    An Experimental Study to Quantify Error Rates Resulting from Measurement Deviation in Area of Origin Reconstructions of Blunt Force Impact Patterns

    The intent of this study was to attempt to quantify error associated with the measurements required in area of origin reconstructions resulting from the analysis of blunt force impact patterns. Mathematical tables were constructed in order to examine trends associated with changing width and length ratios and the influence of impact angle change and area of convergence deviations. The analysis of the trends enabled informed stain selection, mitigating potential error. The analysis of the influence of stain measurement error and gamma angle error was conducted by reconstructing experimentally created blunt force impact patterns using the Tangent Method, comparing the resulting area of origin determinations to reconstructions generated using HemoSpat, a bloodstain pattern analysis software, and then isolating each variable in order to examine its effect on precision and accuracy. A total of 10 blunt force impact patterns were created and initially analyzed utilizing the Tangent Method. The stains selected for the analysis of each pattern were input into HemoSpat software which generated separate and independent results, enabling a comparison of the absolute and relative error rates between the known area of origin and the two methodologies. This also provided a foundation for the examination of each variable’s contribution to absolute and relative error. Finally, artificially induced measurement error was generated by uniformly increasing and decreasing the length, width, and gamma angle values of the selected stains based on an absolute analysis of error. The deviation from the compared values was examined in order to determine if the resulting area of origin determination would adversely affect inferences related to scene analysis. The results indicate that the incorporation of measurement error into a reconstruction creates an error rate that would not substantively affect an area of origin determination or inferences which would typically be rendered based on that determination

    Comparison of two sampling protocols and four home-range estimators using radio-tracking data from urban badgers Meles meles

    Radio-telemetry is often the method of choice for studies of species whose behaviour is difficult to observe directly. However, considerable debate has ensued about the best way of deriving home-range estimates. In recent years, kernel estimators have become the most widely used method, together with the oldest and simplest method, the minimum convex polygon (MCP). More recently, it has been suggested that the local convex hull (LCH) might be more appropriate than kernel methods in cases where an animal’s home range includes a priori inaccessible areas. Yet another method, the Brownian bridge (BB), explicitly uses autocorrelated data to determine movement paths and, ultimately, home ranges or migration routes of animals. Whereas several studies have used simulation techniques to compare these different methods, few have used data from real animals. We used radio-telemetric data from urban badgers Meles meles to compare two sampling protocols (10-minute vs at least 30-minute inter-fix intervals) and four home-range estimators (MCP, fixed kernels (FK), LCH and BB). We used a multi-response permutation procedure and randomisation tests to compare overall patterns of fixes and degree of overlap of home ranges estimated using data from different sampling protocols, and a general linear model to compare the influence of sampling protocols and home-range estimator on the size of habitat patches. The shape of the estimated home ranges was influenced by sampling protocol in some cases. By contrast, the sizes and proportions of different habitats within home ranges were influenced by estimator type but not by sampling protocol. LCH performed consistently better than FK, and is especially appropriate for patchy study areas containing frequent no-go zones. However, we recommend using LCH in combination with other methods to estimate total range size, because LCH tended to produce smaller estimates than any other method.     Neuronal imaging with ultrahigh dynamic range multiphoton microscopy

    Multiphoton microscopes are hampered by limited dynamic range, preventing weak sample features from being detected in the presence of strong features, or preventing the capture of unpredictable bursts in sample strength. We present a digital electronic add-on technique that vastly improves the dynamic range of a multiphoton microscope while limiting potential photodamage. The add-on provides real-time negative feedback to regulate the laser power delivered to the sample, and a log representation of the sample strength to accommodate ultrahigh dynamic range without loss of information. No microscope hardware modifications are required, making the technique readily compatible with commercial instruments.     Mechalele (Self-Playing Ukulele)

    This project involves the design and implementation of a self-playing ukulele. The ukulele plays chords and plucks open strings based on a song or chord progression selected by the user. Each string on the ukulele has a designated hobby servo motor that plucks the string with pre-programmed timing as is shown in Figure 1 (3). The plucking is achieved by attaching a flexible piece of cardboard to the shaft of the servo to act as the guitar pick. By moving the motors together at once, the action of strumming can be approximated. The design uses the last three frets on the ukulele for pressing different strings and allows the ukulele to play different notes and chords. The strings are pressed by small push/pull solenoids (2) that are spring loaded to push the string when active and not touch the string when passive. The output of the system is controlled with user selection buttons that selects song or tuning mode for the ukulele from an LCD display (6). The user can either select tuning or song playing mode using three pushbuttons. In tuning mode, a speaker plays the desired note for a string and the LCD display prompts the user to pluck that string. The frequency of the note is detected from a microphone and the display tells the user if the string is tuned, too high, or too low

    Effective Exploration Behavior for Chemical-Sensing Robots

    Mobile robots that can effectively detect chemical effluents could be useful in a variety of situations, such as disaster relief or drug sniffing. Such a robot might mimic biological systems that exhibit chemotaxis, which is movement towards or away from a chemical stimulant in the environment. Some existing robotic exploration algorithms that mimic chemotaxis suffer from the problems of getting stuck in local maxima and becoming “lost”, or unable to find the chemical if there is no initial detection. We introduce the use of the RapidCell algorithm for mobile robots exploring regions with potentially detectable chemical concentrations. The RapidCell algorithm mimics the biology behind the biased random walk of Escherichia coli (E. coli) bacteria more closely than traditional chemotaxis algorithms by simulating the chemical signaling pathways interior to the cell. For comparison, we implemented a classical chemotaxis controller and a controller based on RapidCell, then tested them in a variety of simulated and real environments (using phototaxis as a surrogate for chemotaxis). We also added simple obstacle avoidance behavior to explore how it affects the success of the algorithms. Both simulations and experiments showed that the RapidCell controller more fully explored the entire region of detectable chemical when compared with the classical controller. If there is no detectable chemical present, the RapidCell controller performs random walk in a much wider range, hence increasing the chance of encountering the chemical. We also simulated an environment with triple effluent to show that the RapidCell controller avoided being captured by the first encountered peak, which is a common issue for the classical controller. Our study demonstrates that mimicking the adapting sensory system of E. coli chemotaxis can help mobile robots to efficiently explore the environment while retaining their sensitivity to the chemical gradient

