    <p>Insect bite hypersensitivity (IBH), which is a cutaneous allergic reaction to antigens from Culicoides spp., is the most prevalent skin disorder in horses. Misdiagnosis is possible, as IBH is usually diagnosed based on clinical signs. Our study is the first to employ IgE levels against several recombinant Culicoides spp. allergens as an objective, independent, and quantitative phenotype to improve the power to detect genetic variants that underlie IBH. Genotypes of 200 Shetland ponies, 127 Icelandic horses, and 223 Belgian Warmblood horses were analyzed while using a mixed model approach. No single-nucleotide polymorphism (SNP) passed the Bonferroni corrected significance threshold, but several regions were identified within and across breeds, which confirmed previously identified regions of interest and, in addition, identifying new regions of interest. Allergen-specific IgE levels are a continuous and objective phenotype that allow for more powerful analyses when compared to a case-control set-up, as more significant associations were obtained. However, the use of a higher density array seems necessary to fully employ the use of IgE levels as a phenotype. While these results still require validation in a large independent dataset, the use of allergen-specific IgE levels showed value as an objective and continuous phenotype that can deepen our understanding of the biology underlying IBH.</p

    Risk Factor Assessment, Sero-Prevalence, and Genotyping of the Virus That Causes Foot-and-Mouth Disease on Commercial Farms in Ethiopia from October 2018 to February 2020

    A cross-sectional serological, active outbreak search and a questionnaire-based survey were carried out to investigate foot-and-mouth disease (FMD) sero-epidemiology in Ethiopia. The circulating serotype of the FMD virus (FMDV) was identified, and the knowledge regarding FMD and husbandry practices was assessed. Using the questionnaire survey, a total of 237 individuals were interviewed, and the majority responded that there is no practice of reporting disease outbreak, free in and out movement of livestock, or share pastures, and they use traditional case management as a means of controlling the disease. A total of 1938 cattle, 490 domestic small ruminants, and 426 swine were sampled randomly for serological analysis using the 3ABC non-structural protein (NSP) enzyme-linked immunosorbant assay. An overall prevalence of 25% in cattle, 5% in domestic small ruminants, and 2% in swine was recorded. Multivariable logistic regression analysis revealed that cattle from the Oromia, Tigray, and Amhara regions had the highest probability of being sero-positive as compared with Addis Ababa odds ratio(OR)(OR: 4 (95% confidence interval (CI)(CI [3–6], 3 (95% CI [2,5]), and 2 (95% CI 2 [1,3]), respectively)). Older cattle (older than three years) and domestic small ruminants (>18 months) had a higher chance of being seropositive (OR: 2, 95% CI [1.6,3]) and (OR: 6, 95% CI [2,18]), respectively). Female and older swine older than three years of age had a higher chance of being sero-positive (p p < 0.05) in cattle. Swine from Bishoftu were less likely to test positive than swine from Addis Ababa (OR: 0.04, 95% CI [0.01–0.3]). From 96 herds, 72 pooled outbreak samples were analyzed by polymerase chain reaction (PCR), virus isolation, serotyping (antigen enzyme linked immunosorbant assay (ELISA)), sequencing, and phylogenetic tree analysis. Six serotype A (G-IV) FMD viruses and three serotype O east African (EA-3 and EA-4) FMDVs were identified. Thus, this study established the lack of disease outbreak reporting, poor husbandry problems, and the prevalence of FMD in three domestic species (cattle, small ruminant, and swine).     The Caspase 8 Inhibitor c-FLIP(L) Modulates T-Cell Receptor-Induced Proliferation but Not Activation-Induced Cell Death of Lymphocytes

    The caspase 8 inhibitor c-FLIP(L) can act in vitro as a molecular switch between cell death and growth signals transmitted by the death receptor Fas (CD95). To elucidate its function in vivo, transgenic mice were generated that overexpress c-FLIP(L) in the T-cell compartment (c-FLIP(L) Tg mice). As anticipated, FasL-induced apoptosis was inhibited in T cells from the c-FLIP(L) Tg mice. In contrast, activation-induced cell death of T cells in c-FLIP(L) Tg mice was unaffected, suggesting that this deletion process can proceed in the absence of active caspase 8. Accordingly, c-FLIP(L) Tg mice differed from Fas-deficient mice by showing no accumulation of B220(+) CD4(−) CD8(−) T cells. However, stimulation of T lymphocytes with suboptimal doses of anti-CD3 or antigen revealed increased proliferative responses in T cells from c-FLIP(L) Tg mice. Thus, a major role of c-FLIP(L) in vivo is the modulation of T-cell proliferation by decreasing the T-cell receptor signaling threshold

    Cytokinesis defects and cancer

    Whole-genome and centrosome duplication as a consequence of cytokinesis failure can drive tumorigenesis in experimental model systems. However, whether cytokinesis failure is in fact an important cause of human cancers has remained unclear. In this Review, we summarize evidence that whole-genome-doubling events are frequently observed in human cancers and discuss the contribution that cytokinesis defects can make to tumorigenesis. We provide an overview of the potential causes of cytokinesis failure and discuss how tetraploid cells that are generated through cytokinesis defects are used in cancer as a transitory state on the route to aneuploidy. Finally, we discuss how cytokinesis defects can facilitate genetic diversification within the tumour to promote cancer development and could constitute the path of least resistance in tumour evolution
