    Temporal and Spatial Cellular Distribution of Neural Crest Derivatives and Alpha Cells during Islet Development

    Recent studies have revealed that signals from neural crest (NC) derivatives regulate the mass, proliferation, and maturation of beta cells in developing fetal pancreas. However, little is known about the cellular distribution of NC derivatives during pancreatic development or the process whereby the developing islets are enclosed. We studied the temporal and spatial distribution of NC derivatives and endocrine cells at each developmental stage. At embryonic day 10.5 (E10.5) of mouse embryo, NC derivatives that migrated to the prospective pancreatic region were distributed in close proximity to pancreatic epithelial cells. As development advanced, most NC derivatives progressively surrounded endocrine rather than exocrine cells, and were distributed in closer proximity to alpha cells rather than to beta cells. At E20, approximately 70% of the NC derivatives enclosing endocrine cells were distributed in close proximity to alpha cells. Moreover, the expression of SynCAM, a Ca2+-independent homophilic trans-cell adhesion molecule, was confirmed from E16.5 on and was more remarkable at the cell boundaries of alpha cells and NC derivatives. These findings suggest that NC derivatives might be distributed in close proximity to alpha cells as a result of homophilic binding of SynCAM expressed by alpha cells and NC derivatives during islet development

    Free Flap Blood Flow Evaluated Using Two-Dimensional Laser Speckle Flowgraphy

    Objective. We investigated the efficiency of laser speckle flowgraphy for evaluating blood flow in free flaps used for plastic surgery. Methods. We measured blood flow using a visual laser meter capable of providing two-dimensional color graphic representations of flow distribution for a given area using a dynamic laser speckle effect. Using laser speckle flowgraphy, we examined the blood flow of 20 free flaps applied following the excision of head and neck tumors. Results. After anastomosis of the feeding and draining blood vessels and sewing the flap, musculocutaneous (MC) flaps showed significantly lower blood flow than jejunal or omental flaps (P < .05). The ratio of blood flow decrease from the edge to the center was significantly greater in MC flaps than in jejunal or omental flaps (P < .001). Conclusion. Laser speckle flowgraphy is useful for the perioperative measurement of blood flow in free flaps used in plastic surgery. This method is a highly useful, practical, and reliable tool for assessing cutaneous blood flow and is expected to be applicable to several clinical fields

    Structural basis of lignocellulose deconstruction by the wood-feeding anobiid beetle Nicobium hirtum

    The details of the lignocellulose deconstruction processes in the digestive systems of wood-feeding insects remain elusive. This study aimed to examine the biochemical conversion of lignocellulose in the digestive system of a wood-feeding anobiid beetle, Nicobium hirtum, one of the most important pests of wooden products in Japan. To this end, N. hirtum larvae were fed with Japanese red pine (softwood) and Japanese beech (hardwood) sapwood diets, as well as an artificial diet containing Shorea wood (hardwood) sapwood sawdust. The structural differences between the original and digested (feces) lignocellulose samples were examined using wet-chemical and two-dimensional (2D) nuclear magnetic resonance (NMR) methods. Cellulose and hemicelluloses, especially mannan in the softwood diet, were preferentially degraded over lignin in the larval digestive system. As a result, lignin was enriched in the digested lignocellulose residues. Lignin compositional analyses based on thioacidolysis and 2D NMR determined that the proportions of oxidized lignin aromatic units were notably increased after digestion. Further, the 2D NMR analyses revealed the accumulation of aldehyde and hydroxypropiovanillone/syringone end-unit structures in lignin, indicating that oxidative and/or reductive modifications of lignin polymers occur in the larval digestive system. Such structural alterations of lignin may facilitate the dissociation of the lignin barrier, thereby liberating polysaccharides for subsequent enzymatic conversion for assimilation and energy

    We report on blood coagulation factor XⅢ(F13)activity in upper gastrointestinal bleeding (UGIB)due to peptic ulcer(PU). 【Methods】From January 2011 to December 2016, 42 patients who had UGIB with PU, performed endoscopic hemostasis(EH), and measured F13 activity (normal range:70‐140%), were retrospectively studied. The clinical signs, peripheral blood, biochemistry, coagulation, F13 activity, and endoscopic findings were examined by logistic regression analysis(LRA)in32patients with successful hemostasis and10patients with rebleeding. 【Results】There were25elderly patients aged70 and over. The average F13 activity was 65.7±22.5%, 68.3±22.1% in the case of successful hemostasis, below the lower limit of normal, and further decreased to 57.5±18.1% in the case of rebleeding. LRA showed association with rebleeding in Forrest classification and F13activity. 【Conclusion】F13activity decreased in many cases of UGIB due to PU, and associated with rebleeding after EH

    A Feynman-graph generator for any order of coupling constants

    A computer program has been developed which generates Feynman graphs automatically for scattering and decay processes in non-Abelian gauge theory of high-energy physics. A new acceleration method is presented for both generating and eliminating graphs. This method has been shown to work quite efficiently for any order of coupling constants in any kind of theoretical model. A utility program is also available for drawing generated graphs.     Remarkable features of ovarian morphology and reproductive hormones in insulin-resistant Zucker fatty (fa/fa) rats

    <p>Abstract</p> <p>Background</p> <p>Zucker fatty (fa/fa) rats are a well-understood model of obesity and hyperinsulinemia. It is now thought that obesity/hyperinsulinemia is an important cause of endocrinological abnormality, but to date there have been no reports on the changes in ovarian morphology or the ovarian androgen profile in rat models of obesity and insulin resistance.</p> <p>Methods</p> <p>In this study we investigated the effects of obesity and hyperinsulinemia on ovarian morphology and the hormone profile in insulin-resistant Zucker fatty rats (5, 8, 12 and 16 weeks of age, n = 6-7).</p> <p>Results</p> <p>Ovaries from 5-week-old fatty rats had significantly greater total and atretic follicle numbers, and higher atretic-to-total follicle ratios than those from lean rats. Ovaries from 12- and 16-week-old fatty rats showed interstitial cell hyperplasia and numerous cysts with features of advanced follicular atresia. In addition, serum testosterone and androstenedione levels significantly declined in fatty rats from age 8 to 16 weeks, so that fatty rats showed significantly lower levels of serum testosterone (12 and 16 weeks) and androstenedione (all weeks) than lean rats. This may reflect a reduction of androgen synthesis during follicular atresia. Serum adiponectin levels were high in immature fatty rats, and although the levels declined significantly as they matured, it remained significantly higher in fatty rats than in lean rats. On the other hand, levels of ovarian adiponectin and its receptors were significantly lower in mature fatty rats than in lean mature rats or immature fatty rats.</p> <p>Conclusions</p> <p>Our findings indicate that ovarian morphology and hormone profiles are significantly altered by the continuous insulin resistance in Zucker fatty rats. Simultaneously, abrupt reductions in serum and ovarian adiponectin also likely contribute to the infertility seen in fatty rats.</p
