In a series of papers, we have proposed a dynamical model for gap junction-coupled networks of class I^* neurons, and investigated its dynamic characters. We found various dynamic states in a model neural network with diffusively coupled class I¤ neuron models, called μ-models. Among others, hierarchies of intermittent transitions attracted attention in relation with real brain dynamics. This paper is devoted to report a mechanism of the first transition appeared in the intermittenly transitory dynamics among an all-synchronized state, various metachronal waves and a weakly chaotic state. We clarify that this intermittent transition is described as an in-out intermittency

    Hayabusa2's Superior Solar Conjunction Phase Trajectory Design, Guidance and Navigation

    Hayabusa2 is the ongoing JAXA’s sample and return mission to the asteroid Ryugu. In late 2018, Ryugu was in superior solar conjunction with the Earth. It is the first time that a spacecraft experiences the blackouts in the communication link with the Earth while hovering around a small celestial body. In this article, the design of the nominal conjunction trajectory flown by the Hayabusa2’s spacecraft is presented. The requirements for the conjunction trajectory were (1) to guarantee a low fuel consumption, (2) to ensure the visibility of the asteroid by the spacecraft’s wide angle camera (60∘ FoV), and (3) to increase the spacecraft altitude to a safety location (∼109 km) from the nominal BOX-A operation of 20 km (Home Position - HP). Finally, (4) to return at BOX-A after the conjunction phase. Given the mission constraints, the designed conjunction trajectory appears to have a fish-shape in the Hill coordinates therefore we renamed it as “ayu” (sweetfish in Japanese) trajectory. The optNEAR tool was developed for the guidance (ΔVs planning) and navigation design of the Hayabusa2’s conjunction mission phase. A preliminary sensitivity analysis in the Hill reference frame proved that the ayu trajectory is a good candidate for the conjunction operation of hovering satellite. The solution in the Hill coordinates is refined in the full-body planetary dynamics (optNEAR Tool) before flight. The ayu conjunction trajectory requires (a) two deterministic ΔVs at the Conjunction Orbit Insertion (COI) point and at the Home-position Recovery Maneuver (HRM) point respectively. (b) Two stochastic ΔVs, known as Trajectory Correction Manoeuvres (TCMs), before and after the deep conjunction phase are also required. The constraint linear covariance analysis in the full-body dynamics is here derived and used for the preliminary guidance and navigation planning. The results of the covariance analysis were validated in a nonlinear sense with a Monte Carlo approach which proved the validity of the semi-analytic method for the stochastic ΔVs planning derived in this paper

    Aflatoxins in Rice Artificially Contaminated with Aflatoxin-producing Aspergillus flavus under Natural Storage in Japan

    Aflatoxin (AFT) contamination is frequent in foods grown in tropical regions, including rice. Although AFTs are generally not found in temperate-region foods, global warming has affected typical temperate-region climates, potentially permitting the contamination of foods with AFT-producing Aspergillus flavus (A. flavus). Here we investigated the AFT production in rice during storage under natural climate conditions in Japan. We examined AFTs in brown rice and rough rice artificially contaminated with A. flavus for 1 year in Japan, and we subjected AFTs in white rice to the same treatment in airtight containers and examined the samples in warm and cold seasons, simulating the storage of white rice in general households. In the brown rice, AFTs increased after 2 months (March) and peaked after 9 months (October). The AFT contamination in the rough rice was minimal. After the polishing and cooking of the brown rice, AFTs were undetectable. In the white rice stored in airtight containers, AFTs increased after 1 month (August) and peaked after 2 months (September). Minimal AFTs were detected in the cold season. Thus, AFT contamination in rice may occur in temperate regions following A. flavus contamination. The storage of rice as rough rice could provide be useful for avoiding AFT contamination

    Factors affecting the choice of suicide method in Okayama: a database analysis from a forensic perspective

