    Registration of N614, A3N615, N616, and N617 Shattercane Genetic Stocks with Cytoplasmic or Nuclear Male Sterility and Juicy or Dry Midribs

    Four shattercane [Sorghum bicolor subsp. drummondii (Nees ex Steud.) de Wet ex Davidse] genetic stocks—N614 (Reg. No. GS-652, PI 665684), A3N615 (Reg. No. GS-651, PI 665683), N616 (Reg. No. GS-653, PI 665685), and N617 (Reg. No. GS-654, PI 665686)—with A3 cytoplasmic male sterility or the nuclear male sterility gene ms3 containing either juicy (dd) or dry (DD) culms were developed jointly by the USDA-ARS; the Iowa Agricultural and Home Economics Experiment Station, College of Agriculture and Life Sciences, Iowa State University; and the Agricultural Research Division, Institute of Agriculture and Natural Resources, University of Nebraska. The stocks were released in July 2011. The source material for these genetic stocks was isolated from an archetypical shattercane population found near Lincoln, NE. Release of these genetic stocks makes available shattercane lines with both A3 cytoplasmic male sterility, and ms3 genetic (nuclear) male sterility to facilitate crossing. These genetic stocks also contain juicy (dd) or dry (DD) culms, a visible genetic marker to facilitate screening progeny resulting from crosses. The genetic stocks have immediate application for basic research involving gene flow from cultivated sorghum [Sorghum bicolor (L.) Moench] to shattercane and on the fitness of offspring resulting from such crosses

    A new method of determining in vitro dry matter digestibility (IVDMD) as recently developed in which the digestion is conducted with the forage samples in filter bags. Our objective was to compare the filter bag and conventional IVDMD analysis methods using smooth bromegrass (Bromus inermis Leyss.), switchgrass (Panicum virgatum L.), and forage sorghum [Sorghum bicolor (L.) Moench] samples. In addition, the filter bag analysis systems for determining non-sequential neutral and acid detergent fiber (NDF and ADF), respectively, were compared with the non-sequential conventional analysis systems. In the filter bag systems, the forage samples are sealed in filter bags and the analyses are conducted on a batch basis rather than on an individual basis as in the conventional IVDMD and fiber analysis procedures. The filter bag analysis methods produced results similar to the conventional methods and ranked the forage samples in the same relative order

    Outcomes in Baby Deliveries among Pregnant Ebola Survivors

    Greater El Monte Community Hospital (GEMCH), the Los Angeles Department of Public Health, and the Centers for Medicare and Medicaid Services assisted in the first documented case of Ebola survivor delivery in the United States. A descriptive qualitative review of GEMCH’s events and the limited documented cases of outcomes of baby deliveries among EVD survivors is discussed. Limited resources and capacity in many developing countries impact adversely on the outcomes of the EVD survivors and their neonates. Three lessons for public health workers emerge: (1) the need for the United States to strengthen their capability to manage EVD cases and other highly contagious and severe infectious diseases; (2) the revealing that EVD survivors can deliver normal, EVD free babies when using the recommended guidelines; (3) The need for health care workers to adopt and share the practical procedures in the Recommended Guidelines by the CDC and LADPH from this event are useful and can be shared with the medical fraternity. This case illustrates that EVD survivors can be equally accepted and treated with success at designated health facilities. Demystifying Ebola and eliminating social stigma surrounding the disease is crucial in this undertaking

    Genetic analysis of seed traits in \u3ci\u3eSorghum bicolor\u3c/i\u3e that affect the human gut microbiome

    Prebiotic fibers, polyphenols and other molecular components of food crops significantly affect the composition and function of the human gut microbiome and human health. The abundance of these, frequently uncharacterized, microbiome-active components vary within individual crop species. Here, we employ high throughput in vitro fermentations of pre-digested grain using a human microbiome to identify segregating genetic loci in a food crop, sorghum, that alter the composition and function of human gut microbes. Evaluating grain produced by 294 sorghum recombinant inbreds identifies 10 loci in the sorghum genome associated with variation in the abundance of microbial taxa and/or microbial metabolites. Two loci co-localize with sorghum genes regulating the biosynthesis of condensed tannins. We validate that condensed tannins stimulate the growth of microbes associated with these two loci. Our work illustrates the potential for genetic analysis to systematically discover and characterize molecular components of food crops that influence the human gut microbiome

