    AbstractThe human MDR1 gene encodes the multidrug transporter P-glycoprotein (Pgp). Although the MDR2/Pgp shares about 80% identity at the amino acid level with the MDRI/Pgp, the MDR2/Pgp cannot act as a multidrug transporter. We examined the drug sensitivity of Saccharomyces cerevisiae expressing either the human MDR1/Pgp or MDR2/Pgp. The human MDR1/Pgp conferred about 4-fold resistance to aureobasidin A, a cyclic depsipeptide antifungal antibiotic, on the drug-sensitive yeast strains. Interestingly the human MDR2/Pgp also conferred about 2.5-fold resistance to aureobasidin A. The resistance to aureobasidin A conferred by the MDR2/Pgp as well as by the MDR1/Pgp was overcome by vinblastine, verapamil, and cyclosporin A, depending on their concentrations, but not by colchicine. Aureobasidin A probably interacts directly with Pgps, because it overcame multidrug resistance of human cells and inhibited azidopine photoaffinity labeling of MDRI/Pgp in human cell membranes. These results suggest the possibility that the human MDR1 and MDR2/Pgps have conserved domain(s) for drug recognition

    An Intermediate-band imaging survey for high-redshift Lyman Alpha Emitters: The Mahoroba-11

    We present results of our intermediate-band optical imaging survey for high-zz Lyα\alpha emitters (LAEs) using the prime focus camera, Suprime-Cam, on the 8.2m Subaru Telescope. In our survey, we use eleven filters; four broad-band filters (BB, RcR_{\rm c}, ii^\prime, and zz^\prime) and seven intermediate-band filters covering from 500 nm to 720 nm; we call this imaging program as the Mahoroba-11. The seven intermediate-band filters are selected from the IA filter series that is the Suprime-Cam intermediate-band filter system whose spectral resolution is R=23R = 23. Our survey has been made in a 34×2734^\prime \times 27^\prime sky area in the Subaru XMM Newton Deep Survey field. We have found 409 IA-excess objects that provide us a large photometric sample of strong emission-line objects. Applying the photometric redshift method to this sample, we obtained a new sample of 198 LAE candidates at 3<z<53 < z < 5. We found that there is no evidence for evolution of the number density and the star formation rate density for LAEs with logL(Lyα)(ergs1)>42.67\log L({\rm Ly}\alpha) ({\rm erg s^{-1}}) > 42.67 between z3z \sim 3 and 5.Comment: 46 pages, 15 figures, PASJ, Vol.57, No.6, in pres

    Accelerating effects of silk fibroin on wound healing in hairless descendants of Mexican hairless dogs

    Abstract: We examined the safety of silk fibroin in the subcutaneous tissues. In addition, we macroscopically and histopathologically evaluated its healing effect on the full-thickness wounds in hairless dogs. We prepared 3 types of matrices including amorphous silk fibroin films, and á-and â-type silk fibroin powder. No toxicity was found in the sites injected with silk fibroin solutions. Macroscopically, silk fibroin films had accelerating effects on wound repair, as compared with occlusive dressings (dried porcine skin and hydrocolloid dressings). Histopathological examinations revealed that silk fibroin films facilitate reepithelialization and the formation of granulation tissues, collagens and elastic fibers. The sites treated with the á-type silk fibroin powder were faster in wound repair than those treated with the â-type silk fibroin powder. The á-type silk fibroin powder absorbed excessive exudate. Microscopically, there are epidermal and dermal regeneration in the sites treated with the á-type silk fibroin powder. These results show that silk fibroin is inert in biological tissues, indicating excellent biocompatibility. Silk fibroin films facilitate reepithelialization, remodeling of connective tissues and collagenization. The á-type fibroin powder is a wound dressing that regulates excessive exudate from the wound and provides a proper moist environment. These results suggest that silk fibroin is a useful dressing material in veterinary clinical medicine

    Effect of Cisternal Administration of Nicardipine Hydrochloride on Cerebral Vasospasm: A Preliminary Report

    In six subarachnoid hemorrhage (SAH) cases due to aneurysmal rupture, effects of cisternal administration of Nicardipine hydrochloride (nicardipine) on vasospasm wer studied. Nicardipine is one of calcium antagonists. All aneurysms were clipped within 48 hr after the onset of SAH with ventricular and cisternal drainages. Subarachnoid clot was removed as much as possible. Nicardipine or mixture of nicardipine and urokinase was administered through cisternal drainage once a day for 10 days postoperatively. The dose of nicardipine was 2 mg or 4 mg, and that of urokinase was 6,000 units or 24,000 units. On the 7th day after the onset of SAH, angiograms were performed before and 30 min after the cisternal administration of nicardipine. Then the diameters of each arteries were compared in order to estimate the vasodilative effects of nicardipine angiographically. In this study nicardipine was considered to have some prophylactic effect on vasospasm because vasospasm was not observed either angiographically or clinically in 2 out of 3 severe SAH cases on CT

    Topological Strings, Flat Coordinates and Gravitational Descendants

    We discuss physical spectra and correlation functions of topological minimal models coupled to topological gravity. We first study the BRST formalism of these theories and show that their BRST operator Q=Qs+QvQ=Q_s+Q_v can be brought to QsQ_s by a certain homotopy operator UU, UQU1=QsUQU^{-1}=Q_s (QsQ_s and QvQ_v are the N=2N=2 and diffeomorphism BRST operators, respectively). The reparametrization (anti)-ghost bb mixes with the supercharge operator GG under this transformation. Existence of this transformation enables us to use matter fields to represent cohomology classes of the operator QQ. We explicitly construct gravitational descendants and show that they generate the higher-order KdV flows.     A NEW OPERATIVE TECHNIQUE FOR ESOPHAGEAL RECONSTRUCTION USING A LONG GASTRIC TUBE OF 3 CM DIAMETER

    The anthors have successfully prepared a 3 cm diameter gastric tube with a more favorable blood supply than the conventional one by performing interrupted suture each of the mucosal layers and seromuscular layers of the stomach separately with pyloromyotomy as a drainage procedure. This gastric tube caused no clavicular pressure on the anastomotic portion through a retrosternal route, so that we could perform end-to-end anastomosis between the cervical esophagus and the gastric tube in a region higher than the clavicle. We report in this paper the clinical findings of our procedure : it was effective in preventing postoperative complications such as suture insufficiency and passage disturbance other than postoperative complaints due to dumping and reflux esophagitis, etc