    The annual number of suicides in Japan increased sharply in 1998, and since that time it has consistently exceeded 30,000 per year. In this study, we analyze a database of personal and background characteristics of 824 cases (605 men, 219 women) who completed suicide in Okayama Prefecture in 2002 and 2003. The data were obtained with cooperation from the police. Using the methodologies in a previous European study as a model, we classified the suicide methods into 8 categories. To examine the generational and regional differences in the choice of methods, we stratified the sample into 4 age groups (&#60;-24, 2544, 4564, and &#62;-65) and 2 regional groups (Okayama/Kurashiki vs. other areas). Our results on gender differences in 7 of the suicide methods were mostly similar to the European data. However, our data showed a remarkably higher proportionate male-to-female mortality ratio for poisoning by other substances (ICD-10, X65-X69 codes) (1.83, 1.15-2.92). In terms of generational differences in the choice of suicide methods, the Mantel-Haenszel test of homogeneity was significant for most of the categories in our study, suggesting an impact of age on how people commit suicide. There were no remarkable regional differences in our sample. An epidemic curve for suicides via carbon monoxide poisoning using charcoal briquets revealed a trend of time clustering not observed in the other 6 means. The database constructed and used in this study contains richer information than conventional death statistics and is expected to provide helpful knowledge and insights for future epidemiological studies.</p

    Molecular basis for the dosing time-dependency of anti-allodynic effects of gabapentin in a mouse model of neuropathic pain

    <p>Abstract</p> <p>Background</p> <p>Neuropathic pain is characterized by hypersensitivity to innocuous stimuli (tactile allodynia) that is nearly always resistant to NSAIDs or even opioids. Gabapentin, a GABA analogue, was originally developed to treat epilepsy. Accumulating clinical evidence supports the effectiveness of this drug for diverse neuropathic pain. In this study, we showed that the anti-allodynic effect of gabapentin was changed by the circadian oscillation in the expression of its target molecule, the calcium channel α2δ-1 subunit.</p> <p>Results</p> <p>Mice were underwent partial sciatic nerve ligation (PSL) to create a model of neuropathic pain. The paw withdrawal threshold (PWT) in PSL mice significantly decreased and fluctuated with a period length about 24 h. The PWT in PSL mice was dose-dependently increased by intraperitoneal injection of gabapentin, but the anti-allodynic effects varied according to its dosing time. The protein levels of α2δ-1 subunit were up-regulated in the DRG of PSL mice, but the protein levels oscillated in a circadian time-dependent manner. The time-dependent oscillation of α2δ-1 subunit protein correlated with fluctuations in the maximal binding capacity of gabapentin. The anti-allodynic effect of gabapentin was attenuated at the times of the day when α2δ-1 subunit protein was abundant.</p> <p>Conclusions</p> <p>These findings suggest that the dosing time-dependent difference in the anti-allodynic effects of gabapentin is attributable to the circadian oscillation of α2δ-1 subunit expression in the DRG and indicate that the optimizing its dosing schedule helps to achieve rational pharmacotherapy for neuropathic pain.</p

    Structural basis for the dual RNA-recognition modes of human Tra2-beta RRM

    Human Transformer2-beta (hTra2-beta) is an important member of the serine/arginine-rich protein family, and contains one RNA recognition motif (RRM). It controls the alternative splicing of several pre-mRNAs, including those of the calcitonin/calcitonin gene-related peptide (CGRP), the survival motor neuron 1 (SMN1) protein and the tau protein. Accordingly, the RRM of hTra2-beta specifically binds to two types of RNA sequences [the CAA and (GAA)2 sequences]. We determined the solution structure of the hTra2-beta RRM (spanning residues Asn110–Thr201), which not only has a canonical RRM fold, but also an unusual alignment of the aromatic amino acids on the beta-sheet surface. We then solved the complex structure of the hTra2-beta RRM with the (GAA)2 sequence, and found that the AGAA tetra-nucleotide was specifically recognized through hydrogen-bond formation with several amino acids on the N- and C-terminal extensions, as well as stacking interactions mediated by the unusually aligned aromatic rings on the beta-sheet surface. Further NMR experiments revealed that the hTra2-beta RRM recognizes the CAA sequence when it is integrated in the stem-loop structure. This study indicates that the hTra2-beta RRM recognizes two types of RNA sequences in different RNA binding modes
