    Importance of geometry of the extracellular matrix in endochondral bone differentiation.

    Subcutaneous implantation of coarse powders (74-420 micron) of demineralized diaphyseal bone matrix resulted in the local differentiation of endochondral bone. However, implantation of matrix with particle size of 44-74 micron (Fine matrix) did not induce bone. We have recently reported that the dissociative extraction of coarse matrix with 4 M guanidine HCl resulted in a complete loss of the ability of matrix to induce endochondral bone; the total loss of biological activity could be restored by reconstitution of extracted soluble components with inactive residue. To determine the possible biochemical potential of fine matrix to induce bone, the matrix was extracted in 4 M guanidine HCl and the extract was reconstituted with biologically inactive 4 M guanidine HCl-treated coarse bone matrix residue. There was a complete restoration of the biological activity by the extract of fine matrix upon reconstitution with extracted coarse matrix. Polyacrylamide gel electrophoresis of the extract of fine matrix revealed similar protein profiles as seen for the extract of coarse matrix. Gel filtration of the 4 M guanidine HCl extract of fine powder on Sepharose CL-6B and the subsequent reconstitution of various column fractions with inactive coarse residue showed that fractions with proteins of 20,000-50,000 mol wt induced new bone formation. These observations demonstrate that although fine bone matrix contains, osteoinductive proteins, matrix geometry (size) is a critical factor in triggering the biochemical cascade of endochondral bone differentiation. Mixing of coarse matrix with Fine results in partial response and it was confined to areas in contact with coarse particles. The results imply a role for geometry of extracellular bone matrix in anchorage-dependent proliferation and differentiation of cells

    For charged black hole, within the grand canonical ensemble, the decay rate from thermal AdS to the black hole at a fixed high temperature increases with the chemical potential. We check that this feature is well captured by a phenomenological matrix model expected to describe its strongly coupled dual. This comparison is made by explicitly constructing the kink and bounce solutions around the de-confinement transition and evaluating the matrix model effective potential on the solutions.Comment: 1+12 pages, 9 figure

    Psychological and physiological adaptations to sperm competition in humans

    Postcopulatory competition between males, in the form of sperm competition, is a widespread phenomenon in many animal species. The extent to which sperm competition has been an important selective pressure during human evolution remains controversial, however. The authors review critically the evidence that human males and females have psychological, behavioral, and physiological adaptations that evolved in response to selection pressures associated with sperm competition. The authors consider, using evidence from contemporary societies, whether sperm competition is likely to have been a significant adaptive problem for ancestral humans and examine the evidence suggesting that human males have physiological and psychological mechanisms that allow for “prudent” sperm allocation in response to variations in the risk of sperm competition

    Diabetic foot disease and oedema

    Diabetic foot ulcers (DFUs) are common and disabling, giving rise to significant morbidity and mortality as well as worldwide socioeconomic problems. Despite treatment, DFUs readily become chronic wounds and may lead to major lower limb amputations. The pathogenesis of DFUs is complex and the main aetiologies are peripheral neuropathy, ischaemia from peripheral arterial disease and biomechanical abnormalities. Microvascular disease is also a significant problem for people with diabetes and contributes to foot ulceration. Successful management of DFUs consists of debridement, infection control, the use of offloading appliances and revascularisation where necessary. Foot ulcers are usually associated with infection and inflammation which lead to surrounding oedema of the foot. Standard offloading devices such as total contact casts and removable cast walkers do not actively reduce foot oedema. There is promising evidence that active oedema reduction by intermittent pneumatic compression in the diabetic foot improves ulcer healing. The objective of this article is to review the association of foot oedema and DFUs, including the role of appliances which reduce oedema.     Modelling the spatial behaviour of a tropical tuna purse seine fleet.

    Industrial tuna fisheries operate in the Indian, Atlantic and Pacific Oceans, but concerns over sustainability and environmental impacts of these fisheries have resulted in increased scrutiny of how they are managed. An important but often overlooked factor in the success or failure of tuna fisheries management is the behaviour of fishers and fishing fleets. Uncertainty in how a fishing fleet will respond to management or other influences can be reduced by anticipating fleet behaviour, although to date there has been little research directed at understanding and anticipating the human dimension of tuna fisheries. The aim of this study was to address gaps in knowledge of the behaviour of tuna fleets, using the Indian Ocean tropical tuna purse seine fishery as a case study. We use statistical modelling to examine the factors that influence the spatial behaviour of the purse seine fleet at broad spatiotemporal scales. This analysis reveals very high consistency between years in the use of seasonal fishing grounds by the fleet, as well as a forcing influence of biophysical ocean conditions on the distribution of fishing effort. These findings suggest strong inertia in the spatial behaviour of the fleet, which has important implications for predicting the response of the fleet to natural events or management measures (e.g., spatial closures)
