    Subtractive genomics approach for in silico identification and characterization of novel drug targets in Neisseria meningitides serogroup B

    Meningococcal disease is a life-threatening illness with annual incidence rates varying from 1 to 1000 per 100 000 persons in different parts of the world. Effective polysaccharide and polysaccharide-protein conjugate vaccines that offer protection against infection with meningococcal serogroups A, C, Y and W-135 have been licensed and are available worldwide. Serogroup B remains the most prevalent cause of meningococcal disease responsible for 32% of all meningococcal disease in the United States, 45 to 80% of the cases in Europe, and for the majority of cases in the rest of the world. The development of a vaccine against serogroup B poses the biggest problem due to the similarity between the B capsular polysaccharide structure and a polysialic acid containing glycopeptides that are a part of human brain tissue. Prevention of meningococcal disease will require the development of an effective vaccine to combat serogroup B, which is the cause of most meningococcal cases in developed countries. The availability of the complete sequence information of Neisseria meningitides serogroup B proteome has made it possible to carry out the in silico analysis of its genome for identification of potential vaccine and drug targets. Our study revealed 1413 proteins which are non-homologous to human genome. Screening these proteins using the Database of Essential Genes (DEG) resulted in the identification of 362 proteins as essential proteins of the bacterium. Analysis of the identified essential proteins, using the KEGG Automated Annotation Server (KAAS) housed at Kyoto Encyclopedia of Genes and Genomes (KEGG) pathways database, revealed 35 enzymes of N. Meningitides that may be used as potential drug targets, as they belongs to pathways present only in the bacterium and not present in humans. Subcelluler localization prediction of these essential proteins revealed that 9 proteins lie on the outer membrane of the pathogen which could be potential vaccine targets. Screening of the functional inhibitors against these novel targets may result in discovery of novel therapeutic compounds that can be effective against Neisseria meningitides Serogroup B

    Synthesis and characterization of carbamates derivatives from 4-amino-1,2,4-triazole

    Carbamate derivatives are proved to be an essential class of molecules because of their application as drugs and prodrugs. Their several derivatives also have been used to craft the drug-target interfaces by incorporating the carbamate moiety. Due to the huge demand in pharma-industries these have been extensively explored and synthesized. The synthesis and characterization of carbamate derivatives from 4-amino-1,2,4-triazoles and 3-(methylthio)-4-amino-1,2,4-triazoles are described. Carbamate derivatives of 4-amino-1,2,4-triazoles with phenyl chloroformate in CH3CN using K2CO3 at ca. ~80°C have been synthesized with good to excellent yield (~81 to 97%). The methodology has been subsequently extended for synthesis of carbamate derivatives of 3-(methylthio)-4-amino-1,2,4-triazoles in which bicarbamate derivatives are formed as major products (~39 to 47%) along with mono carbamate derivatives as minor products (~9 to 22%) in the presence of similar solvent and base. On investigating the effect of solvent and base, the reaction in CH3CN in presence of K2CO3 shows the best results

    Dyskeratosis Congenita- Management and Review of Complications: A Case Report

    Among the inherited bone marrow failure disorders, dyskeratosis congenita is an X-linked inherited disorder arising as a consequence of short telomere and mutations in telomere biology. Production of the altered protein dyskerin, leads to vulnerable skin, nails, and teeth which lead to higher permeability for noxious agents which can induce carcinogenesis accounting for the classical triad of skin pigmentation, nail dystrophy and oral leukoplakia. This condition is fatal and patients succumb to aplastic anemia, malignancy or immunocompromised state. We present a young male with the classic clinical triad and avascular necrosis of both femoral heads, with no evidence of hematologic anomaly or any malignancy. He was managed for osteonecrosis with uncemented total hip arthroplasty for the symptomatic left hip. Our case represents a benign form of such a fatal and rare condition, which if detected and managed early can result in improved quality of life for the patient suffering from this disorder. This patient is under our meticulous follow-up for the last 2 years in order to determine any late development of complications before being labelled as a variant of this syndrome

    Summary There is a huge prevalence of hypovitaminosis D in the Indian population. We studied the efficacy and safety of oral vitamin D supplementation in apparently healthy adult women. Monthly cholecalciferol given orally, 60,000 IU/month during summers and 120,000 IU/month during winters, safely increases 25-hydroxyvitamin D (25 (OH)D) levels to near normal levels.     Microbial Bioagents: Economic multiplication and management of fungal-nematode complex on cumin

    419-421Mass-scale multiplication was done on cheaper substrates for application of bioagents in the management of plant-parasitic nematodes and fungal pathogens. The bioagents were isolated from the local field soils. Out of the 13 isolated fungi, most of the isolates of Trichoderma spp. that were found antagonistic to Fusarium oxysporum f. sp. cumini in dual culture technique, were mass multiplied on cheaper agrowastes. Suitability of 6 substrates was screened and tea waste was found to be best followed by wheat bran and sorghum straw. Trichoderma harzianum (T5) had the ma´imum spore load per gram (SLPG) value on tea waste followed by T. hamatum (T16) on wheat bran. Three isolates of bacteria viz.