    Model selection for prognostic time-to-event gene signature discovery with applications in early breast cancer data

    Model selection between competing models is a key consideration in the discovery of prognostic multigene signatures. The use of appropriate statistical performance measures as well as verification of biological significance of the signatures is imperative to maximise the chance of external validation of the generated signatures. Current approaches in time-to-event studies often use only a single measure of performance in model selection, such as logrank test p-values, or dichotomise the follow-up times at some phase of the study to facilitate signature discovery. In this study we improve the prognostic signature discovery process through the application of the multivariate partial Cox model combined with the concordance index, hazard ratio of predictions, independence from available clinical covariates and biological enrichment as measures of signature performance. The proposed framework was applied to discover prognostic multigene signatures from early breast cancer data. The partial Cox model combined with the multiple performance measures were used in both guiding the selection of the optimal panel of prognostic genes and prediction of risk within cross validation without dichotomising the follow-up times at any stage. The signatures were successfully externally cross validated in independent breast cancer datasets, yielding a hazard ratio of 2.55 [1.44, 4.51] for the top ranking signature.</p

    High quality, patient centred andcoordinated care for Alstrom syndrome: amodel of care for an ultra-rare disease

    Background: Patients with rare and ultra-rare diseases make heavy demands on the resources of both health and social services, but these resources are often used inefficiently due to delays in diagnosis, poor and fragmented care. We analysed the national service for an ultra-rare disease, Alstrom syndrome, and compared the outcome and cost of the service to the standard care. Methods: Between the 9th and 26th of March 2014 we undertook a cross-sectional study of the UK Alstrom syndrome patients and their carers. We developed a semi-structured questionnaire to assess our rare patient need, quality of care and costs incurred to patients and their careers. In the UK all Alstrom syndrome patients are seen in two centres, based in Birmingham, and we systematically evaluated the national service and compared the quality and cost of care with patients’ previous standard of care. Results: One quarter of genetically confirmed Alstrom syndrome UK patients were enrolled in this study. Patients that have access to a highly specialised clinical service reported that their care is well organised, personalised, holistic, and that they have a say in their care. All patients reported high level of satisfaction in their care. Patient treatment compliance and clinic attendance was better in multidisciplinary clinic than the usual standard of NHS care. Following a variable costing approach based on personnel and consumables’ cost, our valuation of the clinics was just under £700/patient/annum compared to the standard care of £960/patient/annum. Real savings, however, came in terms of patients’ quality of life. Furthermore there was found to have been a significant reduction in frequency of clinic visits and ordering of investigations since the establishment of the national service. Conclusions: Our study has shown that organised, multidisciplinary “one stop” clinics are patient centred and individually tailored to the patient need with a better outcome and comparable cost compared with the current standard of care for rare disease. Our proposed care model can be adapted to several other rare and ultra-rare diseases

    Retrieval of Precise Radial Velocities from Near-Infrared High Resolution Spectra of Low Mass Stars

    Given that low-mass stars have intrinsically low luminosities at optical wavelengths and a propensity for stellar activity, it is advantageous for radial velocity (RV) surveys of these objects to use near-infrared (NIR) wavelengths. In this work we describe and test a novel RV extraction pipeline dedicated to retrieving RVs from low mass stars using NIR spectra taken by the CSHELL spectrograph at the NASA Infrared Telescope Facility, where a methane isotopologue gas cell is used for wavelength calibration. The pipeline minimizes the residuals between the observations and a spectral model composed of templates for the target star, the gas cell, and atmospheric telluric absorption; models of the line spread function, continuum curvature, and sinusoidal fringing; and a parameterization of the wavelength solution. The stellar template is derived iteratively from the science observations themselves without a need for separate observations dedicated to retrieving it. Despite limitations from CSHELL's narrow wavelength range and instrumental systematics, we are able to (1) obtain an RV precision of 35 m/s for the RV standard star GJ 15 A over a time baseline of 817 days, reaching the photon noise limit for our attained SNR, (2) achieve ~3 m/s RV precision for the M giant SV Peg over a baseline of several days and confirm its long-term RV trend due to stellar pulsations, as well as obtain nightly noise floors of ~2 - 6 m/s, and (3) show that our data are consistent with the known masses, periods, and orbital eccentricities of the two most massive planets orbiting GJ 876. Future applications of our pipeline to RV surveys using the next generation of NIR spectrographs, such as iSHELL, will enable the potential detection of Super-Earths and Mini-Neptunes in the habitable zones of M dwarfs.Comment: 64 pages, 28 figures, 5 tables.     Phaeochromocytoma and extra adrenal paraganglioma are rare neuroendocrine tumours and have the potential to secrete adrenaline, noradrenaline and dopamine causing a myriad of clinical symptoms. Prompt diagnosis is essential for clinicians and requires a multidisciplinary specialist approach for the clinical and laboratory investigation, diagnosis, treatment and follow-up of patients. This paper is an integrated review of the clinical and laboratory evaluation and treatment of patients suspected to have phaeochromocytoma or paraganglioma, highlighting recent developments and best practices from recent published clinical guidelines.</jats:p
