    Comparative study of planning history, spatial development and sociological significance of the back alley in Yangon and Singapore

    A comparison of two similar types of back alley spaces: Back Drainage Space (BDS) in Yangon and Back-lane in Singapore, is conducted to find out if there are any relationships between their spatial development and lessons that can be learned from the precedent case in Singapore. Commonalities and differences of the back alley are identified by comparing the historical urban context of their formation and development in British colonial cities. The findings suggest that the back alley spaces in both cities, despite divergent circumstances and development after independence, possess common traits as interstitial space between public and private, with a unique way of spatial management based on informal, mutual agreement, which suggests some useful ideas when considering the role of these spaces in the redevelopment of Yangon in the near future

    Functional expression of choline transporter-like protein 1 (CTL1) in small cell lung carcinoma cells: A target molecule for lung cancer therapy

    AbstractCholine is essential for the synthesis of the major membrane phospholipid phosphatidylcholine and the neurotransmitter acetylcholine (ACh). Elevated levels of choline and up-regulated choline kinase activity have been detected in cancer cells. Thus, the intracellular accumulation of choline through choline transporters is the rate-limiting step in phospholipid metabolism and a prerequisite for cancer cell proliferation. However, the uptake system for choline and the functional expression of choline transporters in lung cancer cells are poorly understood. We examined the molecular and functional characterization of choline uptake in the small cell lung carcinoma cell line NCI-H69. Choline uptake was saturable and mediated by a single transport system. Interestingly, removal of Na+ from the uptake buffer strongly enhanced choline uptake. This increase in choline uptake under the Na+-free conditions was inhibited by dimethylamiloride (DMA), a Na+/H+ exchanger (NHE) inhibitor. Various organic cations and the choline analog hemicholinium-3 (HC-3) inhibited the choline uptake and cell viability. A correlation analysis of the potencies of organic cations for the inhibition of choline uptake and cell viability showed a strong correlation (R=0.8077). RT-PCR revealed that choline transporter-like protein 1 (CTL1) mRNA and NHE1 are mainly expressed. HC-3 and CTL1 siRNA inhibited choline uptake and cell viability, and increased caspase-3/7 activity. The conversion of choline to ACh was confirmed, and this conversion was enhanced under Na+-free conditions, which in turn was sensitive to HC-3. These results indicate that choline uptake through CTL1 is used for ACh synthesis. Both an acetylcholinesterase inhibitor (eserine) and a butyrylcholinesterase inhibitor (ethopropazine) increased cell proliferation, and these effects were inhibited by 4-DAMP, a mAChR3 antagonist. We conclude that NCI-H69 cells express the choline transporter CTL1 which uses a directed H+ gradient as a driving force, and its transport functions in co-operation with NHE1. This system primarily supplies choline for the synthesis of ACh and secretes ACh to act as an autocrine/paracrine growth factor, and the functional inhibition of CTL1 could promote apoptotic cell death. Identification of this new CTL1-mediated choline transport system provides a potential new target for therapeutic intervention

    WMO Space-Based Weather and Climate Extremes Monitoring Demonstration Project (SEMDP): First Outcomes of Regional Cooperation on Drought and Heavy Precipitation Monitoring for Australia and Southeast Asia

    To improve monitoring of extreme weather and climate events from space, the World Meteorological Organization (WMO) initiated the space-based weather and climate extremes monitoring demonstration project (SEMDP). Presently, SEMDP is focused on drought and heavy precipitation monitoring over Southeast Asia and the Pacific. Space-based data and derived products form critical part of meteorological services’ operations for weather monitoring; however, satellite products are still not fully utilized for climate applications. Using SEMDP satellite-derived precipitation products, it would be possible to monitor extreme precipitation events with uniform spatial coverage and over various time periods – pentad, weekly, 10 days, monthly and longer time-scales. In this chapter, SEMDP satellite-derived precipitation products over the Asia-Pacific region produced by the Earth Observation Research Center/Japan Aerospace Exploration Agency (EORC/JAXA) and the Climate Prediction Center/National Oceanic and Atmospheric Administration (CPC/NOAA) are introduced. Case studies for monitoring (i) drought in Australia in July-October 2007 and September 2018 and (ii) heavy precipitation over Australia in December 2010 and Thailand and the Peninsular Malaysia in November-December 2014 which caused widespread flooding are also presented. Satellite observations are compared with in situ data to demonstrate value of satellite-derived estimates of precipitation for drought and heavy rainfall monitoring

