Verknüpfung von Lehre und Forschung: Eine universitäre Tradition als didaktische Herausforderung

Die Verknüpfung von Lehre und Forschung gehört zu den konstitutiven Elementen universitärer Hochschulen. Damit ist sowohl eine Spezifität dieses Hochschultypus bezeichnet als auch gleichzeitig ein hoher Anspruch an die Professorinnen und Professoren formuliert: Diese müssen zwei Systeme zusammen bringen, die unterschiedlich funktionieren. Für die Lehre macht diese Verknüpfung nur dann Sinn, wenn Wissenschaftlichkeit und Forschungsorientierung auf unterschiedlichen hochschuldidaktischen Handlungsebenen konzeptionell berücksichtigt werden

The Plasmodium LAP complex affects crystalloid biogenesis and oocyst cell division.

Malaria parasite oocysts located on the mosquito midgut generate sporozoites by a process called sporogony. Plasmodium berghei parasites express six LCCL lectin domain adhesive-like proteins (LAPs), which operate as a complex and share a localisation in the crystalloid - an organelle found in the ookinete and young oocyst. Depletion of LAPs prevents crystalloid formation, increases oocyst growth, and blocks sporogony. Here, we describe a LAP4 mutant that has abnormal crystalloid biogenesis and produces oocysts that display reduced growth and premature sporogony. These findings provide evidence for a role of the LAP complex in regulating oocyst cell division via the crystalloid

Unterstützende Planungsinstrumente für Curricula-Reformprozesse haben verschiedene Ansprüche zu berücksichtigen: Sie sollen u.a. die Zielsetzungen klären, die verschiedenen Fachkulturen und -traditionen berücksichtigen und engagierte Innovation ermöglichen. Die Universität Zürich hat mit einem Schaubild versucht, der gemeinsamen, diskursiven Erarbeitung von Curricula Struktur und Anregung zu geben. Biogenesis of the crystalloid organelle in Plasmodium involves microtubule-dependent vesicle transport and assembly.

Malaria parasites possess unique subcellular structures and organelles. One of these is the crystalloid, a multivesicular organelle that forms during the parasite's development in vector mosquitoes. The formation and function of these organelles remain poorly understood. A family of six conserved and modular proteins named LCCL-lectin adhesive-like proteins (LAPs), which have essential roles in sporozoite transmission, localise to the crystalloids. In this study we analyse crystalloid formation using transgenic Plasmodium berghei parasites expressing GFP-tagged LAP3. We show that deletion of the LCCL domain from LAP3 causes retarded crystalloid development, while knockout of LAP3 prevents formation of the organelle. Our data reveal that the process of crystalloid formation involves active relocation of endoplasmic reticulum-derived vesicles to common assembly points via microtubule-dependent transport. Inhibition of microtubule-dependent cargo transport disrupts this process and replicates the LCCL domain deletion mutant phenotype in wildtype parasites. These findings provide the first clear insight into crystalloid biogenesis, demonstrating a fundamental role for the LAP family in this process, and identifying the crystalloid and its formation as potential targets for malaria transmission control

Berufsbezug dank Forschendem Lernen? Zur Attraktivität einer hochschuldidaktischen Losung

Forschendes Lernen ist an deutschsprachigen Hochschulen inzwischen sehr verbreitet. Der Beitrag argumentiert, dass sich dieser Aufschwung nicht zuletzt der Tatsache verdankt, dass Forschendes Lernen für verschiedene Hochschulkonzeptionen mit jeweils unterschiedlichen Begründungen und Betonungen attraktiv ist. Nicht zuletzt zeigt sich dies betreffend «Berufsbezug»: Forschendes Lernen wird als Antwort gesehen sowohl gegen eine zu enge Interpretation des Berufsbezugs ebenso wie für Forderung nach mehr beruflicher Relevanz der Studiengänge. The heretical elements in the Errores Gazariorum, a treatise written in the Savoyard Valley of Aosta in the second half of the 1430s, and containing one of the first descriptions of the new sect of male and female witches, bring us back to the Piedmont region of the 1380s. People say that a heretical «synagogue» showing almost all the features of what was to become the witches’ sabbath has taken place there. This narrative is the by-product of the «encounter» of a frustrated inquisitor and of a confused person whose mind had been broken by torture and who is at the end of an adventurous spiritual career.Ce sont les éléments hérétiques contenus dans les Errores Gazariorum (une des premières descriptions de la secte des sorciers et sorcières, écrite dans la deuxième moitié des années 1430 dans le Val d’Aoste savoyard) qui nous ramènent dans le Piémont des années 1380 où nous trouvons une « synagogue » pratiquement complète. La défense d’une cause perdue. Le rôle de l’évêque Aymon de Montfalcon dans les procès Jetzer (Berne, 1507-1509)

Cet article se penche sur le rôle joué par l’évêque Aymon de Montfalcon dans les trois procès Jetzer, menés dans les années 1507-1509 à Lausanne et à Berne et concernant des apparitions de la Vierge Marie survenues dans la première moitié de 1507 dans le couvent des dominicains de Berne. L’accusé fut d’abord un jeune frère convers du nom de Hans Jetzer qui avait vu les apparitions, puis ce fut le tour des supérieurs du couvent, suspectés d’être leurs instigateurs. Aymon de Montfalcon essaya de rendre justice aux deux parties. Il voulut d’abord préserver le frère convers d’un traitement trop dur de la part de la ville de Berne ; il voulut ensuite épargner les supérieurs du couvent de la condamnation au bûcher, même s’il les considérait probablement comme coupables, entre autres, du crime de profanation d’hostie qui semble l’avoir particulièrement préoccupé. Dans les deux cas, l’évêque de Lausanne défendait une cause perdue, peut-être aussi parce que, politiquement, il se situait du côté du roi de France qui était en train de perdre, à ce moment-là, la confiance – et les mercenaires – des Confédérés et de la ville de Berne

Palmitoylation of Plasmodium alveolins promotes cytoskeletal function.

S-palmitoylation is a post-translational lipid modification that is widespread among Plasmodium proteins and essential for parasite development. Little is known about the contribution of palmitoylation to the function of individual parasite molecules and structures. Alveolins are major components of the subpellicular network (SPN), a cortical cytoskeleton primarily involved in providing mechanical strength to the cell. We show here that the alveolin IMC1c is palmitoylated on a conserved cysteine motif, and that non-palmitoylated IMC1c displays normal expression, stability and trafficking. However, mutant parasites exhibit reduced osmotic stress resistance and tensile strength. These findings support the hypothesis that alveolin palmitoylation enhances cytoskeletal function by strengthening the connection between the SPN and the adjoining inner membrane complex via lipid anchoring