    The waxy mutation in sorghum and other cereal grains reshapes the gut microbiome by reducing levels of multiple beneficial species

    Waxy starches from cereal grains contain \u3e90% amylopectin due to naturally occurring mutations that block amylose biosynthesis. Waxy starches have unique organoleptic characteristics (e.g. sticky rice) as well as desirable physicochemical properties for food processing. Using isogenic pairs of wild type sorghum lines and their waxy derivatives, we studied the effects of waxy starches in the whole grain context on the human gut microbiome. In vitro fermentations with human stool microbiomes show that beneficial taxonomic and metabolic signatures driven by grain from wild type parental lines are lost in fermentations of grain from the waxy derivatives and the beneficial signatures can be restored by addition of resistant starch. These undesirable effects are conserved in fermentations of waxy maize, wheat, rice and millet. We also demonstrate that humanized gnotobiotic mice fed low fiber diets supplemented with 20% grain from isogenic pairs of waxy vs. wild type parental sorghum have significant differences in microbiome composition and show increased weight gain. We conclude that the benefits of waxy starches on food functionality can have unintended tradeoff effects on the gut microbiome and host physiology that could be particularly relevant in human populations consuming large amounts of waxy grains

    Discovery of diverse and functional antibodies from large human repertoire antibody libraries

    AbstractPhage display antibody libraries have a proven track record for the discovery of therapeutic human antibodies, increasing the demand for large and diverse phage antibody libraries for the discovery of new therapeutics. We have constructed naïve antibody phage display libraries in both Fab and scFv formats, with each library having more than 250billion clones that encompass the human antibody repertoire. These libraries show high fidelity in open reading frame and expression percentages, and their V-gene family distribution, VH-CDR3 length and amino acid usage mirror the natural diversity of human antibodies. Both the Fab and scFv libraries show robust sequence diversity in target-specific binders and differential V-gene usage for each target tested, supporting the use of libraries that utilize multiple display formats and V-gene utilization to maximize antibody-binding diversity. For each of the targets, clones with picomolar affinities were identified from at least one of the libraries and for the two targets assessed for activity, functional antibodies were identified from both libraries

    Experimental Study of the Shortest Reset Word of Random Automata

    In this paper we describe an approach to finding the shortest reset word of a finite synchronizing automaton by using a SAT solver. We use this approach to perform an experimental study of the length of the shortest reset word of a finite synchronizing automaton. The largest automata we considered had 100 states. The results of the experiments allow us to formulate a hypothesis that the length of the shortest reset word of a random finite automaton with nn states and 2 input letters with high probability is sublinear with respect to nn and can be estimated as $1.95 n^{0.55}.

    Pharmacological Characterization of FE 202158, a Novel, Potent, Selective, and Short-Acting Peptidic Vasopressin V 1a Receptor Full Agonist for the Treatment of Vasodilatory Hypotension

    ABSTRACT R), and oxytocin receptor, respectively] contrasting with AVP's lack of selectivity, especially versus the V 2 R (selectivity ratio of 1:18:0.2:92; human V 1a R EC 50 ϭ 0.24 nM). This activity and selectivity profile was confirmed in radioligand binding assays. FE 202158 was a potent vasoconstrictor in the isolated rat common iliac artery ex vivo (EC 50 ϭ 3.6 nM versus 0.8 nM for AVP) and reduced rat ear skin blood flow after intravenous infusion in vivo (ED 50 ϭ 4.0 versus 3.4 pmol/kg/min for AVP). The duration of its vasopressor effect by intravenous bolus in rats was as short as AVP at submaximally effective doses. FE 202158 had no V 2 R-mediated antidiuretic activity in rats by intravenous infusion at its ED 50 for reduction of ear skin blood flow, in contrast with the pronounced antidiuretic effect of AVP. Thus, FE 202158 seems suitable for treatment of conditions where V 1a R activity is desirable but V 2 R activity is potentially deleterious, such as vasodilatory hypotension in septic shock. In addition to the desirable selectivity profile, its shortacting nature should allow dose titration with rapid onset and offset of action to optimize vasoconstriction efficacy and safety
