    Not too big for its mouth: direct evidence of a macrodasyidan gastrotrich preyed in nature by a dileptid ciliate

    Nearly ubiquitous and usually speciose in most aquatic habitats, the meiofaunal-sized gastrotrichs are recognized as an important component of marine and freshwater ecosystems. The common observations that gastrotrichs feed on bacteria, microalgae and biodetritus strongly imply that they play a relevant role in linking the microbial loop to the higher trophic levels. Which are the organisms that in turn prey on gastrotrichs is, however, a substantially unexplored question. Inspecting meiofauna samples collected from shallow sites of the Tyrrhenian coast, we had the chance to spot a wild case of a macrodasyidan gastrotrich predated by a dileptid ciliate. This case is documented here with a set of in-vivo photos, jointly with an unequivocal taxonomic identification of the preyed gastrotrich with Paraturbanella teissieri and a tentative identification of the predator ciliate with Pseudomonilicaryon marinus

    The research project aimed at the investigation of different methods for the storage of fresh-cut fruit. This case study relates to the proposal of an innovative controlled release system to improve the shelf life of ready-to-eat fruit. The methods applied during three research years included the application of active molecules from natural substances, some widely used in commerce and other tested for the first time; first analyzed in vitro and then applied in vivo. The under consideration methods concerned dipping, coating and the layer-by-layer assembly. The analysis carried out on the fruit have monitored pomological traits performances (soluble solids content, titratable acidity, pH, color, flesh firmness), the chemical profile (polyphenoloxidase, carotenoids) and microbial growth

    Use of weaning concentrate in the feeding suckling kids: effects on meat quality

    The effect of a feeding supplementation with starter concentrate on “Capretto” meat production and its qualitative characteristics was investigated. The results showed that the effect of concentrate supplementation did not influence the slaughter weight, slaughter and dissection data, tissue composition and meat chemical composition of the pelvic limb. No differences were found for rheological characteristics of LD meat, except cohesiveness values, which were higher in the concentrate group kids

    MiR-205-5p inhibition by locked nucleic acids impairs metastatic potential of breast cancer cells

    Mir-205 plays an important role in epithelial biogenesis and in mammary gland development but its role in cancer still remains controversial depending on the specific cellular context and target genes. We have previously reported that miR-205-5p is upregulated in breast cancer stem cells targeting ERBB pathway and leading to targeted therapy resistance. Here we show that miR-205-5p regulates tumorigenic properties of breast cancer cells, as well as epithelial to mesenchymal transition. Silencing this miRNA in breast cancer results in reduced tumor growth and metastatic spreading in mouse models. Moreover, we show that miR-205-5p knock-down can be obtained with the use of specific locked nucleic acids oligonucleotides in vivo suggesting a future potential use of this approach in therapy

    Polarization Response in InAs Quantum Dots: Theoretical Correlation between Composition and Electronic Properties

    III-V growth and surface conditions strongly influence the physical structure and resulting optical properties of self-assembled quantum dots (QDs). Beyond the design of a desired active optical wavelength, the polarization response of QDs is of particular interest for optical communications and quantum information science. Previous theoretical studies based on a pure InAs QD model failed to reproduce experimentally observed polarization properties. In this work, multi-million atom simulations are performed to understand the correlation between chemical composition and polarization properties of QDs. A systematic analysis of QD structural parameters leads us to propose a two layer composition model, mimicking In segregation and In-Ga intermixing effects. This model, consistent with mostly accepted compositional findings, allows to accurately fit the experimental PL spectra. The detailed study of QD morphology parameters presented here serves as a tool for using growth dynamics to engineer the strain field inside and around the QD structures, allowing tuning of the polarization response.Comment: 8 pages, 6 figures; accepted for publication in IOP Nanotechnology journa

    Microenvironment in neuroblastoma: Isolation and characterization of tumor-derived mesenchymal stromal cells

    Background: It has been proposed that mesenchymal stromal cells (MSCs) promote tumor progression by interacting with tumor cells and other stroma cells in the complex network of the tumor microenvironment. We characterized MSCs isolated and expanded from tumor tissues of pediatric patients diagnosed with neuroblastomas (NB-MSCs) to define interactions with the tumor microenvironment. Methods: Specimens were obtained from 7 pediatric patients diagnosed with neuroblastoma (NB). Morphology, immunophenotype, differentiation capacity, proliferative growth, expression of stemness and neural differentiation markers were evaluated. Moreover, the ability of cells to modulate the immune response, i.e. inhibition of phytohemagglutinin (PHA) activated peripheral blood mononuclear cells (PBMCs) and natural killer (NK) cytotoxic function, was examined. Gene expression profiles, known to be related to tumor cell stemness, Wnt pathway activation, epithelial-mesenchymal transition (EMT) and tumor metastasis were also evaluated. Healthy donor bone marrow-derived MSCs (BM-MSC) were employed as controls. Results: NB-MSCs presented the typical MSC morphology and phenotype. They showed a proliferative capacity superimposable to BM-MSCs. Stemness marker expression (Sox2, Nanog, Oct3/4) was comparable to BM-MSCs. NB-MSC in vitro osteogenic and chondrogenic differentiation was similar to BM-MSCs, but NB-MSCs lacked adipogenic differentiation capacity. NB-MSCs reached senescence phases at a median passage of P7 (range, P5-P13). NB-MSCs exhibited greater immunosuppressive capacity on activated T lymphocytes at a 1:2 (MSC: PBMC) ratio compared with BM-MSCs (p = 0.018). NK cytotoxic activity was not influenced by co-culture, either with BM-MSCs or NB-MSCs. Flow-cytometry cell cycle analysis showed that NB-MSCs had an increased number of cells in the G0-G1 phase compared to BM-MSCs. Transcriptomic profiling results indicated that NB-MSCs were enriched with EMT genes compared to BM-MSCs. Conclusions: We characterized the biological features, the immunomodulatory capacity and the gene expression profile of NB-MSCs. The NB-MSC gene expression profile and their functional properties suggest a potential role in promoting tumor escape, invasiveness and metastatic traits of NB cancer cells. A better understanding of the complex mechanisms underlying the interactions between NB cells and NB-derived MSCs should shed new light on potential novel therapeutic approaches