    A Field Survey of Window-Opening Behavior and Thermal Conditions in Apartments of Surabaya, Indonesia

    A field survey was conducted in apartments of Surabaya, Indonesia to investigate occupants’ window-opening behavior and their thermal conditions. A total of 347 households were interviewed and about 30 houses were covered for measurements of thermal conditions. It was seen that occupants in the naturally ventilated apartments tend to open windows or doors as much as possible at least during daytime for satisfying air flow and ventilation in particular. The average duration of opening windows/doors was 16-17 hours/day in the naturally ventilated apartments, while the corresponding duration in air-conditioned private apartments was less than 5 hours/day. The results of measurement showed that generally indoor air temperatures was about 6°C lower than the outdoors during 11:00 to 19:00, while it was 3°C higher than the outdoors for the rest of the day, i.e. nighttime. The structural cooling effect was evident in these apartments

    Immune Modulation by Telomerase-Specific Oncolytic Adenovirus Synergistically Enhances Antitumor Efficacy with Anti-PD1 Antibody

    The clinical benefit of monotherapy involving immune checkpoint inhibitors (ICIs) such as anti-programmed death-1 antibody (PD-1 Ab) is limited to small populations. We previously developed a telomerase-specific oncolytic adenovirus, Telomelysin (OBP-301), the safety of which was confirmed in a phase I clinical study. Here, we examined the potential of OBP-502, an OBP-301 variant, as an agent for inducing immunogenic cell death (ICD) and synergistically enhancing the efficacy of OBP-502 with PD-1 Ab using CT26 murine colon cancer and PAN02 murine pancreatic cancer cell lines. OBP-502 induced the release of ICD molecules such as adenosine triphosphate (ATP) and high-mobility group box protein 1 (HMGB1) from CT26 and PAN02 cells, leading to recruitment of CD8-positive lymphocytes and inhibition of Foxp3-positive lymphocyte infiltration into tumors. Combination therapy involving OBP-502 intratumoral administration and PD-1 Ab systemic administration significantly suppressed the growth of not only OBP-502-treated tumors but also tumors not treated with OBP-502 (so-called abscopal effect) in CT26 and PAN02 bilateral subcutaneous tumor models, in which active recruitment of CD8-positve lymphocytes was observed even in tumors not treated with OBP-502. This combined efficacy was similar to that observed in a CT26 rectal orthotopic tumor model involving liver metastases. In conclusion, telomerase-specific oncolytic adenoviruses are promising candidates for combined therapies with ICIs

    A Case of Metastatic Fumarate Hydratase-Deficient–like Renal Cell Carcinoma Successfully Managed by Ipilimumab plus Nivolumab

    We report a 62-year-old male with metastatic fumarate hydratase-deficient renal cell carcinoma (FH-deficient RCC) without fumarate hydratase (FH) mutation (FH-deficient–like RCC). The International Metastatic RCC Database Consortium risk score was intermediate, and immunotherapy with nivolumab and ipilimumab (Ipi/ Nivo) was initiated. Four cycles of Ipi/Nivo and 5 cycles of nivolumab resulted in a complete response of the metastases. Hypophysitis occurred as an immune-related adverse event after four cycles of Ipi/Nivo. The prognosis of patients with FH-deficient RCC is generally poor. Few reports of FH-deficient RCC successfully treated with Ipi/Nivo have been published. Ipi/Nivo can be effective for treating FH-deficient RCC
