    The Influence of Cross-Language Similarity on within- and between-Language Stroop Effects in Trilinguals

    This study investigated effects of cross-language similarity on within- and between-language Stroop interference and facilitation in three groups of trilinguals. Trilinguals were either proficient in three languages that use the same-script (alphabetic in German–English–Dutch trilinguals), two similar scripts and one different script (Chinese and alphabetic scripts in Chinese–English–Malay trilinguals), or three completely different scripts (Arabic, Chinese, and alphabetic in Uyghur–Chinese–English trilinguals). The results revealed a similar magnitude of within-language Stroop interference for the three groups, whereas between-language interference was modulated by cross-language similarity. For the same-script trilinguals, the within- and between-language interference was similar, whereas the between-language Stroop interference was reduced for trilinguals with languages written in different scripts. The magnitude of within-language Stroop facilitation was similar across the three groups of trilinguals, but smaller than within-language Stroop interference. Between-language Stroop facilitation was also modulated by cross-language similarity such that these effects became negative for trilinguals with languages written in different scripts. The overall pattern of Stroop interference and facilitation effects can be explained in terms of diverging and converging color and word information across languages

    EXPRESS: Stratified Distributional Analysis -- a Novel Perspective on RT Distributions

    Response times and their distributions serve as a powerful lens into cognitive processes. We present a novel statistical methodology called Stratified Distributional Analysis (SDA) to quantitatively assess how key determinants of response times (word frequency and length) shape their distributions. Taking advantage of the availability of millions of lexical decision response times in the English Lexicon Project and the British Lexicon Project, we made important advances into the theoretical issue of linking response times and word frequency by analysing RT distributions as a function of word frequency and word length. We tested these distributions against the lognormal, Wald, and Gamma distributions and three measures of word occurrence (word form frequencies obtained from subtitles and contextual diversity as operationalized as discourse contextual diversity and user contextual diversity). We found that the RT distributions were best described by a lnorm distribution across both megastudies when word occurrence was quantified by a contextual diversity measure. The link between the lnorm distribution and its generative process highlights the power of SDA in elucidating mechanisms that govern the generation of RTs through the fitting of probability distributions. Using a hierarchical Bayesian framework, SDA yielded posterior distributions for the distributional parameters at the single-participant level, enabling probabilistic predictions of response times as a function of word frequency and word length, which has the potential to serve as a diagnostic tool to uncover idiosyncratic features of word processing. Crucially, while we applied our parsimonious methodology to lexical decision response times, it is applicable to a variety of tasks such as word-naming and eye-tracking data

    Localized microglia dysregulation impairs central nervous system myelination in development

    Myelination of neuronal axons is a critical aspect of central nervous system development and function. However, the fundamental cellular and molecular mechanisms influencing human developmental myelination and its failure are not fully understood. Here, we used digital spatial transcriptomics of a rare bank of human developing white matter to uncover that a localized dysregulated innate immune response is associated with impeded myelination. We identified that poorly myelinating areas have a distinct signature of Type II interferon signalling in microglia/macrophages, relative to adjacent myelinating areas. This is associated with a surprising increase in mature oligodendrocytes, which fail to form myelin processes appropriately. We functionally link these findings by showing that conditioned media from interferon-stimulated microglia is sufficient to dysregulate myelin process formation by oligodendrocytes in culture. We identify the Type II interferon inducer, Osteopontin (SPP1), as being upregulated in poorly myelinating brains, indicating a potential biomarker. Our results reveal the importance of microglia-mature oligodendrocyte interaction and interferon signaling in regulating myelination of the developing human brain.</p

    An inter-specialty cancer training programme curriculum for Europe

    Introduction Multidisciplinary and multi-professional collaboration is vital in providing better outcomes for patients The aim of the INTERACT-EUROPE Project (Wide Ranging Cooperation and Cutting Edge Innovation As A Response To Cancer Training Needs). The aim was to develop an inter-specialty curriculum. A pilot project will enable a pioneer cohort to acquire a sample of the competencies needed. Methods A scoping review, qualitative and quantitative surveys were undertaken. The quantitative survey results are reported here. Respondents, including members of education boards, curriculum committees, trainee committees of European specialist societies and the ECO Patient Advisory Committee, were asked to score 127 proposed competencies on a 7-point Likert scale as to their value in achieving the aims of the curriculum. Results were discussed and competencies developed at two stakeholder meetings. A consultative document, shared with stakeholders and available online, requested views regarding the other components of the curriculum. Results and Discussion: Eleven competencies were revised, three omitted and three added. The competencies were organised according to the CanMEDS framework with 13 Entrustable Professional Activities, 23 competencies and 127 enabling competencies covering all roles in the framework. Recommendations regarding the infrastructure, organisational aspects, eligibility of trainees and training centres, programme contents, assessment and evaluation were developed using the replies to the consultative document. Conclusions An Inter-specialty Cancer Training Programme Curriculum and a pilot programme with virtual and face-to-face components have been developed with the aim of improving the care of people affected by cancer

    Neurological complications are common in COVID-19. Although SARS-CoV-2 has been detected in patients' brain tissues, its entry routes and resulting consequences are not well understood. Here, we show a pronounced upregulation of interferon signaling pathways of the neurovascular unit in fatal COVID-19. By investigating the susceptibility of human induced pluripotent stem cell (hiPSC)-derived brain capillary endothelial-like cells (BCECs) to SARS-CoV-2 infection, we found that BCECs were infected and recapitulated transcriptional changes detected in vivo. While BCECs were not compromised in their paracellular tightness, we found SARS-CoV-2 in the basolateral compartment in transwell assays after apical infection, suggesting active replication and transcellular transport of virus across the blood-brain barrier (BBB) in vitro. Moreover, entry of SARS-CoV-2 into BCECs could be reduced by anti-spike-, anti-angiotensin-converting enzyme 2 (ACE2)-, and anti-neuropilin-1 (NRP1)-specific antibodies or the transmembrane protease serine subtype 2 (TMPRSS2) inhibitor nafamostat. Together, our data provide strong support for SARS-CoV-2 brain entry across the BBB resulting in increased interferon signaling