    In pre-hospital care, an accurate and quick diagnosis of ST-segment elevation myocardial infarction (STEMI) is imperative to promptly kick-off the STEMI network with a direct transfer to the cardiac catheterization laboratory (cath lab) in order to reduce myocardial infarction size and mortality. Aa atherosclerotic plaque rupture is the main mechanism responsible for STEMI. However, in a small percentage of patients, emergency coronarography does not reveal any significant coronary stenosis. The fluoropyrimidine agents such as 5-Fluorouracil (5-FU) and capecitabine, widely used to treat gastrointestinal, breast, head and neck cancers, either as a single agent or in combination with other chemotherapies, can cause potentially lethal cardiac side effects. Here, we present the case of a patient with 5-FU cardiotoxicity resulting in an acute coronary syndrome (ACS) with recurrent episodes of chest pain and ST-segment elevation.. Our case report highlights the importance of widening the knowledge among cardiologists of the side effects of chemotherapeutic drugs, especially considering the rising number of cancer patients around the world and that fluoropyrimidines are the main treatment for many types of cancer, both in adjuvant and advanced settings

    Determinants of intraregional migration in Sub-Saharan Africa 1980-2000

    Despite great accomplishments in the migration literature, the determinants of South-South migration remain poorly understood. In an attempt to fill this gap, this paper formulates and tests an empirical model for intraregional migration in sub-Saharan Africa within an extended human capital framework, taking into account spatial interaction. Using bilateral panel data between 1980 and 2000, we find that intraregional migration on the subcontinent is predominantly driven by economic opportunities and sociopolitics in the host country, facilitated by geographical proximity. The role played by network effects and environmental conditions is also apparent. Finally, origin and destination spatial dependence should definitely not be ignored

The Fauna Europaea project covers about 230,000 taxonomic names, including 130,000 accepted species and 14,000 accepted subspecies, which is much more than the originally projected number of 100,000 species.     Interleukin-30 feeds breast cancer stem cells via CXCL10 and IL23 autocrine loops and shapes immune contexture and host outcome

    Background Breast cancer (BC) progression to metastatic disease is the leading cause of death in women worldwide. Metastasis is driven by cancer stem cells (CSCs) and signals from their microenvironment. Interleukin (IL) 30 promotes BC progression, and its expression correlates with disease recurrence and mortality. Whether it acts by regulating BCSCs is unknown and could have significant therapeutic implications. Methods Human (h) and murine (m) BCSCs were tested for their production of and response to IL30 by using flow cytometry, confocal microscopy, proliferation and sphere-formation assays, and PCR array. Immunocompetent mice were used to investigate the role of BCSC-derived IL30 on tumor development and host outcome. TCGA PanCancer and Oncomine databases provided gene expression data from 1084 and 75 hBC samples, respectively, and immunostaining unveiled the BCSC microenvironment. Results hBCSCs constitutively expressed IL30 as a membrane-anchored glycoprotein. Blocking IL30 hindered their proliferation and self-renewal efficiency, which were boosted by IL30 overexpression. IL30 regulation of immunity gene expression in human and murine BCSCs shared a significant induction of IL23 and CXCL10. Both immunoregulatory mediators stimulated BCSC proliferation and self-renewal, while their selective blockade dramatically hindered IL30-dependent BCSC proliferation and mammosphere formation. Orthotopic implantation of IL30-overexpressing mBCSCs, in syngeneic mice, gave rise to poorly differentiated and highly proliferating MYC + KLF4 + LAG3 + tumors, which expressed CXCL10 and IL23, and were infiltrated by myeloid-derived cells, Foxp3 + T regulatory cells and NKp46 + RORÎ 3t + type 3 innate lymphoid cells, resulting in increased metastasis and reduced survival. In tumor tissues from patients with BC, expression of IL30 overlapped with that of CXCL10 and IL23, and ranked beyond the 95th percentile in a Triple-Negative enriched BC collection from the Oncomine Platform. CIBERSORTx highlighted a defective dendritic cell, CD4 + T and Î 3δT lymphocyte content and a prominent LAG3 expression in IL30 high versus IL30 low human BC samples from the TCGA PanCancer collection. Conclusions Constitutive expression of membrane-bound IL30 regulates BCSC viability by juxtacrine signals and via second-level mediators, mainly CXCL10 and IL23. Their autocrine loops mediate much of the CSC growth factor activity of IL30, while their paracrine effect contributes to IL30 shaping of immune contexture. IL30-related immune subversion, which also emerged from computational analyses, strongly suggests that targeting IL30 can restrain the BCSC compartment and counteract BC progression
